Lipid supplements can be used to alter fatty acid (FA) profiles of dairy products. For Chanco cheese, however, little information is available concerning effects of lipid supplements on sensorial properties. The objective of this study was to examine effects of supplementation of dairy cow diets with soybean (SO) and hydrogenated vegetable (HVO) oils on chemical and FA composition of milk and cheese and sensory characteristics of cheese. Nine multiparous Holstein cows averaging 169±24d in milk at the beginning of the study were used in a replicated (n=3) 3×3 Latin square design that included 3 periods of 21d. All cows received a basal diet formulated with a 56:44 forage:concentrate ratio. Dietary treatments consisted of the basal diet (control; no fat supplement), and the basal diet supplemented with SO (unrefined oil; 500g/d per cow) and HVO (manufactured from palm oil; 500g/d per cow). Milk fat yield was lower with HVO compared with control and SO. Cheese chemical composition and sensory profile were not affected by dietary treatment. Vaccenic (C18:1 trans-11) and oleic (C18:1 cis-9) acids were higher for SO than for control and HVO. Compared with control and HVO, SO decreased saturated FA and increased monounsaturated FA. The thrombogenic index of milk and cheese produced when cows were fed SO was lower than when cows were fed on control and HVO. The outcome of this study showed that, compared with control and HVO, supplementing dairy cow diets with SO improves milk and cheese FA profile without detrimental effects on the chemical composition of milk and cheese and the sensory characteristics of cheese.
The vertebral column, or spine, provides mechanical support and determines body axis posture and motion. The most common malformation altering spine morphology and function is adolescent idiopathic scoliosis (AIS), a three-dimensional spinal deformity that affects approximately 4% of the population worldwide. Due to AIS genetic heterogenicity and the lack of suitable animal models for its study, the etiology of this condition remains unclear, thus limiting treatment options. We here review current advances in zebrafish phenogenetics concerning AIS-like models and highlight the recently discovered biological processes leading to spine malformations. First, we focus on gene functions and phenotypes controlling critical aspects of postembryonic aspects that prime in spine architecture development and straightening. Second, we summarize how primary cilia assembly and biomechanical stimulus transduction, cerebrospinal fluid components and flow driven by motile cilia have been implicated in the pathogenesis of AIS-like phenotypes. Third, we highlight the inflammatory responses associated with scoliosis. We finally discuss recent innovations and methodologies for morphometrically characterize and analyze the zebrafish spine. Ongoing phenotyping projects are expected to identify novel and unprecedented postembryonic gene functions controlling spine morphology and mutant models of AIS. Importantly, imaging and gene editing technologies are allowing deep phenotyping studies in the zebrafish, opening new experimental paradigms in the morphometric and three-dimensional assessment of spinal malformations. In the future, fully elucidating the phenogenetic underpinnings of AIS etiology in zebrafish and humans will undoubtedly lead to innovative pharmacological treatments against spinal deformities.
Plant-based protein sources, such as soybean, are widely used in fish nutrition due to their market availability, wide distribution and acceptable nutritional quality. However, in some fish species, soybean meal-based diets cause gut inflammation, decreasing both nutrient absorption and growth rates. A suitable alternative to avoid these problems could be the application of additives with anti-inflammatory activity to the diet. In this study, an Aloe vera (Aloe barbadensis Miller, AV) extract was analyzed as a dietary additive to reduce the gut inflammation in Atlantic salmon (Salmo salar) fed with soybean meal (SBM) diet. Fish were distributed in four duplicated groups and fed 28 days with fish meal control diet (FM), AV inclusion diet (AV), FM diet supplemented with AV (FM+AV), SBM diet to induce enteritis and SBM+AV. The fish gut response to these treatments was analyzed in distal intestine by histopathological scores, tissue morphometric measurements and immune gene expression parameters. The score results in fish fed with SBM-based diet clearly showed enteritis, meanwhile fish fed with AV supplemented diet significantly reduced the intestinal SBM signs of damage. These findings were associated to reduction of goblet cells number, lamina propria thickness and sub-epithelial mucosa size, with a significant decrease on pro-inflammatory cytokine il-1β to basal levels, similar to those present in fish fed FM diets. In conclusion, the administration of AV in salmon diet showed a protective intestinal activity against the detrimental effects of SBM, opening the possibility to improve its use as a feed additive in aquafeeds.
