A marine Antarctic psychrotolerant bacterium (strain ANT/505), isolated from sea ice-covered surface water from the Southern Ocean, showed pectinolytic activity on citrus pectin agar. The sequencing of the 16S rRNA of isolate ANT/505 indicates a taxonomic affiliation to Pseudoalteromonas haloplanktis . The supernatant of this strain showed three different pectinolytic activities after growth on citrus pectin. By activity screening of a genomic DNA library of isolate ANT/505 in Escherichia coli , two different pectinolytic clones could be isolated. Subcloning and sequencing revealed two open reading frames (ORF) of 1,671 and 1,968 nt, corresponding to proteins of 68 and 75 kDa, respectively. The deduced amino acid sequence of the two ORFs showed homology to pectate lyases from Erwinia chrysanthemi and Aspergillus nidulans . The pectate lyases contain signal peptides of 17 and 26 amino acids that were correctly processed after overexpression in E. coli BL21. Both enzymes were purified by anionic exchange chromatography. Maximal enzymatic activities for both pectate lyases were observed at 30 ° C and a pH range of 9 to 10. The K m values of both lyases for pectate and citrus pectin were 1 g l -1 and 5 g l -1, respectively. Calcium was required for activity on pectic substrates, whereas the addition of 1 mM ethylenediaminetetraacetic acid (EDTA) resulted in complete inhibition of the enzymes. These two enzymes represent the first pectate lyases isolated and characterized from a cold-adapted marine bacterium.
Summary Mobile genomic islands distribute functional traits between microbes and habitats, yet it remains unclear how their proteins adapt to new environments. Here we used a comparative phylogenomic and proteomic approach to show that the marine bacterium Pseudoalteromonas haloplanktis ANT/505 acquired a genomic island with a functional pathway for pectin catabolism. Bioinformatics and biochemical experiments revealed that this pathway encodes a series of carbohydrate‐active enzymes including two multi‐modular pectate lyases, PelA and PelB. PelA is a large enzyme with a polysaccharide lyase family 1 (PL1) domain and a carbohydrate esterase family 8 domain, and PelB contains a PL1 domain and two carbohydrate‐binding domains of family 13. Comparative phylogenomic analyses indicate that the pathway was most likely acquired from terrestrial microbes, yet we observed multi‐modular orthologues only in marine bacteria. Proteomic experiments showed that P. haloplanktis ANT/505 secretes both pectate lyases into the environment in the presence of pectin. These multi‐modular enzymes may therefore represent a marine innovation that enhances physical interaction with pectins to reduce loss of substrate and enzymes by diffusion. Our results revealed that marine bacteria can catabolize pectin, and highlight enzyme fusion as a potential adaptation that may facilitate microbial consumption of polymeric substrates in aquatic environments.
The interdecadal Pacific Oscillation (IPO) represents the decadal mode of the El Niño-Southern Oscillation phenomenon. As such the IPO is one of the dominant modes of decadal climate variability on both sides of the Pacific Ocean basin. For this paper we utilized a newly developed tree-ring data network comprised of five multi-centennial Vietnamese cypress ring-width chronologies that range from around 11°N-23°N latitude. We combined these data with an average of tree-ring derived drought indices from the North American Drought Atlas over the south central USA, from a box that spans from 27.5°N-35.0°N, 85.0°-110.0°W and contains 55 gridpoints and more than 100 tree ring site chronologies, these 2 locations exhibit rainfall variations that are strongly, negatively correlated with each other and are representative of the influence of the IPO on terrestrial rainfall. The final reconstruction model, weighted most heavily on the three most southerly of the five Vietnamese cypress records, spans from 1350 to 2004, and explains nearly 57% of the variance in the original IPO data for the 5-month season of October-February. The reconstruction model passes all standard statistical tests using a split calibrationverification scheme. We reveal 15 positive and 15 negative phase shifts of the IPO prior to the period of instrumentation, suggesting that the IPO has been active for at least the past seven centuries with varying degrees of intensity. We compare our reconstruction with two related millennial records: the MacDonald and Case (Geophys Res Lett 32(8):L08703, 2005) tree ring-derived reconstruction of the Pacific Decadal Oscillation, and an ice core-derived reconstruction of the IPO from the Law Dome Ice core in Antarctica by Vance et al. (Geophys Res Lett 33(6):L06712, 2015). While there is good general agreement with the latter record, there are three key periods of the past where the two records are out of phase, and we explore the reasons for this disparity. Direct comparison with the related Tripole Index (TPI) shows weaker correlation, likely owing to the stronger relationship between our tree ring data and the equatorial Pacific region relative to the north and south regions of the Pacific that combine to comprise the TPI calculation.
