Работа посвящена проверке предположения о возможном участии системы 2' ,5'олиго(А)синтетаза-РНКаза L в переориентировке клеточного метаболизма в процессе перехода печени от покоя к делению, вызванного частичной гепатэктомией (ЧГЭ). Исследовали активность 2' ,5'олиго(А)синтетазы и состав образуемых олигоаденилатов в ядерной и цитоплазматичекой фракци ях, изолированных из регенерирующей печени в течение первых 12 ч после операции. Обнаружено, что изменения активности происходят в интервале 0,5-3 ч после ЧГЭ, охватывающем период переориентировки клеточного метаболизма. Изменения проявляются в снижении ферментатив ной активности и в повышении соотношения ди-и тримеров 2',5'олигоаденилатов в ядерной фракции и в увеличении ферментативной активности в цитоплазматической фракции, что исходя из свойств олигоаденилатов может приводить соответственно к снижению и повышению активности РНКазы L. Согласно ранее полученным данным, описанные изменения совпадают по времени с задержкой новообразованной РНК в ядре и с перераспределением рибосом на эндоплазматическом ретикулуме. Это поддерживает нашу гипотезу о потенциальной роли системы 2',5'олиго(А)синтетаза-РНКаза L в переориентировке клеточного метаболизма на уровне РНК.
Partial hepatectomy (PHE) stimulates an intact part of the liver to exchange the existing «quiescence» program for the proliferative one. Several changes between 0.5 and 3 h after PHE are considered as manifestations of the abolishment of «quiescence» program' a temporal decrease of either RNA synthesis or accumulation of the newly formed RNA; a restricted variability of RNA transcripts; a partial retention of newly synthesized RNA within the nuclei thereby providing less RNA for the cytoplasm; previously obtained data about transient dissociation of ribosomes from endoplasmic reticulum. An involvement of the 2' ,5'-oligo(A Synthetase-RNAse L system in the process is suggested by up-and down-regulation of 2',5'-oligo(A)synthetase activity in the cytoplasm and in the nucleus, respectively, and by up-regulation of RNAse L content in regenerating liver. The production of IFN alp, an inducer of the 2',5'-oligo(A Synthetase-RNAse L system, is also increased during transition period. A specific role of sinusoidal cells, the main producers of IFN alp in the liver, in the abolishment of the old program is strongly suggested.
Досліджено глутатіонтрансферазну активність цитозолю в зразках плаценти людини з різним генотипом цитохрому Р450 1А1 і глутатіонтрансфераз класу Р1 та МІ, а також експресію глутатіонтрансферази Р1 у культурі клітин хоріокарциноми людини за умов короткочасної індукції цитохрому Р450 1А1 бензпіреном. Виявлено, що в клітинах плаценти у фізіологічних умовах і внаслідок короткочасної індукції цитохрому Р450 1АЇ глутатіонтрансферазна ак тивність цитозолю та експресія глутатіонтрансферази позитивно корелюють з активністю/ек спресією цитохрому Р450 JAJ.
For the first time the prevalence of Ñ677Ò methylenetetrahydrofolate reductase (MTHFR) genotypes was detected in a cohort of Ukrainian population. We have analyzed the association of polymorphism of MTHFR alone and in its combination with polymorphic genes coding for glutathione S-transferase P1, glutathione S-transferase M1, and cytochrome P450 1A1 with cytosolic glutathione S-transferase activity in human placenta samples. The substitution of Ñ677÷Ò677 in at least one allele of MTHFR gene is accompanied by the tendency of GST activity to increase. The tendency remains irrespective of allelic variants of other three genes in specified combinations. The possible relation between the MTHFR allelic variants and cytosolic glutathione S-transferase activity is discussed.
