Die Phasen werden getrennt und die waBrige Phase wird rnit CH2C12 extrahiert. Die vereinigten organischen Phasen werden mit gesattigter NaCI-Losung gewaschen, mit Na2S04 getrocknet und eingeengt. Der Ruckstand wird mit ToluoVDiethylether (9 i 1) an Kjeselgel 60 Merck(R) sc getrennt: Der Vorlauf enthalt 1, die gelbe Hauptfrdktion besteht aus 2') (Ausb. 30 %) und der orange-rot gefarbte Nachlauf aus wenig 3. 5,S'-Dimethoxy-2,2'-dipivnioyl-l '-hydroxy-3,3'-binaphrhyl-l ,I-chinon (3)Der Ansatz aus 0,26 g (1 mmol) 1 und 0,6 g Tetrabutylammoniumhydrogensulfat in 30 ml CH2C12 sowie 1,5 g (5,5 mmol) NO(S0,K)z in 30 ml 15proz. Na2C03-Losung wird 8-12 h intensiv bei RT geriihrt. Wenn die DC-Kontrolle den Verbrauch von 1 anzeigt, wird die Reaktion abgebrochcn und der Ansatz wie bei 2 aufgearbeitet und sc getrennt. In mehreren Mitt. haben wir uber die Validitat der kontinuierlichen Ultrafiltration zur raschen und sicheren Bestimmung von Proteinbindungsparametern berichtet. Voraussetzung fiir die Anwendbarkeit der Methode auf hochgradig an Protein gebundene Substanzen ist die Vefigbarkeit einer Detektionsmethode, die es gestattet, den Arzneistoff im DurchfluRverfahren in Konzentrationen zu messen, die zwei bis drei GroRenordnungen unter seinem Plasmaspiegel liegen. Im Gegensatz zu anderen Anticoagulantien vom 4-Hydroxycumarintyp zeigt Acenocoumarol (Sintrom') keine Fluoreszenz. Dies hatte zur Folge, daB seine EiweiBbindung bisher nur diskontinuierlich unter Verwendung radioaktiv markierten Materials14) oder fur unphysiologisch niedrige Proteinkonzen-trationen5r6) respektive kontinuierlich in unphysiologisch hohen Substanzkonzentrationen') durch UV-Detektion bestimmt werden konnte.Es lassen sich jedoch reproduzierbare Bestimmungen im UV-Bereich bis zu Konzentrationen von cf = 10nmolll entsprechend 2,5 bis 5 Milliextinktionen durchfiihren, wenn ein empfindlicher und stabiler Detektor vorhanden ist . Die DurchfluBkuvette des UV-Detektors mu13 bei diesen Messungen exakt thermostatisiert werden. Aus dem unter derartigen Bedingungen von uns erhaltenen kontinuierlichen Satz von MeBergebnissen sind die Werte fiir einige Konzentrationen herausgegriffen und in Tab. 1 zusammengefdt worden. Man erkennt, daB a im Bereich von 2,5-10pmoM praktisch konstant und etwa doppelt so groB wie bei Warfarin ist. Hieraus extrapolieren wir, daB auch im therapeutisch genutzten Konzentrationsbereich um ctot = 1 pmoYl') ahnliche Bindungsverhaltnisse vorliegen werden. Die Konzentration an ungebundenem Acenocoumarol steigt wegen c,= a-c,,,,.iiber den MeRbereich linear an. Die Wechselwirkung der anticoagulanten Titelverbindungen wurde im Konzentrationsbereich von 2,5-15 pmoUI rnit Phenylbutazon in Konzentrationen bis zu 0,6mmoV1 untersucht. Die freie Konzentration von Phenprocoumon wurde um bis zu 310 %, die von Warfarin bis zu 378 % erhöht. Fiir Methylsulfinylwarfarin (MSW) wurde nur eine Steigerung um 33 % beobachtet, die iiberdies bei Phenylbutazonkonzentrationen von 0,2-0,6mmoU1 nahezu konstant war. MSW sollte daher zur Erhöhung der Arzneimittelsicherheit bei der Thromboseprophylaxe mit oralen Anticoagulantien beitragen können.Interactions of the title anticoagulants in concentrations of 2.5 to 15 pmoUL with phenylbutazon up to concentrations of 0.6 mmol/L were determined. Zur Überprüfung dieser These haben wir daher die Verdrängung von Phenprocoumon, Warfarin und MSW aus ihrer Bindung an Humanserumalbumin (HSA) durch Phenylbutazon gemessen. Die Proteinbindung von drei Methylthioderivaten des Warfarins wurde mit Hilfe der kontinuierlichen Ultrafiltration bestimmt. Das 4''-Methylsulfinylwarfarin (2) hat im Konzentrationsbereich 2.5-12.5 p o V l einen 7-9 mal hoheren freien Anteil a als Warfarin (1). Da 2 auch im Tierexperiment giinstige pharmakokinetische Eigenschaften zeigt, konnte es zur Verbesserung der Arzneimittelsicherheit auf dem Sektor der Anticoagulantien beitragen.
