Abstract:Seed treatment refers to the application of certain agents physical, chemical or biological to the seed prior to sowing in order to suppress, control or repel pathogens, insects and other pests that attack seeds, seedlings or plants and it ranges from a basic dressing to coating and pelleting. Introduction and ban of arsenic (used from 1740 until 1808) is the key milestones in the history of modern seed treatment till then a continuous research and advancement in this technology is going on. The technological advancement prepared a roadmap for refining existing seed treatment technologies and future work on technologies like fluid drilling as a way to sow germinated seeds where gel can also serve as a delivery system for other materials, seed priming advances the early phase of germination without redicle emergence. Another advanced technology, solid matrix priming (SMP) has been evaluated as a means to advances the germination of seeds and serve as a carrier for useful material too. Physical and biological seed treatments alone an alternative to chemicals or in combination with a chemical treatment are being used worldwide because of their environmental safety and socioeconomic aspects. Biological seed treatments are expected to be one of the fastest growing seed treatment sectors in the near future, in part because they are easier to register at Environment Protection Agency (EPA). Lack of awareness to seed treatments at farmer's level is one of the limiting factors in disease management and hence, efforts should be made at farmer's level to adopt the technology. Keeping the all above facts in mind, selected seed treatment technologies with their improvement and significance will be discussed in this review.

During physiologic stresses, like micronutrient starvation, infection, and cancer, the cytosolic moonlighting protein glyceraldehyde‐3‐phosphate dehydrogenase (GAPDH) is trafficked to the plasma membrane (PM) and extracellular milieu (ECM). Our work demonstrates that GAPDH mobilized to the PM, and the ECM does not utilize the classic endoplasmic reticulum–Golgi route of secretion; instead, it is first selectively translocated into early and late endosomes from the cytosol via microautophagy. GAPDH recruited to this common entry point is subsequently delivered into multivesicular bodies, leading to its membrane trafficking through secretion via exosomes and secretory lysosomes. We present evidence that both pathways of GAPDH membrane trafficking are up‐regulated upon iron starvation, potentially by mobilization of intracellular calcium. These pathways also play a role in clearance of misfolded intracellular polypeptide aggregates. Our findings suggest that cells build in redundancy for vital cellular pathways to maintain micronutrient homeostasis and prevent buildup of toxic intracellular misfolded protein refuse.—Chauhan, A. Abstract:The genus Trichoderma contains species that are of a great economic importance due to their ability to act as biological control agents against a large variety of fungal plant pathogens. In the present investigation thirty isolates of the Trichoderma sp. were obtained from the rhizosphere soils of different plants at different locations at Nainital, Almora, Udham Singh Nagar, Derhadun, Haridwar and Tehri Garhwal districts of Uttarakhand (India). The isolates were characterized on the basis of their cultural and morphological characteristics. The cultural characteristics included linear growth, colony colour, pigmentation and growth pattern. Morphological characteristics studied were structure, shape and arrangement of conidiophores, phialides and conidia. Out of thirty isolates, 6 isolates namely PB10, PB13, PB23, PB26, PB27 and PB28 were identified as T. virens and remaining 24 isolates as T. harzianum.

Heat shock protein-70kDa (Hsp70) is a member of molecular chaperone family, involved in the proper folding of various proteins. Hsp70 is important for tumor cell survival and is also reported to be involved in enhancing the drug resistance of various cancer types. Hsp70 controls apoptosis both upstream and downstream of the mitochondria by regulating the mitochondrial membrane permeabilization (MMP) and apoptosome formation respectively. In the present study, we have elucidated the role of Hsp70 in Gambogic acid (GA) induced apoptosis in bladder cancer cells. We observed that functional inhibition of Hsp70 by Pifithrin-μ switches GA induced caspase dependent (apoptotic) cell death to caspase independent cell death. However, this cell death was not essentially necrotic in nature, as shown by the observations like intact plasma membranes, cytochrome-c release and no significant effect on nuclear condensation/fragmentation. Inhibition of Hsp70 by Pifithrin-μ shows differential effect on MMP. GA induced MMP and cytochrome-c release was inhibited by Pifithrin-μ at 12h but enhanced at 24h. Pifithrin-μ also reverted back GA inhibited autophagy which resulted in the degradation of accumulated ubiquitinated proteins. Our results demonstrate that Hsp70 plays an important role in GA induced apoptosis by regulating caspase activation. Therefore, inhibition of Hsp70 may hamper with the caspase dependent apoptotic pathways induced by most anti-cancer drugs and reduce their efficacy. However, the combination therapy with Pifithrin-μ may be particularly useful in targeting apoptotic resistant cancer cells as Pifithrin-μ may initiate alternative cell death program in these resistant cells.
