We have previously shown that high density lipoprotein is the most abundant protein in the carp plasma and displays bactericidal activity in vitro. Therefore the aim of this study was to analyze the contribution of its principal apolipoproteins, apoA-I and apoA-II, in defense. Both apolipoproteins were isolated by a two step procedure involving affinity and gel filtration chromatography and were shown to display bactericidal and/or bacteriostatic activity in the micromolar range against Gram-positive and Gram-negative bacteria, including some fish pathogens. In addition, a cationic peptide derived from the C-terminal region of carp apoA-I was synthesized and shown to posses antimicrobial activity (EC 50 ¼ 3-6 lM) against Planococcus citreus. This peptide was also able to potentiate the inhibitory effect of lysozyme in a radial diffusion assay at subinhibitory concentrations of both effectors. Finally, limited proteolysis of HDL-associated apoA-I with chymotrypsin in vitro was shown to generate a major truncated fragment, which indicates that apoA-I peptides liberated in vivo through a regulated proteolysis could also be involved in innate immunity.
Mycobacterium avium ssp. paratuberculosis (MAP) is the etiologic agent of paratuberculosis and it potentially plays a role in Crohn's disease. In humans, the main route of transmission of MAP might be the intake of contaminated milk and dairy products. Considering that MAP has already been detected in many types of cheese in different counties, and that Coalho cheese is an important dairy product in northeastern Brazil, the aim of this study was to report the first detection of MAP in retail Coalho cheese in Brazil by PCR and culture. Of 30 retail Coalho cheese samples, 3 (10%) amplified fragments of a similar size to that expected (626 bp) were obtained and viable MAP was recovered by culture from 1 (3.3%) sample. The DNA from the positive culture sample was sequenced and showed 99% identity with the insertion sequence IS900 deposited in GenBank. It was possible to identify the presence of MAP-specific DNA in the analyzed samples for the first time in Brazil, and to recover viable cells from retail Coalho cheese.
Green manure may lead to physical, chemical, and biological improvements to the soil. However, the information on its use in the Brazilian Northeast is scarce. The aim of this study was to assess the phenological cycle and phytomass productivity of seven legume species. This study was conducted in the second semester of 2015 in a completely randomized design with seven treatments and four replications. Plant height, fresh mass of shoot (FMS), dry mass of shoot (DMS), fresh mass of root (FMR), and dry mass of root (DMR) were determined at 100 days after sowing. Germination, flowering, and pod maturation were also assessed. The highest averages of FMS were observed in Crotalaria breviflora and Crotalaria mucronata, followed by Canavalia ensiformis, whereas the highest averages of DMS were observed in C. breviflora, C. ensiformis, C. mucronata, and Cajanus cajan cv. IAPAR 43. The highest averages of FMR were obtained by C. breviflora and C. mucronata, followed by C. cajan cv. Fava Larga. The species C. mucronata, C. cajan cv. IAPAR 43, and C. breviflora presented the highest averages of DMR. In addition, C. juncea, C. ensiformis, and C. cajan cv. IAPAR 43 were the earlier species regarding flowering, while C. cajan cv. Fava Larga was the later species. C. juncea was the earliest among all studied species regarding pod maturation. The tested legume species obtained promising initial results during the assessment period, being able to meet the expectations of biomass production and contributing to soil sustainability in this region. AVALIAÇÃO DE LEGUMINOSAS COM POTENCIAL DE USO PARA ADUBOS VERDES NOS TABULEIROS LITORÂNEOS DO PIAUÍ RESUMO -A adubação verde pode atribuir melhorias físicas, químicas e biológicas aos solos, porém, são escassas as informações de uso no nordeste brasileiro. Objetivou-se avaliar o ciclo fenológico e as produtividades de fitomassa de