Beragam jenis tumbuhan obat yang telah lama digunakan secara tradisional kini dipopulerkan kembali, salah satunya sirih (Piper betle L.). Daun sirih dikenal sebagai bahan untuk menginang dengan keyakinan bahwa daun sirih dapat menguatkan gigi, menyembuhkan luka-luka kecil di mulut, menghilangkan bau mulut, menghentikan perdarahan gusi dan sebagai obat kumur. Oleh karena itu, penelitian ini bertujuan untuk menganalisis total fenol pada ekstrak segar daun sirih hijau. Pengambilan sampel daun sirih hijau berdasarkan usia, yaitu dipilih daun sirih hijau yang berusia 1 bulan. Pembuatan ekstrak dilakukan dengan metode maserasi. Daun dicuci hingga bersih kemudian dikeringkan dengan cara diangin-anginkan, dirajang, ditimbang lalu dimasukkan ke dalam erlenmeyer dan ditambahkan metanol 87,5 ml. Filtrasi dengan kertas saring, filtrat disimpan di lemari pendingin, lalu residu kembali dimaserasi dengan metanol 87,5 ml selama 2x24 jam pada suhu 27oC, residu kemudian disaring kembali dengan kertas saring. Analisis senyawa fenolik menggunakan metode spektrofotometri UV-Vis. Penetapan kadar dilakukan dengan menggunakan pereaksi Folin-Cioceltau. Panjang gelombang yang digunakan pada penetapan kadar senyawa fenolik yaitu 785 nm dan aquabidest sebagai blanko dengan tiga kali pengulangan. Kurva baku yang digunakan adalah asam galat dengan konsentrasi 0; 50; 100; 150; 200 ppm. Data hasil penelitian berupa nilai absorbansi pengulangan I sebesar 0,046 sehingga diperoleh kadar I = 166,71 ppm; absorbansi pengulangan II sebesar 0,049 sehingga diperoleh kadar II = 165,10 ppm; serta absorbansi pengulangan III sebesar 0,050 sehingga diperoleh kadar III = 164,56 ppm. Penelitian ini menyimpulkan kadar rata-rata total fenol pada ekstrak daun sirih hijau yang diperoleh adalah sebesar 165,45 ppm.Kata kunci : sirih hijau, total fenol, Piper betle
