Entre 51 genotipos de trigo cristalino (T. turgidum var. durum) de habito invernal se identificó a Mirlo 26 y Elinia 48, como resistentes a la raza BBG/ BPC de Puccinia triticina E. (agente causal de roya de la hoja) en etapa de plántula. En la progenie de la cruza con el cultivar susceptible de primavera Atred se determinó que en Elinia 48 la resistencia fue condicionada por un gen dominante y otro recesivo mientras que en Mirlo 26 se debió a un gen recesivo. La cruza entre los dos genotipos resistentes no produjo progenie susceptible en la F2 ni en las familias F3, por lo que se sugiere la similitud de por lo menos un gen de resistencia en ambos progenitores. Mirlo 26 es susceptible mientras que Elinia 48 permaneció resistente a la raza BBB/BNJ virulenta a Lr61 identificada en el 2008, por lo que fue posible determinar que el gene recesivo identificado en Mirlo 26 es el gen que ambos progenitores tienen en común. En las poblaciones F2 y F3 de la cruza Mirlo 26 x Elinia 48 inoculadas con la raza BBB/BNJ virulenta sobre el gen recesivo la segregación fue de 3:1, que corresponde a la segregación para un gen dominante. Como resultado de las cruzas de los progenitores de invierno con el progenitor de primavera se dispone ahora genotipos de trigo cristalino de primavera que poseen el gen dominante y líneas con el gen recesivo.
El cultivo del trigo en México actualmente enfrenta la roya de la hoja (Puccinia triticina E.) que ocasiona pérdidas hasta de 100% del rendimiento. Esta investigación evaluó la respuesta de tres genotipos de trigo: “Mirlo 26”, “Elinia 48” y “Atred” a la roya de la hoja BBG/BPC bajo condiciones in vitro. Los dos primeros, considerados tolerantes (R) y el tercero susceptible (S). La germinación in vitro de los tres genotipos se logró en medio de cultivo MS (1962) adicionado con sacarosa (30 g L-1); 2,4-D (20.3 μM) y Kin (5.8 μM). Además de generar plántulas, las semillas formaron callos. De las plántulas se disecaron secciones transversales basales de tallo y se sembraron en medio MS (1962) con BA (11.1 μM) y AIA (1.0 μM) para inducir organogénesis directa. En este mismo medio se sembraron los callos para inducir organogénesis indirecta. La regeneración de plántulas enraizadas se logró en ambas rutas morfogénicas en el mismo medio de cultivo y concentraciones hormonales a las cuatro semanas. La aclimatación fue 90% en sustrato de agrolita y peat-moss (1:1). La evaluación de roya de la hoja en plantas aclimatadas tuvo porcentajes de infección de 21% en el genotipo “Atred” (S), 10% en “Mirlo 26” (R) y 10% en “Elinia 48” (R). En plántulas in vitro los porcentajes fueron de 70% para “Atred”, 40% para “Mirlo 26” y 30% para “Elinia 48” al aplicar 0.03 mg mL-1 de esporas. Este estudio es el primer reporte que evalúa la tolerancia de genotipos de trigo a la roya de la hoja usando el cultivo de tejidos vegetales in vitro.
La roya de la hoja, causada por el hongo Puccnia triticina E., es una enfermedad que cuando se presenta, genera pérdidas hasta del 100 % en el rendimiento de grano del trigo cristalino (Triticum turgidum ssp. durum L.). Como medida de protección genética en la construcción piramidal de nuevos genotipos resistentes es necesario identificar en el germoplasma con resistencia a esta enfermedad, el número de genes involucrados y su acción génica. Con este propósito, durante el ciclo de cultivo otoño-invierno 2017- 2018, se cruzaron los progenitores susceptibles Atred#1 y Atred#2 con las líneas resistentes de trigo cristalino procedentes de Etiopía WC-2 no. 100, DW-K2 no. 47, Oda, 2000/01 population FR. no. 43, 2000/01 population 37-30 BDI no. 63 y 2000/01 population 37-30 BDI no. 12. Las generaciones filiales de las cruzas se obtuvieron alternadamente en CIMMYT-Batán, Estado de México y CIMMYT-CENEB, Ciudad Obregón, Sonora, hasta obtener las Familias F3 y F4. Con base en los análisis de segregantes de tales familias, se encontró que, en las seis líneas etíopes, la resistencia a la roya de la hoja es conferida por dos genes dominantes.
