Macroautophagy/autophagy is a cellular process in which cytosolic contents are degraded by lysosome in response to various stress conditions. Apart from its role in the maintenance of cellular homeostasis, autophagy also involves in regulation of cell cycle progression under nutrient-deprivation conditions. However, whether and how autophagy is regulated by the cell cycle especially during mitosis remains largely undefined. Here we show that WIPI2/ATG18B (WD repeat domain, phosphoinositide interacting 2), an autophagy-related (ATG) protein that plays a critical role in autophagosome biogenesis, is a direct substrate of CUL4-RING ubiquitin ligases (CRL4s). Upon mitosis induction, CRL4s are activated via neddylation, and recruit WIPI2 via DDB1 (damage specific DNA binding protein 1), leading to polyubiquitination and proteasomal degradation of WIPI2 and suppression of autophagy. The WIPI2 protein level and autophagy during mitosis could be rescued by knockdown of CRL4s or treatment with MLN4924/Pevonedistat, a selective inhibitor of CRLs, via suppression of NAE1 (NEDD8 activating enzyme E1 subunit 1). Moreover, restoration of WIPI2 rescues autophagy during mitosis and leads to mitotic slippage and cell senescence. Recent experimental literature (Sol. Energ. Mat. Sol. Cells, 2012, 105, 192) has reported that nitrogen doped Cu 2 O is a possible material for novel intermediate band solar cells. The doping effects of nitrogen on the crystal structure, electronic structure, and optical properties of Cu 2 O have been studied by an ultrasoft pseudopotential plane wave method based on first-principles calculations. The results show that nitrogen doping slightly widens the band gap of Cu 2 O, and form an intermediate band in the gap located at about 0.9 eV from the VBM (or CBM). This intermediate band is predominantly formed by hybridization between the N-2p states and the Cu-3d states. N-doped Cu 2 O is very likely to absorb at a maximum across the solar light spectrum, from the near infrared region to the ultraviolet region. Based on these results, N-doped Cu 2 O is considered to be a perfect intermediate band material for a novel kind of solar cells.

In human leukaemia, resistance to chemotherapy leads to treatment ineffectiveness or failure. Previous studies have indicated that cancers with increased levels of aerobic glycolysis are insensitive to numerous forms of chemotherapy and respond poorly to radiotherapy. Whether glycolysis serves a key role in drug resistance of leukaemia cells remains unclear. The present study systematically investigated aerobic glycolytic alterations and regulation in K562/adriamycin (ADM) multidrug‑resistant (MDR) and ADM‑sensitive K562 leukaemia cells in normoxia, and the association between drug resistance and improper glycometabolism. The cell proliferating activity was assessed with an MTT colorimetric assay, glycolysis, including glucose consumption, lactate export and key‑enzyme activity was determined by corresponding commercial testing kits. The expression levels of hexokinase‑II (HK‑II), lactate dehydrogenase A (LDHA), glucose transporter‑4 (GLUT‑4), AKT, p‑AKT473/308, mammalian target of rapamycin (mTOR), p‑mTOR, c‑Myc and hypoxia‑inducible factor‑1α (HIF‑1α) were analyzed by western blot or reverse transcription‑quantitative polymerase chain reaction (RT‑qPCR). K562/ADM cells exhibited increased glucose consumption and lactate accumulation, increased lactate dehydrogenase, hexokinase and pyruvate kinase activities, and reduced phosphofructokinase activity. In addition, K562/ADM cells expressed significantly more HK‑II and GLUT‑4. Notably, inhibition of glycolysis effectively killed sensitive and resistant leukaemia cells and potently restored the sensitivity of MDR cells to the anticancer agent ADM. The AKT serine/threonine kinase (AKT)/mechanistic target of rapamycin (mTOR) signalling pathway, a crucial regulator of glycometabolic homeostasis, mediated over‑activation and upregulation of c‑Myc expression levels in K562/ADM cells, which directly stimulated glucose consumption and enhanced glycolysis. In conclusion, the present study demonstrated that MDR leukaemia cells exhibit increased aerobic glycolytic activity and that this may be responsible for resistance to chemotherapeutics in leukaemia MDR cells via activation of the AKT‑mTOR‑c‑Myc signalling pathway. Therefore, inhibition of aerobic glycolysis may be a potential therapeutic strategy to efficiently treat multidrug resistance in relapsed or refractory leukaemia and cancers.

The E3 ubiquitin ligase HUWE1/Mule/ARF-BP1 plays an important role in integrating/coordinating diverse cellular processes such as DNA damage repair and apoptosis. A previous study has shown that HUWE1 is required for the early step of DNA damage-induced apoptosis, by targeting MCL-1 for proteasomal degradation. However, HUWE1 is subsequently inactivated, promoting cell survival and the subsequent DNA damage repair process. The mechanism underlying its regulation during this process remains largely undefined. Here, we show that the Cullin4B-RING E3 ligase (CRL4B) is required for proteasomal degradation of HUWE1 in response to DNA damage. CUL4B is activated in a NEDD8-dependent manner, and ubiquitinates HUWE1 in vitro and in vivo. The depletion of CUL4B stabilizes HUWE1, which in turn accelerates the degradation of MCL-1, leading to increased induction of apoptosis. Accordingly, cells deficient in CUL4B showed increased sensitivity to DNA damage reagents. More importantly, upon CUL4B depletion, these phenotypes can be rescued through simultaneous depletion of HUWE1, consistent with the role of CUL4B in regulating HUWE1. Collectively, these results identify CRL4B as an essential E3 ligase in targeting the proteasomal degradation of HUWE1 in response to DNA damage, and provide a potential strategy for cancer therapy by targeting HUWE1 and the CUL4B E3 ligase.

Network-on-chip (NoC) is promising for the communication paradigm of the next-generation multiprocessor system-on-chip (MPSoC). As communication has become an integral part of on-chip computing, and even the performance bottleneck, researchers are paying much attention to its implementation and optimization. Traditional techniques that model communication inaccurately will lead to unexpected runtime performance, which is on average 90.8% worse than the predicted results based on observation, and are not suitable for the deep optimization of communication-intensive scenarios. In this paper, techniques are presented for the NoC-based MPSoCs that integrate optimization on interprocessor communications with the objective of minimizing the schedule length. A fine-grained integer-linear programming (ILP) model is proposed to properly address the communication latency with a network contention, which generates runtime scheduling with trivial performance difference from the predictions. We further propose a heuristic algorithm, unified priority-based scheduling (UPS), to effectively solve the contention problem in polynomial time by assigning priorities to messages. Evaluation results show that the solutions obtained by the ILP model outperform the state-of-the-art techniques by 31.1%, and UPS improves application performance by 34.7% and 44.4% compared with acquainted first-in-first-out (FIFO)-based and random-based methods. In addition, UPS achieves averagely 8.3% approximated results with the optimal solutions generated by ILP. A case study on H.264 high-definition television (HDTV) decoder and the digital signal processor (DSP) filter benchmarks achieves significant improvement on the performance and the results prediction accuracy, as well as the prominent reduction in the number of network contention and energy consumption.
