The calcium-independent receptor of ␣-latrotoxin (CIRL), a neuronal cell surface receptor implicated in the regulation of exocytosis, is a natural chimera of the cell adhesion protein and the G protein-coupled receptor (GPCR). In contrast with canonic GPCRs, CIRL consists of two heterologous non-covalently bound subunits, p120 and p85, due to endogenous proteolytic processing of the receptor precursor in the endoplasmic reticulum. Extracellularly oriented p120 contains hydrophilic cell adhesion domains, whereas p85 resembles a generic GPCR. We determined that the site of the CIRL cleavage is located within a juxtamembrane Cys-and Trp-rich domain of the N-terminal extracellular region of CIRL. Mutations in this domain make CIRL resistant to the cleavage and impair its trafficking. Therefore, we have named it GPS for G protein-coupled receptor proteolysis site. The GPS motif is found in homologous adhesion GPCRs and thus defines a novel receptor family. We postulate that the proteolytic processing and twosubunit structure is a common characteristic feature in the family of GPS-containing adhesion GPCRs.Cell adhesion receptors provide physical links between cell plasma membranes and the extracellular matrix. These receptors have large extracellular domains that contain characteristic structural modules that are directly involved in cell-to-cell and cell-to-matrix interaction. They can also function as signaling receptors, providing the cell with critical information required for proper tissue growth and development. The signaling function of cell adhesion receptors has been primarily attributed to their connection to the tyrosine phosphorylation pathway via tyrosine kinase or phosphatase domains in their intracellular region or to the recruitment of other tyrosine kinases upon activation. The recent discovery of heptahelical receptors with large extracellular cell adhesion-like domains opens an intriguing possibility that cell-to-cell and cell-to-matrix interaction can be also coupled to G protein signaling (1, 2).By this mechanism, cells should be able to produce fast and sensitive responses to physical contacts with the extracellular environment.Putative cell adhesion GPCRs 1 are linked to different cellular functions in a variety of tissues. Receptor-like protein-tyrosine phosphatase sigma (PTP ) is essential for neuronal development and function. Here we report that PTP is a target of ␣-latrotoxin, a strong stimulator of neuronal exocytosis. ␣-Latrotoxin binds to the cell adhesion-like extracellular region of PTP . This binding results in the stimulation of exocytosis. The toxin-binding site is located in the C-terminal part of the PTP ectodomain and includes two fibronectin type III repeats. The intracellular catalytic domains of PTP are not required for the ␣-latrotoxin binding and secretory response triggered by the toxin in chromaffin cells. These features of PTP resemble two other previously described ␣-latrotoxin receptors, neurexin and CIRL. Thus, ␣-latrotoxin represents an unusual example of the neurotoxin that has three independent, equally potent, and yet structurally distinct targets. The known structural and functional characteristics of PTP , neurexin, and CIRL suggest that they define a functional family of neuronal membrane receptors with complementary or converging roles in presynaptic function via a mechanism that involves cellto-cell and cell-to-matrix interaction.
CIRL (the calcium-independent receptor of α-latrotoxin), a neuronal cell surface receptor implicated in the regulation of exocytosis, is a member of the GPS family of chimeric cell adhesion/G protein-coupled receptors. The predominant form of CIRL is a membrane-bound complex of two subunits, p120 and p85. Extracellularly oriented p120 contains hydrophilic cell adhesion domains, whereas p85 is a heptahelical membrane protein. Both subunits are encoded by the same gene and represent products of intracellular proteolytic processing of the CIRL precursor. In this study, we demonstrate that a soluble form of CIRL also exists in vitro and in vivo. It results from the further cleavage of CIRL by a second protease. The site of the second cleavage is located in the short N-terminal extracellular tail of p85, between the GPS domain and the first transmembrane segment of CIRL. Thus, the soluble form of CIRL represents a complex of p120 non-covalently bound to a 15 amino acid residue N-terminal peptide fragment of p85. We have previously shown that mutations of CIRL in the GPS domain inhibit intracellular proteolytic processing and also result in the absence of the receptors from the cell surface. Our current data suggest that although CIRL trafficking to the cell membrane is impaired by mutations in the GPS region, it is not blocked completely. However, at the cell surface, the non-cleaved mutants are preferentially targeted by the second protease that sheds the extracellular subunit. Therefore, the two-step proteolytic processing may represent a regulatory mechanism that controls cell surface expression of membrane-bound and soluble forms of CIRL.