sete espécies de leguminosas. O trabalho foi conduzido durante o segundo semestre de 2015, em delineamento inteiramente casualizado, com sete tratamentos e quatro repetições. Aos 100 dias após semeadura (DAS) foram determinadas a altura das plantas, massa fresca (MFPA) e seca da parte aérea (MSPA), massa fresca (MFR) e seca da raiz (MSR). Avaliou-se também a germinação, florescimento e maturação das vagens. As maiores médias para MFPA foram obtidas pelas Crotalaria breviflora e Crotalaria mucronata, seguidas da Canavalia ensiformis. As mais produtivas para MSPA foram C. breviflora, C. ensiformis, C. mucronata, e Cajanus cajan Cv IAPAR 43. As maiores médias para MFR foram obtidas pelas C. breviflora e C. mucronata, seguidas do C. cajan Cv Fava Larga. A C. mucronata, C. cajan Cv IAPAR 43 e C. breviflora foram as espécies que obtiveram as maiores médias de MSR. ResumoO efeito da época de diferimento sobre a produção e composição química da forragem do capim-Marandu (Brachiaria brizantha cv. Marandu), durante a estação seca, foi avaliado em experimento conduzido em Parnaíba, Piauí. O delineamento experimental foi o de blocos casualizados em um esquema fatorial de 3 x 4, sendo três épocas de diferimento (23 de março, 19 de abril e 17 de maio) e quatro épocas de utilização (12 de julho, 9 de agosto, 6 de setembro e 4 de outubro) com três repetições. O diferimento em março ou abril, com utilizações em julho a outubro proporciona maiores produções de matéria seca verde, contudo, o diferimento em maio e utilização em julho apresentou melhor relação material vivo/material morto. A forragem com maiores relação folha/colmo, teores de proteína bruta e cinzas, e menores teores de fibra em detergente neutro e fibra em detergente ácido, foram verificadas com utilizações nos meses de julho e agosto, independentemente da época de diferimento. Visando conciliar rendimento e composição química da forragem do capim-Marandu, recomenda-se o diferimento em abril ou maio para utilização em julho ou agosto, respectivamente. The present study analyzed the effects of different concentrations of the hexane extract of A. oleraceae (HEAO) (Jambú) on the germ cells of semi-engorged Rhipicephalus microplus female ticks, through a morpho-histological study, evaluating the effectiveness of the extract in the genesis of the individuals. To perform this analysis, 100 semi-engorged females were divided into five groups with 20 individuals each: groups I and II, respectively constituted by distilled water control and 50% ethanol + 1% DMSO, and groups III, IV, and V constituted by treatment with HEAO in the concentrations of 12.5, 25.0 and 50.0 mg/mL, respectively. All the ticks were immersed in the different concentrations of the extract or in distilled water for 5 minutes, dried and conditioned in BOD incubator for 7 days. The individuals of the treatment groups revealed the action of this extract showing alterations in the germ cells of the females from the different groups when compared with those from the groups I and II (control groups). These alterations were mainly related to the size and shape of the oocytes; number of yolk granules; presence, number, size and location of vacuoles in the cytoplasm of all the germ cells; and the presence of nuclear alterations in these cells as well. Thus, it was demonstrated that the concentrations of HEAO affected the germ cells of R. microplus ticks. ABSTRACT:The present work describes the antibacterial activity of the essential oil and the ethanol extract from leaves of Alpinia zerumbet (colônia) on Staphylococcus aureus strains isolated from cows with subclinical mastitis and standard strains ATCC 29213 and ATCC 25923, using the agar diffusion method. Ten treatments containing different concentrations of essential oil and ethanol extract (100.0; 50.0; 25.0; 12.5 and 6.3 mg.mL ) and the control group (50% ethyl alcohol and 1% Tween solutions) were used for antimicrobial testing. The major constituents of the essential oil were p-cimeno (32.72%), 1.8-cineol (24.05%) and 4-terpineol (20.23%), which were