WSterisch wenig gehinderte prim. Amine, Alkylendiamine, Aminoalkohole und Aminosauren reagieren mit Trimethylsilyl-diathylarnin in Gegenwart von wenig (NH4)2SO4 zu den entsprechenden N.N-Bis-trimethylsilyl-Derivaten, die sich mit Ausnahme des AminosaureDerivates durch bemerkenswerte Hydrolysebestiindigkeit auszeichnen.
Die physikalivchen Daten nnd IIt-Spektren einiger Aininosaure-trirnetliylsilylester werden angegeben.Die in der vorangegangenen Veroffentlichung dieser Reihe erstmalig beschriebenen Aminosaure-trialkylsilylester erwieseri sich alp therinisch recht bestandig und erschienen deshalb zur gas-chroinatographischen Trennung von Sminosauren4) geeignet. I n diesem Zusammenhang tvurden nun die physikalischen Daten aller uns zugiinglichen AminosSure-triniethylsilylester bestimmt und das Verhalten weiterer funktioneller Gruppen in den Aminosiiuremolekiilen bei Silylierixng imd Aminolyse untersucht .Zur Synthese der Aminosiiure-trimethylsilylester wurden die hereits beschriebenen z,
K. RUHLMANN und J. HILS 21 1 20 Stdn. bei 37" inkubiert. AnschlieBende Chromatographie im Losungsmittel A zeigte drei Substanzen rnit den RF-Werten 0.1 (3'-Uridy/siiure), 0.06 und 0.32 neben einer Spur Uridin. Der Fleck mit RF= 0.32 wurde ausgeschnitten, mit Wasser eluiert, und die Losung lyophilisiert. Der Ruckstand wurde 12 Min. rnit 20-proz. Essigsaure bei 100" behandelt. Nach Entfernen der Essigsaure wurde 24 Stdn. rnit 0.3 n KOH bei 37Ohydrolysiert. Nach Neutralisation rnit dem Ionenaustauscher MERCK I wurde im Losungsmittel A chromatographiert. Das Chromatogramm zeigte nur 3'-Uridy/shure und Uridin im Verhaltnis I . 1 : 1. Das von Ribonuclease nicht hydrolysierte Material (RF = 0.06) wurde rnit Wasser eluiert und die Losung lyophilisiert. Der Ruckstand (6 optische Dichteeinheiten OD) wurde in 25 A Wasser gelost, 25 A Trispuffer (0.05rn, pH 8) sowie 10 A einer Losung von a/kalischer Phosphafuse (10 mg Protein/20 ccm) zugegeben, und die Losung 6 Stdn. bei 37" inkubiert. Das Chromatogramm (Losungsmittel A) zeigte keine Hydrolyseprodukte. u b e r die Si-N-Bindung, XIV1) Darstellung, Eigenschaften und Reaktionen von Aminosaure-trialkylsilylestern werden beschrieben.Die Existenz von Aminosaure-trialkylsilylestern (Aminosauresilylestern) wurde schon mehrfach vermutet 2J) und ihre Darstellung wiederholt, jedoch erfolglos, versucht.So gelang es nicht, Cbo-aminosaure-trialkylsilylester hydrierend zu spaltenz), und Solvolyseversuche an N-Trialkylsilyl-aminosaure-trialkylsilylestern (Silylaminosauren) fiihrten auch mit einem UnterschuR an Solvolyseagenz stets nur zu den freien Aminosauren4.5).Es zeigte sich nun, da8 beim Ruhren von Silylaminosauren in einer NH3-Atmosphare bei Raumtemperatur Aminosauresilylester gebildet werden : 2 R3Si-NH-CH-COzSiR3 + NH3 -D R3Si-NH-SiR3 + 2 HzN-CH-C02SiR3
