0rganver~inderungen bei Ratten, welche mit gro•en Mengen synthetischer oestrogener Stoffe behandelt wurden 1. I. Mitteilung. Mit 10 Textabbildungen. Die Entdeckung, da~ verhs einfache Substanzen, wie z.B. das 4.4'-Dioxydiphenyl bei kastrierten Ratten die spezifische Wirkung des weiblichen Sexualhormons ersetzen kann, verdanken wir Dodds und Lawson. Diese Forscher haben ferner beschrieben, dal~ das Phenolderivat des ~therischen Ols dos Anethol, das P-anol, (p-propenylphenol), eine ~hnlich, jedoch sti~rker wirkende Substanz ist. Dodds, .Fitzgerald und Lawson konnten in einer ganzen Reihe der Diphenyls eine ~hnliche oestrogene Wirkung nachweisen. Dieselben Forscher entdeckten auch, da~ die Einfiihrung yon ~thylenbindungen zwischen die Kohlenatome a. und fl die Wirksamkeit der Prs stark erhSht. Sie gelangten derart zu einer ganzen Reihe yon Stilbenderivaten, unter welchen nach Dodds, Lawson, Robinson, Goldberg, Kreitmair und Siecl~mann das 4.4'-Dioxyg-/~-di~thylstilben oder Di-~thy]stilboestron das wirksamste ist. Die Formel lautet: ClsH2002.Oestrogene Einheit bei kastrierten Ratten 0,3--0,4 ~ ; es ist also 2--3real so wirksam wie das Oestron. Nach Kreitmair und Sieckmann soil die wirksame Gabe des Oestrostilbens 0,13 ~, die des Diazetats des Oestrostilbens 0,17 7, die des Di-Propionates 0,31 y betragen. Die wirks~me Dosis des Oestrons ist 0,17y, die des Oestradiolbenzoats 0,1 y. Das Oestrostilben ist also etwas wirksamer, als der eigene essigsaure Ester, beide sind 2mal so wirksam, wie der propionsaure Ester und 5mal wirksamer als das Oestron. Das Oestrostilben erreicht beinahe die Wirksamkeit des Oestradiolbenzoates. Nach den Untersuchungen yon Kreitmair und Siec~mann soll das Oestrostilben bei peroraler Gabe 20real so wirksam sein wie das k6rpereigene Oestron.Nach den Angaben der oben erw~hnten Verfasser und zahlreicher Nachuntersucher stimmt die qualitative Wirkung der synthetischen 1 Der Sehriftwaltung des Arehivs fiir Gynakologie iibergeben am 31. Die Entdeekungen yon Dodds und Mitarbeitern, dab gewisse synthetiseh hergestellte Substanzen eine mit jener des Follikelhormons fibereinstimmende oestrogene Wirkung austiben, veranlaBte eine groBe Reihe yon Forsehern zur biologischen Priifung dieser Stoffe. In unserer vorausgehenden Mitteilung haben wit jene Untersuehungen aufgezghlt, naeh welehen die synthetischen oestrogenen Stoffe sowohl in kleinen, wie aueh in groBen Gaben, in ihren qualitativen und quantitativen Wirkungen mig den Wirkungen des Follikelhormons weitgehend tibereinstimmen. Eine natiirliehe Folge dieser theoretiseh hoehinteressanten Versuehe war, dab die Kliniker die synthetischen oestrogenen Substanzen frtihzeitig in der Heilkunde zu verwert.en versuehten. Heute weist aueh bereits dieser loraktisehe Tell der Frage eine ausgedehnte Literatur auf. Guldenberg ist es zum erstenmM gelungen, mit 20 mg Stilboestrol und 30 KE synthetisehem Progesteron bei einer operativ kastrierten Kranken eine wirkliehe Menstruation zu erzeugen. H. Buschbeclc und K. Hausknecht besehreiben 3 Fglle, in welehen sie bei seit Jahren amenorrhoisehen Kranken mit 4, bzw. 5real 5 mg Stilboestrol Menstruationen, bei einer 58j/~hrigen Frau nach 7jghriger Menopause eine Sehleimhauthyperplasie hervorrufen konnten. Dieselben Forseher kamen in ihren therapeutisehen Versuehen zu dem Ergebnis, dab die Wirkung des Stilboestrols aui die Geb~rmuttermuskulatur, auf die Seheide, auf die Milchabsonderung mit jener des Follikelhormons vSllig tibereinstimmt and dab es auch die Ausfallserseheinungen hervorragend gut beeinfluBt, l~ber gleiehe Erfolge beriehten aueh Erhardt, Krammann und Schii[er auf Grund ihrer in/~hnlieher Riehtung ausgefiihrten Untersuehungon. Die klinisehen Untersuehungen haben bewiesen, dab die synthetisehen oes~rogenen Substanzen im Indikationsgebie~ des kSrpereigenen Follikelhormons iiberall mit vollem Erfolg beniitz~ werden kSnnen.
