Der hydrolytische Abbau von polymeren Kohlenhydraten stellt ein mehrgleisiges Geschehen dar. Neben der unter Anlagerung von Wasser sich vollziehenden Losung von Sauerstoffbriicken und der daraus resultierenden Bildung von niedriger molekularen Sacchariden ist in Abhangigkeit von den Milieubedingungen auch mit sekundaren Vorgangen desmolytischer bzw. resynthetischer Art zu rechnen. Dabei kann es sich -vom molekiilzerstorenden Abbau abgesehen -einmal um die im sauren Milieu ablaufenden Prozesse der ,,Reversion" handelnl. Sie setzen eine gewisse Mindestkonzentration der Reaktionspartner voraus, sind also eindeutig konzentrationsabhangig und fuhren zu relativ stabilen, von den Ausgangssacchariden in der Struktur abweichenden Produkten ; am Aufbau konnen dabei einheitliche (einfache Reversion) oder uneinheitliche Konstituenten (gemischte Reversion) beteiligt sein. Zum andern ist an jene Prozesse zu denken, die in der Enzymchemie als ,,Gr u p p enii b e r t r a g ung" bzw. als ,,Tr a n sg 1 y cosi d i erung'I2 bezeichnet werden. Hierbei entstehen labile Zwischenprodukte, die mit voranschreitender Umsetzung wieder gespalten werden. Eine solche ,,enz y m at i s c h e" Transglycosidierung ist weitgehend konzentrationsunabhangig, die in eigenen Versuchen als Analogon erwiesene ,,s a u r el' Transglycosidierung aber wohl von der Konzentration abhangig'. Das Schrifttum verzeichnet ein iiberaus reichhaltiges experimentelles Material iiber die Vorgange der Reversion und der Transglycosidierung. Die sich daraus ergebenden, vorangehend skizzierten Moglichkeiten solcher sekundarer Umsetzungen bei der partiellen wie auch der totalen Hydrolyse mussen -wenn man sich nicht der Gefahr von Fehlschlussen aussetzen willkritisch in Erwagung gezogen werden. Darauf ist insbesondere bei reaktionskinetischen Betrachtungen iiber die Hydrolyse von Sacchariden Bedacht zu * Auszugsweise wurde von dem einen von uns (T.) uber dieses Thema in einem Vortrag auf dem VIII. GlucoamylaseAktivitats-Bestimmung Es wird ein Verfahren zur Bestimmung der Aktivititt von Glucoamylase mitgeteilt, wobei als Einheit die Freisetzung von I yMol Glucose je min aus ZuLKowsKI-Starke zugrunde gelegt wird. Zur Bestimmung der gebildeten Glucose wird Glucoseoxydase/Peroxydase herangezogen. Als Kriterium fiir die mijgliche Anwesenheit von &-Amylase werden zusatzlich die reduzierenden Gruppen unter Einsatz von 3,5-Dinitrosalicylsaure ermittelt. Eine Erhohung der mittels der letztgenannten Technik gefundenen Aktivitatswerte weist auf die gleichzeitige Anwesenheit von a-Amylase hin. Modellgemische, bestehend aus Glucoamylase und a-Amylase mikrobieller Hcrkunft, werden gemit0 der beiden genannten Verfahren analysiert. Es zeigt sich, daL? Glucoamylase auch in Gegenwart gewiser Anteile an a-Amylase mit befriedigender Genauigkeit selektiv erfaabar ist. Die Methode wird auf Handelsenzymprsparate angewendet und hinsichtlich Reproduzierbarkeit diskutiert. Die Verwendung von Glucoamylase (a-1,4-Glucan-Glucohydrolase, EC 3.2.1.3) mikrobieller Herkunft zur enzymatischen Starkekonversion sowie zum AufschluB anderer kohlenhydrathaltiger Hohstoffe gewinnt gegenwartig zunehmend an Bedeutung [I]. Die Bestimmung ihrer Aktivitat in Handels-Enzympraparaten setzt eine gesicherte Analytik voraus, wobei insbesondere die meist gleichzeitige Anwesenheit von a-Amylase (&-I ,4-Glucan-q-Glucanohydrolase, EC 3.2.1.1) und Maltase (a-D-Glucosid-Glucohydrolase, EC 3.2.1.20) zu beriicksichtigen ist. Wie das Schrifttum hierzu ausweist, wird Glucoamylase z. Zt. nach mehreren, voneinander abweichenden Methoden ermittelt. Hieraus resulticrt, daBsehr zum Nachteil einer vergleichbaren Aussage iiber die Wirksamkeit vom Enzympraparatenauch die Einheit der Aktivitat unterschiedlich definiert wird. Das Hauptproblem der spezifischen Erfassung von Glucoamylase in Gegenwart anderer carbohydratischer AktivitLten (z. B. &-Amylase, Maltase) besteht in der selektiven Ermittlung der gebildeten Glucose, und zwar zu einem Zeitpunkt, da vonseiten der a-Amylase (in Verbindung mit Maltase) praktisch noch keine Glucose in Freiheit gesetzt wurde; hierzu sei em-ahnt, daL? mit einer partiellen Aufspaltung der durch a-Amylase-Wirkung gebildeten Ma!tose stets zu rechnen ist, da Glucoamylase zugleich auch MaltaseaktivitiLt aufweist. Die Bestimmung der ,,saccharogenen" Aktivitat mittels reduktometrischer Methoden [z -41 besitzt lediglich orientierenden Charakter, es sei denn, es liegen hochgereinigte Praparatefrei von a-Amylasezur Untersuchung vor. Eine genauere Aussage vermittelt dic von PAZUR 11. a. [5] beschriebene papierchromatographische Auftrennung der inkubierten LGsung; jedoch verhindert auch hierbei die Anwesenheit von a-Amylase (+ Maltase) eine exakte Bestimmung dcr Glucoamylase, sofern die Beteiligung der beiden letztgenannten Enzyme bei der Glucosebildung nicht mit Sicherheit auszuschliel3en ist.