Подтверждена обнаруженная нами ранее абсолютная зависимость активности рибозима от присутствия в реакционной среде спермидина (помимо Mg). Определена оптимальная концентрация ионов Mg , составившая 5-10 мМ. При определении температурного оптимума реакции, равного 30 °С, выявлено активирующее влияние 20 % формамида при температуре 15 °С. Установлена зависимость эффективности расщепления субстратной РНК от времени реакции.
Методом часткового нуклеазного гидролізу показано, що іонний склад середовища суттєво впливає на третинні взаємодії в молекулі рибозиму, змінюючи його просторову структуру.
Представлены исследования хромосомно-плазмидного детерминирования признаков антибиотикорезистентности у штаммов кампилобактерий, выделенных из различных источников. Анализ антибиотикограмм штаммов кампилобактерий показал, что 12 из них (31,6 %) обладали множественной лекарственной устойчивостью. Установлено наличие плазмид, детерминирующих антибиотикорезистентность у 7 из 38 исследован ных культур (18,4 %). Обработка бромистым этидием приводила к изменению фенотипических свойств плазмидосодержащих штаммов. После элиминации плазмид моле кулярной массой (мм) (40-70)-10 6 утрачивалась устойчивость штаммов к тетрацик лину и эритромицину, а плазмид мм (1,5-6) • /О 6-к канамицину. Плазмидный ха рактер множественной устойчивости кампилобактерий может служить дополнительным критерием эпидемической опасности штаммов. Введение. Имеются единичные сообщения, посвященные генетической организации бактерий рода Campylobacter. В частности показано, что штаммы кампилобактерий различных видов содержат плазмидную ДНК, функция которой заключается в детерминировании антибиотико резистентности. Ряд работ посвящен изучению устойчивости кампило бактерий к тетрациклину, обусловленной плазмидой pUA466 [1], и ка намицину, зависящей от наличия плазмиды pS1178 [2]. Однако в ра ботах разных авторов имеются некоторые противоречия в характерис тике плазмидной ДНК У изучаемых штаммов, касающиеся ее количе ства и величины, что определяется, вероятно, использованием разных методов выделения плазмид [3]. При организации комплексного эпидемиологического надзора необ ходимо установление природы устойчивости и спектра антибиотикоре зистентности, обусловленного трансмиссивными R-факторами. В настоя щее время мы не располагаем данными литературы по исследованию плазмидного профиля отечественных штаммов кампилобактерий. В связи с этим задачей нашей работы явилось изучение хромосом но-плазмидного детерминирования признаков антибиотикорезистентнос ти у штаммов кампилобактерий, выделенных из различных источников. Материалы и методы. Исследованы 38 штаммов бактерий рода Campylobacter различного происхождения, полученных от больных, сельскохозяйственных животных и птиц, объектов окружающей среды, и эталонные. Их характеристика приведена в таблице. В качестве плотной питательной среды для штаммов кампилобак терий использовали железо-эритрит кровяной агар [4]. Культивиро вание проводили при 42 °С в атмосфере 8-10 % С0 2 , создаваемой при помощи коммерческих газогенерирующих пакетов «Кампилогаз» (НИИ «Синтез», Борислав), в течение 48-72 ч. Чувствительность кампилобактерий к антибиотикам определяли методом диффузии в агар с использованием дисков.