Context:Paramignya trimera (Oliv.) Burkill (Rutaceae) has been used to treat liver diseases and cancer. However, the anti-inflammatory effects of this medicinal plant and its components have not been elucidated.Objective: This study investigated chemical constituents of the P. trimera stems and evaluated anti-inflammatory effects of isolated compounds.Materials and methods: Cytotoxicity of isolated compounds (5–40 μM) toward BV2 cells was tested using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) for 24 h. Inhibitory effects of isolated compounds (5-40 μM) on nitrite and PGE2 concentrations were determined using Griess reaction and PGE2 ELISA kit, respectively (pretreated with the compounds for 3 h and then stimulated for 18 h with LPS). Inhibitory effects of compounds (5-40 μM) on iNOS and COX-2 protein expression were evaluated by Western blot analysis (pretreated with the compounds for 3 h and then stimulated for 24 h with LPS).Results: Seven coumarins were isolated and identified as: ostruthin (1), ninhvanin (2), 8-geranyl-7-hydroxycoumarin (3), 6-(6′,7′-dihydroxy-3′,7′-dimethylocta-2′-enyl)-7-hydroxycoumarin (4), 6-(7-hydroperoxy-3,7-dimethylocta-2,5-dienyl)-7-hydroxycoumarin (5), 6-(2-hydroxyethyl)-2,2-dimethyl-2H-1-benzopyran (6), and luvangetin (7). Compounds 1–4 and 7 inhibited NO and PGE2 production in LPS-stimulated BV2 cells, with IC50 values ranging from 9.8 to 46.8 and from 9.4 to 52.8 μM, respectively. Ostruthin (1) and ninhvanin (2) were shown to suppress LPS-induced iNOS and COX-2 protein expression.Discussion and conclusion: The present study provides a scientific rationale for the use of P. trimera in the prevention and treatment of neuroinflammatory diseases. Ostruthin and ninhvanin might have potential therapeutic effects and should be considered for further development as new anti-neuroinflammatory agents.
Autumn precipitation over Central Vietnam is associated with an increase in the occurrence of tropical cyclones that lead to frequent flooding and pose a significant threat to lives and property. The present analyses reveal a pronounced decadal oscillation of autumn precipitation in Central Vietnam within the 8-11 year frequency band that is modulated by the East Pacific-North Pacific (EP-NP) teleconnection. The negative phase of the EP-NP pattern is associated with a positive sea surface temperature (SST) anomaly in the South China Sea (SCS) that induces low-level convergence, enhances convection, and increases precipitation over Central Vietnam and adjacent islands including Hainan (China) and the Philippines. This circulation feature around the SCS is embedded in a large-scale circulation associated with SST anomalies across the Pacific Ocean-i.e., cooling in the Eastern and Central tropical Pacific sandwiched by warming in the North and South Pacific as well as the Western Pacific Ocean. The positive phase of the EP-NP features opposite SST and circulation anomalies, with the result being reduced rainfall in Central Vietnam. This out-of-phase relationship and shared decadal spectral coherence between the EP-NP index and autumn precipitation in Central Vietnam might be useful for future climate predictions and flood management.
For the biopesticide, the observed decline in detectable protein was due to biological factors, possibly including the germination of B. thuringiensis spores, and was favoured by higher temperature. In contrast, for purified proteins, the decline in detectable protein was slower at low temperature, probably because the conformational changes of the soil-adsorbed protein, which cause fixation and hence reduced extraction efficiency, are temperature dependent. © 2016 Society of Chemical Industry.
Based on molecular and morphological data, the new genus Billolivia with five new species, B. longipetiolata, B. minutiflora, B. poilanei, B. vietnamensis and B. violacea, is described. IUCN conservation assessments are provided for the species.
