N‐Acyl‐thioamide 1 wurden durch Acylierung von Thioamiden und zum ersten Mal durch Anlagerung von Grignard‐Reagenz an Benzoyl‐isothiocyanat dargestellt. IR‐ und NMR‐spektroskopische Untersuchungen ergeben, daß bei aliphatischen N‐Acyl‐ und N‐Benzoyl‐thioamiden die E,Z′‐Konfiguration, bei N‐Acyl‐thiobenzamiden mit kleinen Acylresten dagegen die Z,E′‐Konfiguration vorherrscht. N‐Acyl‐thioamide 1 mit α‐ständigen H‐Atomen reagieren mit Acylhalogeniden zu den Keten‐S,N‐acylalen 3 und 4.
Thioamide reagieren mit Diphenylcyclopropenon zu N-(E)-[2-Phenyl-cinnamoyl]-thioamiden 2 (Tab. l), aus denen durch Cyclisierung 1.3-Thiazin-4-one 3 (Tab. 2) bzw. 4 (Tab. 3) hervorgehen.
Aus den bei der Alkylicrung von Tbioamiden priinar cntstchenden Thioimidsaureesterhydrohalogcniden koniien die freien Thioirnidwurccqter dargestellt und thermisch in die Thioamidc umgelagert werden Die Reaktron verlauft intermolekular und erfordert eine hinrcichcnde Stabilitat des Alkylreqtes als Carbeniutn-Ion. In zwei Fallen konnten die entstandenen Thioamide wieder in die Thioirntdsaureester-hydrochloride zuruckveraandelt wcrdcn. Mit verschiedenen Carbinolen reagiercn die Thioainide bevorzugt zu N-Alkylierungsprodukten.Thioamidc odcr dcren Anionen sind ambifunktionelle Nucleophile. Die Alkylicrung crfolgt jcdoch im allgenxineii nur am Schwefel zu Thioimidsiureester-hydrohalogeniden 11).Lcdiglich in Ausnahnicfallen sind die isonieren N-substituierten Thioamide durch direkte Alkylierung des Thioamids zuganglich. So reagiert nach Bredereck, Gompper und Bitzer 2 , Thiobenzamid mit Tritylchlorid in Pyridin zu N-Trltyl-thiobenzamid. Hurd und Mitarbb.3) haben gezeigt, daB die N-Alkylierung thermodynnmisch gesteuert und priinar der Angriff am Sch wcfcl bcvorzu gt ist.Wir suchten nach Alkylierungsinitteln, rnit denen man, jc nach Wahl der Reaktionsbedingungen, eine Alkylierung am Schwefel-oder am Stickstoffatom erreichen kann. AuBcrdem wurde gepriift, ob rich Thioimidsaureester in Thioamide umlagern lassen. Eiae derartige Umlagerung, die bei Carbonamiden als Muu??m-Chapmu/z-Umlagerung4) schon seit langem bekannt ist, gelang bei Thioamiden bisher noch nicht eindeutig. Aus der L.6sang kristallisierten nach Zusatz von Petrolither (60--70") 0.90 g (65 x) Carbinol aus. 3 nus 2 e : 2.0 g 2 e wurden niit 2w HzS04 und Ather his zur vollstindigen Auflosung geschuttelt. Aus den uber Calciumchlorid getrockneten, eingeengten und rnit Petrolather versetzten Atherextrakten fielen in der Kalte 0.90 g 3 aus (SO x), Als Nebenprodukte wurden im Dunnschichtchromatogramm Thioheiizamid und 4.4'-Dimethoxy-be.nzhydrol nachgewiesen. Alkalische Hydrulyse von ?'l~ioimidsuureester-hydr~huiogeni~ei~ 4.4'-Dimethoxy-benzhydrylmrr~uptun nus 2e: 1 S O g 2e wurden in 20proz. Natronlnuge suspendiert. Nach I stdg. Riihren wurde mit Chloroform extrahiert, getrocknet, eingedampft, in wenig Ather aufgenomnien und rnit Pctrolathcr ( 3 0 -5 0 ) versetzt. In der Kalte fielen 0.60 g (60 %) farblose Kristalle aus. Durch Alkylierung von N-Acyl-thioamiden oder durch Acylierung von Thioimidsaureestern wurden die N-Acyl-thioimidsaureester 10 synthetisiert, die bei Vorhandensein a-standiger H-Atome mit den N-Acyl-keten-S,N-acetalen 15 in einem Tautomeriegleichgewicht stehen konnen. Wahrend N-Acyl-thioacetimidsaureester ausschlieBlich in der ThioimidsaureesterForm vorliegen, findet man im Gleichgewicht bei den or-Alkyl-oder or-Phenyl-substituierten Vertretern die beiden Tautomeren in meBbaren Konzentrationen nebeneinander und bei den orp-disubstituierten Derivaten nur noch die Keten-S,N-acetale. Fur die Tautomerisierung ergab sich eine Kinetik 1. Ordnung. Imine-Enamine Tautomerism, I. -Tautomeric Equilibrium between N-Acylimidothiolates and N-Acylketene S,N-AcetalsAlkylation of N-acylthioamides or acylation of imidothiolates afforded the N-acylimidothiolates 10, which may be in tautomeric equilibrium with N-acylketene S,N-acetals 15 if H atoms are present in the or-position. N-Acylthiolacetimidates occur exclusively in the thiolimidate form whereas or-alkyl or or-phenyl substituted compounds show an equilibrium with both tautomers in measurable concentrations. orpDisubstituted compounds exist only in the ketene S,N-acetal form. The tautomerization is a first order reaction.
