BACKGROUND Once peroxidase purification can improve substrate degradation capacity, peroxidase from rice bran was purified by expanded bed adsorption (EBA) coupled to diafiltration/ultrafiltration (DF/UF) and applied for deoxynivalenol (DON) reduction. RESULTS The DF/UF step was optimized according to membrane molecular weight cutoff (MWCO), DF cycles number, operation temperature and CaCl2 addition to the DF buffer. DON reduction tests were carried out with both clarified and purified peroxidase. The use of MWCO of 10 kDa, 10 °C, six DF cycles and adding 4 mmol L−1 CaCl2 to DF buffer promoted enzyme purification of 4.4‐fold and enzymatic recovery of 79.4%. The global purification process combining two techniques resulted in enzyme purification of 75.1‐fold with an enzymatic recovery of 22.8%. Peroxidase purified by EBA combined with DF/UF resulted in DON reduction of 81.7% in a model system. CONCLUSION Peroxidase purification was achieved by scale‐up integrated strategies. The use of purified peroxidase from agricultural by‐products could be a promising alternative for enzymatic DON levels reduction. © 2017 Society of Chemical Industry
RESUMOAs peroxidases pertencem à família das oxidorredutases, e têm por função catalisar a oxidação de uma variedade de substratos orgânicos tendo o peróxido de hidrogênio como molécula aceptora. Inúmeras técnicas são utilizadas na purificação de peroxidases incluindo a cromatografia de troca iônica em coluna de leito expandido, a qual apresenta vantagens como maior produtividade de processo e interação adsorvente-molécula alvo. Este trabalho teve como objetivo determinar as curvas de ruptura em três diferentes graus de expansão, visando à posterior purificação de peroxidase por cromatografia de troca iônica em coluna de leito expandido. Neste estudo, foi verificado que o uso do grau de expansão 2,5 proporcionou maior Q10% (0,1851 U/mL de resina) frente às demais condições estudadas, se mostrando o mais adequado para posterior purificação de peroxidase por esta técnica.Palavras-chave: capacidade dinâmica de adsorção, leito expandido, Streamline TM SP. INTRODUÇÃOAs peroxidases são enzimas presentes em todos os organismos capazes de catalizar reações de óxido-redução, graças a vários peróxidos que atuam como receptores de elétrons (FAWAL et al., 2013). Estas enzimas estão amplamente distribuídas na natureza e podem ser extraídas de vegetais, tecidos animais e alguns fungos (BALASUBRAMANIAN; BOOPATHY, 2013). Considerando sua natureza oxidativa, especificidade e atuação em condições moderadas, inúmeras aplicações são atribuídas às peroxidases em áreas como biomédica, clínica, biotecnológica e industrial (SONTÜRK; RAMAZAN;ÖZDEMIR, 2014). O uso final das peroxidases irá definir se estas necessitam ser submetidas a um processo de purificação.Dentre as ténicas utilizadas na purificação de peroxidases destaca-se cromatografia de troca iônica, a qual se baseia na competição dos íons de interesse e contaminantes pelos grupos carregados da fase estacionária (PESSOA & KILIKIAN, 2005). A utilização desta técnica em modo expandido tem se mostrado como alternativa na obtenção de peroxidases purificadas (GAUTÉRIO et al., 2015), apresentando vantagens como obtenção do produto purificado em grandes volumes, maior interação adsorvente-molécula alvo e possibilidade de aumento de escala (TOLEDO et al., 2007). O comportamento do processo de adsorção em função da expansão do leito pode ser verificado através das curvas de ruptura, as quais indicam a quantidade do produto de interesse que pode se ligar ao trocador iônico (NAYAK; PONRATHNAM; RAJAN, 2001). De forma prática, a curva de ruptura serve como indicativo do volume que deve ser alimentado à coluna cromatográfica sem que haja perda do composto durante a alimentação.
