The effects of an electric current on ethylene biosynthesis were investigated in cucumber (Cucumis sativus L.) fruit that were producing almost no ethylene. Direct currents at 0.5 to 3.0 milliamperes induced much ethylene synthesis, with a rapid continuous increase in the rate, which reached a peak within 5 to 6 hours and then decreased. The rate of production was greater with a stronger current. Ethylene production was not observed after the use of a sine-wave altemating current (60 hertz) at 3 milliamperes, the magnitude at which a direct current had the greatest effect. The activity of 1-aminocyclopropane-1-carboxylic acid (ACC) synthase and ethylene forming enzyme (EFE) increased before the rise in ethylene production. ACC synthase and EFE were activated sixfold and fourfold, respectively, by 2 hours. The concentration of ACC increased linearly up to 6 hours and then decreased. Ethylene induction by an electric current was suppressed almost completely by the infiltration of the cucumbers with 5 millimolar aminooxyacetic acid, an inhibitor of ACC synthase, and was also suppressed 70% by 5 millimolar salicylic acid, an inhibitor of EFE. The results indicate that the ethylene induced by the direct current was synthesized via the ACCethylene pathway as a result of electrical stress, a new kind of stress to be identified.
Melon (Cucumis melo L.) has a long history of cultivation worldwide. During cultivation, domestication, and selection breeding, the sugar content of mature melon fruits has been significantly increased. Compared with un-sweet melon and wild melon, rapid sucrose accumulation can occur in the middle and late stages of sweet melon fruit development. During the evolution and development of melon fruit, the phloem unloading pathway has not been identified and analyzed. In this study, the phloem unloading pathway and the function of related sugar transporters in cultivated and wild melon fruits were analyzed by CFDA and esculin tracing, cytological pathway observation, qRT-PCR, and gene function analysis, etc. Results show that phloem unloading pathway of wild melon fruit is largely symplastic, whereas phloem unloading pathway of cultivated melon fruit shifts from symplastic to apoplasmic during development. According to fruit grafting experiment, fruit sink accumulates sugars independently. The results of the correlation analysis showed that the expression amounts of several sucrose transporter genes were positively correlated with the sucrose content of melon fruit. Furthermore, CmSWEET10 was proved to be a sucrose transporter located on the plasma membrane of the phloem and highly expressed in the premature stage of sweet melon fruits, which means it may be involved in phloem apoplast unloading and sucrose accumulation in sweet melon fruits. Finally, we summarized a functional model of related enzymes and sugar transporters involved in the apoplast unloading of sweet melon fruits during enlargement and sucrose accumulation.
Purpose Arctium lappa L. is one of the medicinal and food homologous plants in China, which is rich in nutrients and medicinal ingredients. The use of plant growth promoting (PGP) endophytic bacteria is a useful alternative in agricultural production to reduce the use of chemical fertilizers. The aim of this study was to analysis the diversity of endophytic bacteria in different cultivars of A. lappa L. collected from two different geographical locations in China and evaluate PGP traits of the isolates and their potential PGP ability in greenhouse condition. Methods Endophytic bacterial community was investigated by culture-dependent and culture-independent methods. Isolates were screened and investigated for multiple PGP traits, and representative strains were inoculated host seedlings to evaluate the growth promoting effect. Results A total of 348 endophytic bacteria were obtained and they distributed into four phyla, 30 genera and 73 different species. In addition, high throughput sequencing revealed more abundant bacterial community, including 17 bacterial phyla, and 207 different known genera. A high proportion of PGP traits were detected, including production of indole acetic acid, siderophore, ammonia and phosphate solubilization. Four representative strains with multiple PGP traits of the most prevalent genera were further selected for host inoculation and growth promoting evaluation, and they significantly increase seedlings length, root length and fresh weight. Conclusion This study demonstrated that A. lappa L. harbors abundant endophytic bacteria, and plant genotype and geographical origin affect their composition. Moreover, some endophytic bacteria showed good potential for the development of microbial fertilizer in the future.
