The neurotransmitter serotonin (5-HT) is involved in mood disorder aetiology and it has been reported that (organophosphate) OP exposure affects 5-HT turnover. The aim of this study was to elucidate the mechanism underlying OP effects on the adult 5-HT system. First, acute in vivo administration of the OP diazinon (0, 1.3, 13 or 39 mg/kg i.p.) to male Hooded Lister rats inhibited the activity of the cholinergic enzyme acetylcholinesterase in blood and in the hippocampus, dorsal raphe nucleus (DRN), striatum and prefrontal cortex. Diazinon-induced cholinesterase inhibition was greatest in the DRN, the brain's major source of 5-HT neurones. Second, acute in vivo diazinon exposure (0 or 39 mg/kg i.p.) increased the basal firing rate of DRN neurones measured ex vivo in brain slices. The excitatory responses of DRN neurones to α1-adrenoceptor or AMPA/kainate receptor activation were not affected by in vivo diazinon exposure but the inhibitory response to 5-HT was attenuated, indicating 5-HT1A autoreceptor down-regulation. Finally, direct application of the diazinon metabolite diazinon oxon to naive rat brain slices increased the firing rate of DRN 5-HT neurones, as did chlorpyrifos-oxon, indicating the effect was not unique to diazinon. The oxon-induced augmentation of firing was blocked by the nicotinic acetylcholine receptor antagonist mecamylamine and the AMPA/kainate glutamate receptor antagonist DNQX. Together these data indicate that 1) acute OP exposure inhibits DRN cholinesterase, leading to acetylcholine accumulation, 2) the acetylcholine activates nicotinic receptors on 5-HT neurones and also on glutamatergic neurones, thus releasing glutamate and activating 5-HT neuronal AMPA/kainate receptors 3) the increase in 5-HT neuronal activity, and resulting 5-HT release, may lead to 5-HT1A autoreceptor down-regulation. This mechanism may be involved in the reported increase in risk of developing anxiety and depression following occupational OP exposure.
Os resultados sugerem que alterações do metabolismo mineral e a pHPT podem ocorrer após um ano do Tx, principalmente em pacientes com níveis elevados de PTH pré-Tx.
OBJETIVO: Analisar a eficácia da suspensão de carvão inerte na marcação de lesões impalpáveis mamárias e as alterações morfológicas associadas ao seu uso, além de determinar se há ou não prejuízo na interpretação destas lesões pelo patologista. MÉTODOS: Foram atendidos 135 casos de lesões impalpáveis mamárias, previamente marcadas com suspensão de carvão. As lâminas coradas pelo método de HE contendo pigmentos de carvão foram analisadas ao microscópio óptico, onde se realizou avaliação quantitativa e qualitativa da resposta inflamatória e determinação da presença ou não de prejuízo ao diagnóstico anatomopatológico. Os índices avaliados quantitativamente quanto à resposta inflamatória foram as contagens de linfócitos, células gigantes e neutrófilos, todos observados e quantificados em campos de grande aumento. Foi, ainda, avaliada a quantidade e distribuição de carvão presente nas lesões. RESULTADOS: Na avaliação quantitativa e qualitativa da resposta inflamatória observou-se, independente da quantidade de carvão, granulomas a "corpo estranho" em todas as amostras. Em relação à resposta inflamatória linfocitária 5,19% das amostras não apresentaram infiltração linfocítica, sendo que em 82,22% ela era de discreta intensidade e em 12,59% de moderada intensidade. Quanto à resposta inflamatória aguda, observou-se ausência total de exsudação neutrofílica em 42,96% dos espécimes, exsudação neutrofílica discreta em 42,22%, moderada em 11,11% e acentuada, sob forma de microabscessos, em 3,7% dos casos. Nesta série foram observados cinco nódulos de carvão, posicionados paralesionalmente, e que não geraram interferência na análise morfológica. CONCLUSÃO: Este método é marcador eficaz de lesões impalpáveis, de baixo custo, alta efetividade, sem prejuízo à análise histológica, além de ser de fácil execução; é confortável para a paciente e de grande auxílio ao cirurgião e patologista na localização destas lesões.
The effectiveness of hemodialysis is completely dependent on the quality of the patient’s vascular access; thus, appropriate assessment of ateriovenous fistulas and grafts is of critical importance. Point of care ultrasound (POCUS) can be an additional useful skill at the bedside for assessment of the hemodialysis vascular access. In this paper we discuss the basic terminology and techniques that can be employed in the POCUS exam of vascular accesses. We also delineate the current recognized criteria for access maturation and common issues that can be diagnosed with the support of POCUS.
