In dieser Zeitschrift hat F. Suter 1 ) die Frage behandelt, ob Brenztraubensäure ein Spaltungsprodukt von Hornsubstanz sei, und sie verneinend beantwortet. Frische Tyrosinmutterlauge wurde mit Schwefelsäure stark angesäuert und mit einer Mischung von Äther und Alkohol extrahiert. Der Rückstand der ätherischen Lösung wurde mit Wasser aufgenommen und mit Quecksilberchlorid gefällt; das Filtrat wurde mit Schwefelwasserstoif behandelt; eine Bildung von Thiomilchsäure konnte weder durch Prüfung mit Quecksilberchlorid, noch durch die Kupfersulfatreaktion nachgewiesen werden, woraus geschlossen wird, daß keine Brenztraubensäure aus der Hornsubstanz gebildet würde. Es wird nicht angegeben, warum nicht, statt dieses komplizierten Verfahrens, die empfindliche Reaktion auf Brenztraubensäure mit salzsaurem Phenylhydrazin in Verwendung kam. Daß die Bedeutung eines mit dieser Reaktion erlangten positiven Resultats nicht durch den negativen Befund von Suter entkräftet wird, liegt auf der Hand.In einer Abhandlung, 2 ) worin ich zuerst die Bildung des Cystins aus Hornsubstanz mitteilte, wurde angegeben, daß nach dem Zersetzen der Hornsubstanz mit Salzsäure eine schwerflüchtige, jodoformgebende Substanz in das Destillat überging, als die Zersetzungsflüssigkeit bei niedrigem Drucke abdestilliert wurde; der Schwerflüchtigkeit wegen konnte diese Substanz
Die Frage über die Bindungsart des Schwefels in den Proteinstoffen ist zwar mehrmals Gegenstand der Bearbeitung ge\vesen, aber mit nur wenig entscheidenden Resultaten. Aus Untersuchungen, w r elche in Liebig's Laboratorium ausgeführt wurden, weiss man, dass die Behandlung mit Lauge nur einen Theil des Schwefels als Schwefelalkali abspaltet, welches Bleioxyd schwärzt, und dass ein grosser Theil des Schwefels im Moleküle zurückbleibt. Man hat seitdem zwei verschiedene Bindimgsformen des Schwefels angenommen und von oxydirtem und nicht oxydirtem resp. von fest gebundenem und locker gebundenem oder bleischwärzendem Schwefel gesprochen, welcher in verschiedenen Proteinkörpern in verschiedener Menge vorkommt.Die Art der Verkettung dieses locker gebundenen, bleischwärzenden Schwefels ist durch die bisherigen Untersuchungen keineswegs aufgeklärt worden. Die Ansichten über die Bedeutung desselben gehen auch ziemlich weit auseinander.Die Proteintheorie von Mulder betrachtete seiner Zeit den locker gebundenen Schwefel zwar nicht als einen Bestandtheil des Kernes der Proteinkörper, nahm aber an, dass er in einer Seitenkette gebunden sei, welche für die verschiedenen Eigenschaften der einzelnen Proteinkörper Bedeutung habe. Einmal hat Suter 1 ) bei der Bearbeitung von Hornsubstanz aus Tyrosinmutterlauge, die Fäulnisgeruch zeigte und an deren Oberfläche Schimmelpilze reichlich wucherten, a-Thiomilchsäure darstellen können. Da dies aber nicht bei der Bearbeitung von frischer Tyrosinmutterlauge gelang, schloß er, daß die Thiomilchsäure sekundär durch die Pilze gebildet worden war. Seitdem hat Friedmann 2 ) die Frage zu neuer Bearbeitung aufgenommen. Er konnte aus Rinderhorn, Gänsefedern, Menschenhaaren und Wolle durch Kochen mit Salzsäure a-Thiomilchsäure (oder deren Disulfid) darstellen; daneben glaubt er Thioglykolsäure gefunden zu haben. Seine Schlußfolgerung, daß die -Thiomilchsäure primär abgespalten wird, setzt aber die Richtigkeit der Angaben Suters voraus, daß keine Bindung von Schwefel bei der Behandlung der Hornzersetzungsflüssigkeiten mit Schwefelwasserstoff stattfinde und weiter, daß keine Bildung von Brenztraubensäure bei der Zersetzung der Proteuistoffe vorkomme. Die erste dieser Angaben ist aber wenig überzeugend, da Suter dieselbe durch keinen Beleg stützt 3 ) und die unumgänglichen Fehler bei der Ausführung der Analysen
Bekanntlich ist E. Baumann durch eine Reihe von Arbeiten, welche von ihm und seinen Schülern ausgeführt wurden, zu der Ansicht gelangt, daß das Cystin (aus Harn) ein Disulfid der -Aminothiomilchsäure sei. E. Friedmann 1 ) findet bei der Besprechung dieser Arbeiten, daß B au mann für die -Stellung des Schwefels keinen bindenden Beweis geliefert hat. Selber konnte Friedmann bei Bearbeitung des Cystins (aus Hornspänen) nach Joche m ß-Thiomilchsäure darstellen und in der Form des Disulfids rein gewinnen; die Prüfung der rohen Thiomilchsäure durch die Farbenreaktion mit Kupfersulfat zeigte keine -Thiomilchsäure an. Durch Oxydation von Cystin (oder Cystein) mit Brom konnte er eine Sulfosäure des Cysteins, Cysteinsäure genannt, darstellen. Durch Erhitzen dieser Säure mit Wasser auf 235° gelang, unter Abspaltung von Kohlensäure, die physiologisch sehr bedeutungsvolle Überführung derselben in Taurin. Die Bindung des Schwefels und der Aminogruppe an verschiedene KohlenstofTatome wurde dadurch erwiesen.Zu derselben Auffassung bezüglich der Bindung des Schwefels und der Aminogruppe gelangte C.
