These results suggest that a direct DNA detection protocol is more sensitive, accurate and faster than traditional culture-based methods. It can be useful to detect T. rubrum in patients undergoing antifungal therapy and who have been reported mycologically cured on the basis of a culture-based method.

A total of 63 dermatophyte strains belonging to 6 species (Trichophyton rubrum (16), T. mentagrophytes (16), T. tonsurans (20), T. violaceum (2), Microsporum canis (7), and Epidermophyton floccosum (2)) were tested for their in vitro susceptibility to a range of antifungal drugs using disk diffusion and Clinical Laboratory Standards Institute M38-A (CLSI, formerly NCCLS) broth microdilution methods. The antifungals used were ciclopirox (CIC), terbinafine (TER), griseofulvin (GRE), fluconazole (FLC), itraconazole (ITR), posaconazole (POS), and ravuconazole (RAV). In the broth microdilution assay terbinafine was found to have the highest activity followed by ravuconazole, posaconazole, ciclopirox, itraconazole, griseofulvin and fluconazole. In the disk diffusion assay terbinafine produced the largest inhibition zone diameters (IZDs) on Dermasel agar media followed by ravuconazole, griseofulvin, posaconazole, and itraconazole. A significant correlation was not observed between the minimum inhibitory concentrations (MICs) and IZDs, but some correlations were observed for POS, RAV, and TER (correlation coefficients r=-0.507, -0.249, -0.267, P<0.05, respectively). MICs obtained by the microdilution method with ITR and GRE did not correlate with IZDs obtained in disk diffusion assays in this study. The ciclopirox (20 microg/disk) and fluconazole (25 microg/disk) did not produce well defined inhibition zone diameters on Dermasel agar medium.

The in vitro susceptibility of 15 strains of six genera of zygomycetes including Rhizopus oryzae (Rhizopus arrhizus), Rhizopus stolonifer, Mucor circinelloides (three), Absidia corymbifera (three), Rhizomucor pusillus (three), Cunninghamella bertholletiae (two), and Syncephalastrum racemosum (two) to nine antifungal agents were determined by the NCCLS M38-A broth microdilution method. Geometric means of the minimal inhibitory concentrations (MIC) were: amphotericin B 0.07 mg l(-1) (range 0.03-0.5 mg l(-1)), nystatin 0.83 mg l(-1) (range 0.25-4 mg l(-1)), itraconazole 0.59 mg l(-1) (range 0.03 to >8 mg l(-1)), voriconazole 6.50 mg l(-1) (range 2 to >8 mg l(-1)), ciclopiroxolamine 1.59 mg l(-1) (range 0.5-4 mg l(-1)), and amorolfine 9.19 mg l(-1) (range 1 to >16 mg l(-1)). All strains were resistant to 5-flucytosine, fluconazole (MIC >64 mg l(-1)) and caspofungin (MIC >16 mg l(-1)).

The process of aging is characterized by numerous changes in the body which has an overall negative effect on the health and lifestyle of elderly. Nutrition deserves special attention as an individual reaches old age. It plays a vital role in affecting the quality of life, including physical, mental and social health. The physiological decline in food intake is very common among older age and this result in nutritional deficiencies. These increased nutritional deficiencies are the major risk factors for certain chronic diseases and deteriorated age related health. Thus, the adoption of nutritional intervention can be a measure to tackle the current situation of nutritional deficiencies and promote a healthy lifestyle.

Neuronal migration in brain is followed by differentiation of committed neurons and simultaneous apoptosis of uncommitted preneuronal cells due to a limiting supply of trophic factors and nutrients. We have dissected differentiation and apoptosis by designing a simple in vitro model for this nutrient deprivation using engineered neuronal cell lines stably transfected with a promoterless segment (G‐21) of the intronless human serotonin1A receptor (5‐HT1A‐R) gene. Despite the use of widely different heterologous promoters (cytomegalovirus and Rous sarcoma virus) for the stable expression of G‐21, a dramatic increase in expression of the 5‐HT1A‐R (five‐ to 15‐fold) and its mRNA was always observed during degeneration and apoptosis of nutrient‐deprived neuronal cells. Involvement in this induction of a 170‐bp 5′‐end untranslated sequence (5′‐UT) (tail end of the 500‐bp natural promoter) of G‐21 was confirmed by stable transfection of neuronal cells with an SV‐40 promoter‐driven construct harboring the 5′‐UT and the reporter chloramphenicol acetyltransferase (CAT) cDNA. Presence of the 5′‐UT resulted in a threefold increase in CAT expression during nutrient deprivation in randomly chosen clones. The induction was also observed in the endogenous 5‐HT1A‐R, expressed by embryonic day 16 mouse hippocampal neurons, subsequent to nutrient deprivation and onset of degeneration. A trophic role of the 5‐HT1A‐R has been suggested in earlier studies. Considering the example of protective heat shock proteins, which are induced during various types of stress, our results suggest that stressed neuronal cells undergoing degeneration and apoptosis synthesize increased levels of 5‐HT1A‐R as a final attempt to survive.
