Memories are not stored as exact copies of our experiences. As a result, remembering is subject not only to memory failure, but to inaccuracies and distortions as well. Although such distortions are often retained or even enhanced over time, sleep's contribution to the development of false memories is unknown. Here, we report that a night of sleep increases both veridical and false recall in the DeeseRoediger-McDermott (DRM) paradigm, compared to an equivalent period of daytime wakefulness. But while veridical memory deteriorates across both wake and sleep, false memories are preferentially preserved by sleep, actually showing a non-significant improvement. The same selectivity of false over veridical memories was observed in a follow-up nap study. Unlike previous studies implicating deep, slow-wave sleep (SWS) in declarative memory consolidation, here veridical recall correlated with decreased SWS, a finding that was observed in both the overnight and nap studies. These findings lead to two counterintuitive conclusions -that under certain circumstances sleep can promote false memories over veridical ones, and SWS can be associated with impairment rather than facilitation of declarative memory consolidation. While these effects produce memories that are less accurate after sleep, these memories may, in the end, be more useful.
Transgenic expression in the hypothalamus of syndecan-1, a cell surface heparan sulfate proteoglycan (HSPG) and modulator of ligand-receptor encounters, produces mice with hyperphagia and maturity-onset obesity resembling mice with reduced action of alpha melanocyte stimulating hormone (alphaMSH). Via their HS chains, syndecans potentiate the action of agouti-related protein and agouti signaling protein, endogenous inhibitors of alphaMSH. In wild-type mice, syndecan-3, the predominantly neural syndecan, is expressed in hypothalamic regions that control energy balance. Food deprivation increases hypothalamic syndecan-3 levels several-fold. Syndecan-3 null mice, otherwise apparently normal, respond to food deprivation with markedly reduced reflex hyperphagia. We propose that oscillation of hypothalamic syndecan-3 levels physiologically modulates feeding behavior.
TNF-␣-converting enzyme (TACE, herein denoted as Adam17) proteolytically sheds several cell-surface inflammatory proteins, but the physiologic importance of the cleavage of these substrates from leukocyte subsets during inflammation is incompletely understood. In this study, we show that Adam17-null neutrophils have a 2-fold advantage in their initial recruitment during thioglycollate-induced peritonitis, and they roll slower and adhere more readily in the cremaster model than wild-type neutrophils. Although CD44 and ICAM-1 are both in vitro substrates of Adam17, their surface levels are not altered on Adam17-null neutrophils. In contrast, L-selectin levels are elevated up to 10-fold in Adam17-null circulating neutrophils, and their accelerated peritoneal influx, slower rolling, and increased adhesion in the cremaster muscle are dependent on L-selectin. Analysis of mixed chimeras shows that enhanced L-selectin levels and accelerated influx were both cell-intrinsic properties of neutrophils lacking Adam17. In contrast, Adam17-null monocytes display no acceleration of infiltration into the peritoneum in spite of elevated L-selectin surface levels, and their peritoneal influx was independent of L-selectin. Therefore, our data demonstrate substrate and myeloid cell-type specificity of Adam17-mediated cleavage of its substrates, and show that neutrophils and monocytes use distinct mechanisms for infiltration of tissues. IntroductionRecruitment of specific subsets of leukocytes to sites of injury is a complex, multistep process that includes leukocyte tethering and rolling, activation and firm adhesion, and transmigration, 1,2 as well as more recently elaborated steps of slow rolling, adhesion strengthening, intraluminal crawling, paracellular and transcellular migration, and migration through the basement membrane into the tissue. 3 This process can be characterized as a sequential adhesion cascade that is mediated by quantitative and qualitative changes of several distinct surface receptors on leukocytes and the endothelium. Whereas much has been learned about the mechanisms involved in leukocyte adhesion to endothelial cells, less is known about the molecules responsible for de-adhesion and the transition between the sequential steps. However, transendothelial migration is a very rapid event, and it only takes minutes to reach the subendothelial basement membrane once a leukocyte sticks to the endothelium. 4 A useful method to rapidly modulate leukocyte-endothelial interactions is cell-surface proteolysis, a mechanism that can instantly alter the cell-surface protein repertoire. The pathogenesis of Bombyx mori cytoplasmic polyhedrosis virus (BmCPV) infection is unclear, although accumulating evidence indicates that circular RNAs (circRNAs), which act as competing endogenous RNAs or positive regulators, play important roles in regulating gene expression in eukaryotes and, thus, may play a role in BmCPV infections. To explore the expression and biological functions of circRNAs in the silkworm midgut following BmCPV infection, silkworm circRNA expression profiles of normal midgut tissue (control) and BmCPV-infected midgut tissue (test) were determined using high-through sequencing. A total of 9,753 and 7,475circRNAs were detected from the control and test samples, respectively. The two samples shared 6,085 circRNAs, while 646 and 737 circRNAs were expressed specifically in the control and test samples, respectively. A total of 3,638 circRNAs were shown to be differentially expressed, and 400 circRNAs were substantially differentially expressed with a fold-change ≥ 2.0 (p< 0.05 and a false discover rate < 0.05), of which 294 were up-regulated and 106 were down-regulated following infection. