Atherosclerosis represents the most significant risk factor for coronary artery disease (CAD), the leading cause of death in developed countries. To better understand the pathogenesis of atherosclerosis, we applied a likeli hoodbased model selection method to infer genedisease causality relationships for the aortic lesion trait in a segregating mouse population demonstrating a spectrum of susceptibility to developing atherosclerotic lesions. We identified 292 genes that tested causal for aortic lesions from liver and adipose tissues of these mice, and we experimentally validated one of these candidate causal genes, complement component 3a receptor 1 (C3ar1), using a knockout mouse model. We also found that genes identified by this method overlapped with genes progressively regulated in the aortic arches of 2 mouse models of atherosclerosis during atherosclerotic lesion development. By comparing our gene set with findings from public human genomewide association studies (GWAS) of CAD and related traits, we found that 5 genes identified by our study overlapped with published studies in humans in which they were identified as risk factors for multiple atherosclerosisrelated pathologies, including myocardial infarction, serum uric acid levels, mean platelet volume, aortic root size, and heart failure. Candidate causal genes were also found to be enriched with CAD risk polymorphisms identified by the Wellcome Trust Case Control Consortium (WTCCC). Our findings therefore validate the ability of cau sality testing procedures to provide insights into the mechanisms underlying atherosclerosis development.

LITMUS was a single-centre, Phase 2a study designed to investigate whether the gene biomarker FGL2/IFNG previously reported for the identification of tolerance in murine models could identify operationally tolerant liver transplant recipients. Multiplex RT-PCR was used to amplify eight immunoregulatory genes in peripheral blood mononuclear cells (PBMC) from 69 adult liver transplant recipients. Patients with PBMC FGL2/IFNG ≥ 1 and a normal liver biopsy underwent immunosuppression (IS) withdrawal. The primary endpoint was the development of operational tolerance. Secondary endpoints included correlation of tolerance with allograft gene expression and immune cell markers. Twenty-eight of 69 patients (38%) were positive for the PBMC tolerance biomarker and 23 proceeded to IS withdrawal. Nine of the 23 patients had abnormal baseline liver biopsies and were excluded. Of the14 patients with normal biopsies, eight (57%) have achieved operational tolerance and are off IS (range 12-57 months). Additional studies revealed that all of the tolerant patients and only one non-tolerant patient had a liver gene ratio of FOXP3/IFNG ≥1 prior to IS withdrawal. Increased CD4 + T regulatory T cells were detected both in PBMC and livers of tolerant patients following IS withdrawal. Higher expression of SELE (gene for E-selectin) and lower expression of genes associated with inflammatory responses (GZMB, CIITA, UBD, LSP1 and CXCL9) were observed in the pre-withdrawal liver biopsies of tolerant patients by RNA sequencing. These results suggest that measurement of PBMC FGL2/IFNG may enrich for the identification of operationally tolerant liver transplant patients, especially when combined with intragraft measurement of FOXP3/IFNG.

An external rotor transverse flux motor using a combined-type stator core is proposed for a direct drive application in this paper. The stator core is combined by two kinds of components that can both be manufactured conveniently by generic laminated silicon steel used in traditional motors. The motor benefits from the predominance of low manufacturing cost and low iron loss by using a silicon-steel sheet. Firstly, the basic structure and operation principles of the proposed motor are introduced. Secondly, the expressions of the electromagnetic torque and the cogging torque are deduced by theoretical analysis. Thirdly, the basic characteristics such as permanent magnet flux linkage, no-load back electromotive force, cogging torque and electromagnetic torque are analyzed by a three-dimensional finite element method (3D FEM). Then, the influence of structure parameters on the torque density is investigated, which provides a useful foundation for optimum design of the novel motor. Finally, the torque density of the proposed motor is calculated and discussed, and the result shows that the proposed motor in this paper can provide considerable torque density by using few permanent magnets.
