RESUMENEl uso de sustrato con lombricompuesto favorece el crecimiento en cultivos hortícolas, siendo importante conocer sus propiedades para dosificar la cantidad adecuada. Este trabajo tuvo como objetivo analizar las modificaciones producidas en un sustrato según la dosis de lombricompuesto adicionado y evaluar el efecto de las mezclas sobre la calidad del plantín de lechuga. El ensayo se realizó en un invernadero en Luján, Argentina (34°36' S, 59°04' W). Se sembró lechuga mantecosa cv. Tizona, utilizando sustratos compuestos por distintas combinaciones porcentuales de lombricompuesto y una mezcla comercial formulada con turba y perlita: 1) 0% lombricompuesto, 2) 20% lombricompuesto, 3) 40% lombricompuesto. Las mezclas se analizaron físicoquímicamente. En las plantas se midió número de hojas, peso fresco y seco de parte aérea, raíz y total. El diseño fue en bloques completos aleatorizados con 4 repeticiones. Se realizó análisis de la varianza, evaluando diferencias entre medias por la prueba de Tukey y análisis de regresión para estudiar el tipo de respuesta en los parámetros físicoquímicos de los sustratos por la incorporación de lombricompuesto en el rango de 0 a 40%. La adición de lombricompuesto en los sustratos aumentó linealmente el pH, la conductividad eléctrica, el nivel de nutrientes, la densidad aparente y la porosidad total, y disminuyó linealmente la materia orgánica y cuadráticamente el porcentaje de poros ocupados por agua. Las plantas respondieron al agregado de lombricompuesto adquiriendo mayor tamaño con 20%.Palabras clave: almácigo, vermicompuesto, Eisenia foetida. Alstroemeria (Alstroemeriaceae) es un género nativo de Sudamérica. Sus especies, que se distribuyen desde Venezuela hasta Argentina y Chile, tienen alto impacto en el mercado de plantas ornamentales, debido al color y atractiva forma de sus flores. El cultivo in vitro de meristemas aumenta la eficiencia de la propagación de las plantas y su calidad, por contribuir con el saneamiento de enfermedades sistémicas. El presente estudio tuvo como objetivo desarrollar un protocolo eficiente de cultivo in vitro de meristemas de rizoma de Alstroemeria híbrida var. Fiesta de 15 INTA, a partir del agregado de reguladores de crecimiento bencilaminopurina (BAP) y ácido naftalenacético (ANA) a un medio de cultivo base. Se evaluó la cantidad total de plantas completas alcanzadas, producción de hojas, longitud aérea y radical, formación de callo, necrosis y mortandad de los explantos sembrados sobre MS; MS + 1 mg L-1 BAP; MS + 0,2 mg L-1 ANA; y MS + 0,2 mg L-1 ANA + 1 mg L-1 BAP. Se realizó un análisis de la varianza y la evaluación de diferencias entre medias por el test de Tukey. La suplementación de 0,2 mg L-1 ANA al medio MS es suficiente para el cultivo de meristemas de rizoma de Alstroemeria var. Fiesta de 15 INTA, ya que su empleo resultó en una mayor producción aérea y radical y, consecuentemente de plantas completas, además de un alto porcentaje de sobrevivencia de plantas luego de su transferencia al invernáculo. El Instituto de Floricultura, perteneciente al Instituto Nacional de Tecnología Agropecuaria (INTA) lleva a cabo diversos proyectos relacionados con el acceso y el uso sustentable de los recursos fitogenéticos nativos del país con fines de investigación y productivos. Dentro de estas actividades, este Instituto realiza el mejoramiento del género Calibrachoa (plantas conocidas vulgarmente como calibrachoas) con la finalidad de obtener cultivares ornamentales para el mercado nacional e internacional. En relación con los aspectos agronómicos, la infección por patógenos es uno de los factores de mayor importancia que limitan la producción de calibrachoa. Por ello, en primer lugar se realizó la identificación de la especie de potyvirus que se encontraba infectando estas plantas y finalmente se detectó a potato virus Y como patógeno de los cultivares de calibrachoa por primera vez en la Argentina. Asimismo, resultó fundamental analizar el rol de las semillas en la transmisión de PVY y la evaluación de la influencia de los factores ambientales sobre este proceso, por lo que se realizaron cruzamientos dirigidos entre plantas infectadas con PVY, y entre plantas infectadas con PVY y plantas sin infectar, durante el verano, el otoño y la primavera. A partir de la técnica RT-PCR, se confirmó que esta especie viral se transmite por semillas tanto en el caso de uno como de ambos padres infectados, independientemente de las condiciones de la época del año, y que existe una incidencia importante del tejido materno sobre la cantidad de semillas producidas y la cantidad de plantas logradas a partir de los cruzamientos. Por otro lado, se llevó a cabo la identificación de genotipos resistentes a patógenos, cuyo conocimiento permite incorporar fuentes de variabilidad a los programas de mejoramiento, evitando problemas sanitarios, por lo que se llevó a cabo la inoculación (por transmisión mecánica) de plantas silvestres de calibrachoa y de petunia provenientes del banco de germoplasma del Instituto de floricultura (Calibrachoa caesia, C. thymifolia, C. linoides, Petunia inflata y P. integrifolia) con un extracto proveniente de plantas de calibrachoa fehacientemente infectadas con PVY. Posteriormente, mediante observación de la sintomatología y detección mediante ELISA y RT-PCR, se vio que todas las plantas analizadas se infectaron con el virus, lo que mostró la ausencia de resistencia en estas plantas a PVY. A su vez, se llevó a cabo la inoculación de C. thymifolia y P. integrifolia con la cepa patógena Nigrospora oryzae a modo de evaluar el comportamiento y de detectar posibles fuentes de resistencia a este hongo, y finalmente se vio que C. thymifolia presenta resistencia a N. oryzae, mientras que P. integrifolia es susceptible. Por otra parte, en este trabajo se llevó a cabo la optimización de un protocolo de regeneración de plantas de Calibrachoa cv. Pampa Salmón INTA a partir de meristemas apicales, mediante el análisis comparativo de diferentes medios de cultivo. Finalmente, se vio que la combinación de MS ½ (Murashige y Skoog ½) + 0,01 mg L-1 6-bencilaminopurina (BAP) resultó en la obtención de un alto porcentaje de regeneración de plantas completas, además de una mayor producción aérea y radical que, consecuentemente, permitieron una alta tasa de sobrevivencia de plantas luego de su transferencia al invernáculo. A partir de este avance, se pudo realizar el cultivo de meristemas con el objetivo de sanear las plantas de C. cv. Pampa Salmón INTA infectadas con PVY, lo que permitió erradicar el PVY en el 74% de los casos, confirmado mediante RT-PCR. Por lo tanto, dada la importancia comercial de C. hybrida y frente al escaso conocimiento de la etiología de las enfermedades que la afectan, este trabajo permitió generar información sobre la interacción patógenos-hospedante y su manejo.