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses were conducted to determine the principal functions of the substantially differentially regulated genes. circRNA-miRNA interaction networks were constructed based on a correlation analysis between the differentially expressed circRNAs and the nature of their microRNA (miRNA) binding sites. The network inferred that 13 miRNAs interacting with 193 circRNAs were among the 300 most abundant relationships. bmo-miR-3389-5p, bmo-miR-745-3p, and bmo-miR-3262 were related to 30, 34, and 34 circRNAs, respectively. circRNA_8115, circRNA_9444, circRNA_4553, circRNA_0827, and circRNA_6649 contained six, five, four, four, and four miRNA binding sites, respectively. We further found that alternative circularization of circRNAs is a common feature in silkworms and that the junction sites of many silkworm circRNAs are flanked by canonical GT/AG splicing signals. Our study is the first to show the circRNA response to virus infection. Thus, it provides a novel perspective on circRNA-miRNA interactions during BmCPV pathogenesis, and it lays the foundation for future research of the potential roles of circRNAs in BmCPV pathogenesis.
In order for wireless body area networks to meet widespread adoption, a number of security implications must be explored to promote and maintain fundamental medical ethical principles and social expectations. As a result, integration of security functionality to sensor nodes is required. Integrating security functionality to a wireless sensor node increases the size of the stored software program in program memory, the required time that the sensor's microprocessor needs to process the data and the wireless network traffic which is exchanged among sensors. This security overhead has dominant impact on the energy dissipation which is strongly related to the lifetime of the sensor, a critical aspect in wireless sensor network (WSN) technology. Strict definition of the security functionality, complete hardware model (microprocessor and radio), WBAN topology and the structure of the medium access control (MAC) frame are required for an accurate estimation of the energy that security introduces into the WBAN. In this work, we define a lightweight security scheme for WBAN, we estimate the additional energy consumption that the security scheme introduces to WBAN based on commercial available off-the-shelf hardware components (microprocessor and radio), the network topology and the MAC frame. Furthermore, we propose a new microcontroller design in order to reduce the energy consumption of the system. Experimental results and comparisons with other works are given.
Cage-based aquaculture has been growing rapidly in recent years. In some locations, cage-based aquaculture has resulted in the clustering of large quantities of cages in fish farms located in inland lakes or reservoirs and coastal embayments or fjords, significantly affecting flow and mass transport in the surrounding waters. Existing studies have focused primarily on the macro-scale flow blockage effects of fish cages, and the complex wake flow and associated near-field mass transport in the presence of the cages remain largely unclear. As a first step toward resolving this knowledge gap, this study employed the combined Particle Image Velocimetry and Planar Laser Induced Fluorescence (PIV-PLIF) flow imaging technique to measure turbulence characteristics and associated mass transport in the near wake of a steady current through an aquaculture cage net panel in parametric flume experiments. In the near-wake region, defined as ~3M (mesh size) downstream of the net, the flow turbulence was observed to be highly inhomogeneous and anisotropic in nature. Further downstream, the turbulent intensity followed a power-law decay after the turbulence production region, albeit with a decay exponent much smaller than reported values for analogous grid-generated turbulence. Overall, the presence of the net panel slightly enhanced the lateral spreading of the scalar plume, but the lateral distribution of the scalar concentration, concentration fluctuation and transverse turbulent scalar flux exhibited self-similarity from the near-wake region where the flow was still strongly inhomogeneous. The apparent turbulent diffusivity estimated from the gross plume parameters was found to be in reasonable agreement with the Taylor diffusivity calculated as the product of the transverse velocity fluctuation and integral length scale, even when the plume development was still transitioning from a turbulent-convective to turbulent-diffusive regime. The findings of this study provide references to the near-field scalar transport of fish cages, which has important implications in the assessment of the environmental impacts and environmental carrying capacity of cage-based aquaculture.
