In this study, three good biofilm-forming yeast strains, including Candida viswanathii TH1, Candida tropicalis TH4 and Trichosporon asahii B1, were isolated from oil-contaminated water and sediment samples collected in coastal zones of Vietnam. These strains were registered in the GenBank database with the accession numbers JX129175, JX129176 and KC139404 for strain TH1, TH4 and B1, respectively. The biofilm formed by a mixture of these organisms degraded 90, 85, 82 and 67% of phenol, naphthalene, anthracene and pyrene, respectively, after a 7-day incubation period using an initial concentration of 600 ppm phenol and 200 ppm of each of the other compounds. In addition, this biofilm completely degraded these aromatic compounds, which were from wastewater collected from petroleum tanks in Do Xa, Hanoi after 14 days of incubation based on gas chromatography mass spectrometry analysis. To the best of our knowledge, reports on polycyclic aromatic hydrocarbon and phenol degradation by biofilm-forming yeasts are limited. The results obtained indicate that the biofilm formed by multiple yeast strains may considerably increase the degradation efficiency of aromatic hydrocarbon compounds, and may lead to a new approach for eliminating petroleum oil-contaminated water in Vietnam.
This work is aimed to assess the aerobic biotransformation of a branched side chain alkylbenzene, iso-pentylbenzene, by Candida viswanathii TH1. The yeast Candida viswanathii TH1 isolated from oil-polluted sediments collected in coastal zones in Vietnam exhibited as a strain that could better transform branched aromatic hydrocarbons in biofilm (pellicle) than in planktonic form. During incubation of TH1 as biofilm with iso-pentylbenzene, the seven intermediates produced were benzoic acid, phenylacetic acid, 2-methyl-4-phenyl-butan-1-ol, 2-hydroxy-phenylacetic acid, 2-methyl-4-phenylbutyric acid, succinic acid and iso-valerophenone as revealed by gas chromatography/mass spectra and high-performance liquid chromatography analyses. The occurrence of these intermediates showed that iso-pentylbenzene could be oxidized not only via mono- but also by a sub-terminal oxidation pathway. This is the first study on iso-pentylbenzene transformation by a biofilm-forming Candida viswanathii strain. The catabolic versatility of the biofilm-forming strain TH1 and its use for mono and sub-terminal oxidation during the transformation of iso-pentylbenzene enhance our understanding of the degradation of branched side chain phenylalkanes and give new insight into the potential role of such species in the transformation of other recalcitrant aromatic compounds.
Aromatic hydrocarbons such as naphthalene, pyrene are recalcitrant compounds found in oil contaminated areas including petroleum storage tanks, oil exploiting companies. These components are difficult to be degraded/transformed in the lack of oxygen conditions. Among anaerobic and micro-aerobic microorganisms, photosynthetic purple bacteria are the dominant group. Photosynthetic purple bacteria (PPB) are considered as aquatic organisms which are able to grow in anaerobic conditions by photosynthesis but without oxygen. This bacterial group has flexible metabolic types depending on living conditions, then they are widely distributed in nature. There are numerous publications on planktonic PPB which could use naphthalene and pyrene as carbon and energy sources. However, there is no publication on biofilm formed by PPB to degrade their aromatic compounds. In this research, 4 biofilm-forming PPB strains including DQ41, PY2, PY6 and DG12 were screened and estimated their pyrene and napthalene degradation capacity. These organisms demonstrated high biofilm forming ability. As biofilm types, their utilization efficiencies were upper 79% with the initial concentrations of naphthalene and pyrene of 200 and 250 ppm, respectively. These results may contribute to enlarge the number of biofilm-forming microorganisms to degrade/transform aromatic hydrocarbons in polluted area treatment in Vietnam.
Hiện nay, việc xử lý phenol trong nước thải ô nhiễm dầu theo phương pháp phân hủy sinh học được nhiều nhà khoa học trên thế giới cũng như ở Việt Nam quan tâm nghiên cứu. Chủng vi khuẩn DX3 được phân lập từ bể chứa nước thải kho xăng dầu Đỗ Xá, Thường Tín, Hà Nội sau 3 lần làm giàu liên tiếp trên môi trường muối khoáng Gost có bổ sung 50 mg/l phenol. Khuẩn lạc chủng vi khuẩn DX3 có hình tròn, màu trắng đục, nhớt, đường kính khoảng 1,5-2 mm, không ăn sâu vào thạch. Dưới kính hiển vi điện tử quét, tế bào có dạng hình que ngắn, kích thước tế bào vi khuẩn 1,14 µm × 2,67 µm, hai đầu vi khuẩn tròn, màng tế bào mỏng, nhẵn, có tiên mao ngắn xung quanh. Dựa vào đặc điểm hình thái và so sánh trình tự gene mã hóa 16S rRNA, đã xác định được chủng Bacillus sp. DX3 có độ tương đồng cao tới 99% với loài Bacillus megaterium IAM 13418. Trình tự nucleotide này đã được đăng ký trên GenBank với mã số KC845545. Chủng vi khuẩn này có khả năng phân hủy 99,95% phenol với nồng độ ban đầu là 150 mg/l trong môi trường khoáng sau 7 ngày nuôi cấy ở 30 o C. Với những kết quả trên, chủng vi khuẩn Bacillus sp. DX3 đã góp phần làm phong phú thêm số lượng các chủng vi khuẩn thuộc chi Bacillus nói riêng và hệ vi sinh vật có khả năng phân hủy phenol cao trong nước thải ô nhiễm dầu nói chung để phục vụ cho công nghệ phân hủy sinh học nguồn nước thải sau này. Trong số các chủng đó, chủng QN-B1 có khuẩn lạc tròn, gọn, bề mặt khô cằn, ở giữa có nhân lồi, trắng, đường kính 3-4mm là chủng có các khả năng ấy tốt hơn các chủng còn lại. Kết quả phân tích trình tự vùng bảo thủ cho nấm (ITS1-5.8r RNA-ITS2) và kết hợp với các đặc điểm hình thái, sinh lý, sinh hóa chủng B1 được định tên là Trichosporon asahii QN-B1. Chủng đã được đăng ký trên ngân hàng gen với mã số KC139404. Chủng này có khả năng phân hủy phenol và tạo biofilm tốt ở pH từ 3 đến 7; với nồng độ NaCl là 1,5% và ở nhiệt độ 30 o C, đồng thời chủng có khả năng sinh trưởng và phát triển tốt ở nồng độ phenol lên tới 200ppm. Sử dụng các điều kiện cho khả năng tạo màng của chủng, sau 7 ngày nuôi cấy, chủng QN-B1 đã phân hủy được 90% phenol, với nồng độ ban đầu là 200 ppm. Các kết quả này đã mở ra khả năng ứng dụng các chủng nấm men tạo màng sinh học nhằm xử lý các hợp chất gây ô nhiễm môi trường là các hydrocarbon thơm nói chung và phenol nói riêng.Từ khóa: Trichosporon asahii, màng sinh học, nấm men, phân hủy sinh học.
