Las fosfolipasas A2 (PLA2) son enzimas que pertenecen a una gran familia de proteínas, actualmente clasificadas en 12 grupos; que comparten similar función enzimática y estructural. De acuerdo con las características bioquímicas y el origen celular, las fosfolipasas se clasifican como: cÍtosólica (cPLA2 ), secretora (sPLA2) e intracelular (iPLA2). La relación estructura-actividad, de este grupo de proteínas es un reto para bioquímicos, biólogos moleculares,toxicólogos, farmacólogos y fisiólogos. Las PLA2 se han identificado en tejidos de mamíferos, en diversos venenos principalmente de serpientes y en algunas bacterias y plantas. Numerosas funciones fisiológicas y fisiopatológicas sin toxicidad se le han atribuido a las PLA2 de mamíferos. En contraste, las PLA2 de venenos son tóxicas e inducen efectos farmacológicos que son mediados probablemente por receptores de membrana, las diversas isoenzimas de las PLA2, de venenos están involucradas en un proceso evolutivo acelerado debido al rápido cambio en los exones, más no, en los intrones ni en las regiones reguladoras de los genes, estas modificaciones son completamente opuestas a las que ocurre en los genes de isoenzimas ordinarias. En esta revisión se describe la enzimología, la regulación celular, los últimos criterios de clasificación, los receptores de membrana, la diversidad biológica de las fosfolipasas de secreción en venenos, mamíferos y bacterias, además se presenta las nuevas estrategias terapéuticas para el tratamiento de enfermedades en las que se encuentran involucradas.
Objetivo. Aislar y caracterizar in silico un transcrito del gen de fosfolipasa A 2 (PLA 2 ) aislado del veneno de Lachesis muta de la Amazonía peruana. Materiales y métodos. Se amplificó el transcrito del gen sPLA 2 mediante la técnica de RT-PCR a partir de RNA total utilizando cebadores específicos, el producto de DNA amplificado se insertó en el vector pGEM para su posterior secuenciación. Mediante análisis bioinformático de la secuencia nucleotídica se determinó un marco de lectura abierta de 414 nucleótidos que codifica 138 aminoácidos, incluyendo16 aminoácidos del péptido señal, el peso molecular y el pI fueron de 13 976 kDa y 5,66 respectivamente. Resultados. La secuencia aminoacídica denominada Lm-PLA 2 -Perú, contiene Asp49, así como Tyr-28, Gly-30, Gly-32, His-48, Tyr52, Asp99 importantes para la actividad enzimática. La comparación de Lm-PLA 2 -Perú con las secuencias aminoacídicas de los bancos de datos mostró 93% de similitud con las sPLA 2 de Lachesis stenophrys y más del 80% con otras sPLA 2 de venenos de la familia Viperidae. El análisis filogenético de la secuencia nucleotídica del transcrito del gen sPLA 2 indica que Lm-PLA 2 -Perú se agrupa con otras sPLA 2 [Asp 49 ] ácidas previamente aisladas del veneno de Bothriechis schlegelii con un 89% de identidad. El modelaje tridimensional de Lm-PLA 2 -Perú, presenta una estructura característica de sPLA 2 del Grupo II formada por tres hélices-α, una lámina-β, una hélice corta y un lazo de unión con calcio. Conclusión. La secuencia nucleotídica corresponde al primer transcripto del gen de PLA 2 clonado a partir del veneno de la serpiente Lachesis muta, que habita en la selva del Perú. The comparison with the amino acid sequence data banks showed of similarity between PLA 2 from Lachesis stenophrys (93%) and other PLA 2 snake venoms and over 80% of other sPLA 2 family Viperidae venoms. Con el objetivo de estandarizar un método rápido para la detección de C. jejuni en pollos se tomaron hisopados cloacales de 50 pollos de 7 semanas de edad en diferentes mercados del distrito de Lima. Se extrajo el ADN por solventes orgánicos y se detectó la presencia de C. jejuni mediante la reacción en cadena de la polimerasa anidada utilizando cebadores específicos para los genes ribosómicos 16S y gen hipO. Paralelamente, utilizando métodos de filtración bacteriológicos convencionales se aisló C. jejuni en medios selectivos. Los métodos rápido y convencional permitieron detectar C. jejuni en el 48/50 (96%) Y 29/50 (58%) de las muestras analizadas, respectivamente. Los hallazgos de este estudio indican que el método rápido tiene mayor sensibilidad que el convencional.