determined by gas chromatographymass spectrometry and gas chromatography -flame ionization detector (CG-MS/FID). Ellagic acid and three flavonoids (rutin, quercetin and campferol) were detected in the ethanol extract by means of high performance liquid chromatography-photodiode array detector (HPLC-PDA). All strains showed sensitivity to the treatments with essential oil and the ethanol extract. The best response was obtained with A. zerumbet essential oil at a 100 mg.mL ) e o grupo controle (álcool etílico a 50% e Tween a 1%). Os constituintes majoritários do óleo essencial foram p-cimeno (32,72%), 1,8-cineol (24,05%) e 4-terpineol (20,23%), sendo esses determinados por cromatografia a gás acoplada a espectrometria de massas e cromatografia a gás com detector de ionização de chama (CG-EM/DIC). No extrato etanólico foi detectado o ácido elágico e três flavonoides: rutina, quercetina e campferol, por meio de cromatografia a líquido de alta eficiência acoplada a detector de arranjo de diodo (CLAE-DAD). Todas as cepas apresentaram sensibilidade aos tratamentos com óleo essencial e extrato etanólico. A melhor resposta foi obtida com o óleo essencial de A. zerumbet que, na concentração de 100 mg.mL -1 proporcionou inibição total do crescimento bacteriano. Esses resultados sugerem o potencial antibacteriano do óleo essencial e do extrato etanólico de A. zerumbet no controle da mastite bovina. The aim of this study was to evaluate the acaricidal activity of essential oils from three species of plants with intermediary concentrations of 1,8-cineole against the tick species Rhipicephalus (Boophilus) microplus. For this purpose, five serial concentrations (100.0, 50.0, 25.0, 12.5, 6.2 mg/mL) of essential oils from Mesosphaerum suaveolens (L.) Kuntze, Ocimum gratissimum L. and Alpinia zerumbet (Pers.) B. L. Burtt & R. M. Sm. were used on larval packet and adult immersion tests. The essential oils were analysed by gas chromatography-mass spectrometry (GC/MS) and gas chromatography-flame ionization detection (GC-FID), being detected 35.8, 24.7 and 24.0% of 1.8-cineol in the oils of M. suaveolens, O. gratissimum and A. zerumbet, respectively. The lethal concentration (LC 50) of each oil for larvae and engorged females was calculated through Probit analysis. All essential oils showed high efficacy (≥ 95.0%) on engorged females at the 100.0 mg/mL concentration. In regards to larvae, O. gratissimum (LC 50 = 11.9 mg/mL) was the most potent, followed by the A. zerumbet (LC50 = 19.7 mg/mL) and the M. suaveolens (LC50 = 51.6 mg/mL) essential oils. These results show that other compounds interfere with 1,8-cineole action.
The use of synthetic acaricides is currently the main method to control ticks. However, the indiscriminate use of these chemicals can lead to the selection of resistant individuals and in the accumulation of chemical residues in the environment, contaminating the soil and water streams, consequently affecting the flora, fauna, and the human beings as well. In this sense, the objective of this study was to investigate the acaricidal effect of crude ethanolic extract of Acmella oleracea (L.) R.K. Jansen aerials parts at different concentrations on fed males and semi-engorged females of A. cajennense s.s. An in vitro bioassay (Adult Immersion Test) was carried out to determine the lethal concentration 50 (LC) of ethanolic extract, calculated by Probit analysis. The results showed that the fed males were sensitive to all the concentrations of A. oleracea ethanolic extract, and mortality rate progressively increased (15-65%) in higher ethanolic extract concentrations. However, semi-engorged females were not sensitive to all the concentrations used here. In the highest concentration (100mg/mL), a mortality rate of 100% was observed after 72h of exposure, indicating that the acaricidal effect would probably be dose-dependent. The LC values obtained for the fed A. cajennense s.s males and semi-engorged females were 29.4534mg/mL (limits: 24.4467-41.3847mg/mL) and LC=17.6335mg/mL (limits: 5.2506-23.5335mg/mL), respectively.
