O mercado de produtos zero lactose está expandindo e conquistando, a cada dia, mais e mais consumidores, principalmente aqueles com diferentes graus de intolerância a lactose. O custo operacional para fabricação destes produtos é elevado, existindo a necessidade de estudos que visem maior eficiência no processo, garantindo maior rentabilidade para a indústria e um produto mais acessível ao consumidor. O método de imobilização é uma das alternativas a serem utilizadas, pois possibilita a reutilização das enzimas imobilizadas. Com isto, o objetivo deste estudo foi imobilizar a enzima beta-galactosidase de Kluyveromyces lactis em alginato de sódio com cloreto de cálcio e avaliar a atividade desta enzima imobilizada na hidrólise de soro de queijo, a cada reciclo. Os testes foram realizados nas seguintes condições: pH 6,2 do tampão fosfato de potássio/hidróxido de sódio, pH 6,3 do soro doce, 0,75 mL de solução enzimática, 0,8% (m/v) de alginato de sódio, 0,3% (m/v) de cloreto de cálcio e temperatura de 34 °C. Os resultados indicaram que a maior atividade da enzima foi observada no primeiro ciclo, hidrolisando 52% da lactose presente no soro, seguido pelo primeiro reciclo, 15%, segundo reciclo, 5%, terceiro reciclo, 3% e quarto reciclo, 2%. A diminuição na eficiência da hidrólise ao longo dos reciclos se deu possivelmente por escape da enzima através dos poros do suporte. As concentrações finais de lactose nos soros hidrolisados, do primeiro ciclo até o quarto reciclo não reduziram a ponto de atender a legislação para produtos zero lactose, no entanto, foi possível imobilizar a enzima e realizar a hidrólise do substrato de acordo com a metodologia estudada. Portanto, mais estudos visando o aumento da eficiência dessa hidrólise se mostram promissores.
