ResumenRuta graveolens L. y R. chalepensis L. son plantas usadas en medicina folclórica como antiespasmódicos, antihelmínticos, antimicrobianos, emenagogos y abortivos. En el presente trabajo se evaluó el efecto del extracto acuoso liofilizado (EAL) de las hojas de R. chalepensis en embriones postimplantacionales de ratón. Ratonas albinas preñadas recibieron intraperitonalmente (ip) 10 mg de ruda liofilizada/kg de peso corporal (grupo tratado, n=12) durante el periodo post-implantacional (día 9 -día 17 post-cópula), y un grupo control(C), n=18, que recibió sólo agua destilada durante el mismo período. El EAL de ruda no afectó negativamente el peso de la madre pero sí del útero durante el tratamiento (p<0,05). En el grupo tratado la frecuencia de reabsorciones fetales fue mayor (p<0,05) y el peso fetal fue significativamente menor en comparación con el control (p<0,01). Además en el grupo tratado se evidenció la presencia de malformaciones esqueléticas. En conclusión, encontramos que el EAL de R. chalepensis muestra efectos embriotóxicos en ratones expuestos durante el período postimplantacional.Palabras clave: Ruta chalepensis, embriotoxicidad, teratogenicidad, ratón, postimplantacional. AbstractRuta graveolens L. and R. chalepensis L. are plants used in folkloric medicine as antispasmodics, antihelmintics, antimicrobials, emmenagoges and abortifacients. In this work we evaluated embryotoxic effect of liophilized aqueous extract (LAE) of leaves of R. chalepensis on postimplantational mouse embryos. Pregnant albino mice received intraperitoneally (ip) 10 mg of liophilzed rue/kg of body weight (treated group, n=12) during post implantational period (day 9 -day 17 postcopula), and a control (C) group, n=18, received only distilled water during the same period of time. The LAE of R. chalepensis did not affect negatively the weight of mother but affected the weight of the uterus during the period of treatment (p<0,05). The frequency of fetal resorptions was greater (p<0,05) and a significant decrease of fetal weight (p<0,01) was evidenced in fetuses of the treated group in comparison with the C group. Also there was occurrence of skeletal malformations in the treated group. Efecto del extracto acuoso de Origanum vulgare L. (Lamiaceae) en embriones preimplantacionales de ratón
Efecto de ResumenMaytenus macrocarpa (Chuchuhuasi) es un árbol nativo de la región Amazónica del Perú utilizado como planta medicinal para el tratamiento de muchas enfermedades, sin embargo su efecto sobre el sistema reproductor masculino aún no ha sido elucidado. El objetivo del presente estudio fue evaluar el efecto del extracto acuoso de M. macrocarpa en dosis diarias durante 7 días sobre los parámetros reproductivos de ratones machos. Se utilizó machos maduros de la cepa C57BL divididos en 2 grupos (n=10), Grupo Control (C): NaCl 0.9% y Grupo Tratamiento (T): Extracto acuoso de Chuchuhuasi, ambos suministrado diariamente vía sonda esofágica y al octavo día fueron eutanizados. Se registraron el peso corporal y de los órganos reproductivos: testículo, epidídimo y conducto deferente, se evaluaron la concentración, movilidad y morfología espermática. En nuestros datos observamos diferencias significativas (Prueba t-Student P=0.05) en el peso de la cabeza y cuerpo del epidídimo AbstractMaytenus macrocarpa (chuchuhuasi) is native tree of the Peruvian Amazon used as traditional medicine for the treatment of many diseases, but its effect on the male reproductive system has not yet been elucidated. The aim of this study is evaluate the effect of aqueous extract of M. macrocarpa in daily doses for 7 days on reproductive parameters of male mice. We used C57BL mature male mice divided into 2 groups (n= 10), Control Group (C): 0.9% NaCl and Treatment group (T): Aqueous extract of Chuchuhuasi, both supplied daily via oral gavages. At the eight day of treatment the mice were euthanized. The weight of the body and reproductive organs: testis, epididymis and vas deferens, were registered. Concentration, motility and sperm morphology were evaluated. The results showed significantly differences (t-Student test P<0.05) in the weight of the head and body epididymis (C: 19.25±1.1 vs T: 21.26±2.0), vas deferens (C: 10.61±0.7 vs T: 11.75±0.5), progressive sperm motility (C: 42.16±5.2 vs T: 25.82±8.4) and immobile sperm (C: 36.05±4.9 vs T: 48.51±7.2). No difference in sperm count was observed. The sperm normal morphology diminished with ingest of M. macrocarpa (tStudent test p <0.05) (C: 39.72±1.3 vs T: 30.78±4.9). We conclude that the aqueous extract of chuchuhuasi, has a negative effect on the male reproductive system of mice.Keywords: Maytenus macrocarpa, Chuchuhuasi, mouse, sperm quality, infertility. RESUMENSe evaluó la posible ocurrencia de Hepatozoon caimani en una población de "caimán blanco" Caiman crocodilus en un zoológico de Lima, Perú. Se colectó muestras de sangre periférica de 10 especímenes de ambos sexos y de diversas edades, clínicamente sanos y mantenidos en cautiverio en el Parque Zoológico Huachipa. Se obtuvo tres frotices de sangre por animal y fueron teñidos con May-Grunwald-Giemsa 2%. Se identificó la presencia de hemoparásitos intraeritrocitarios del género Hepatozoon, determinándose por sus aspectos morfológicos que los gamontes corresponden a H. caimani. Three blood smears were prepared per animal and stained with May-Grunwald-Giemsa 2%. Intraerythrocitic haemoparasites of the genus Hepatozoon were identified and due to the morphological aspect they were considered to be H. caimani.
La "maca", Lepidium meyenii Walp, especie vegetal nativa del Perú, es un recurso de gran valor nutricional. Además son varias las investigaciones realizadas en la última década en las cuales se ha comprobado sus efectos sobre la fertilidad de mamíferos. En este trabajo, evaluamos el posible efecto del extracto liofilizado del hipocótilo de L. meyenii Walp en el desarrollo de los embriones preimplantacionales de Mus musculus. Se suministró el liofilizado intra-peritonealmente, en una concentración de 0 g/Kg (grupo control) y 1 g/Kg de peso corporal a dos grupos de ratonas preñadas respectivamente, desde el día 1 hasta el día 4 de preñez. Se evaluaron el estadío de desarrollo, condición y morfología embrionarias, no encontrándose efectos tóxicos del extracto acuoso liofilizado de "maca" en el desarrollo normal de los embriones pre-implantacionales de ratón.
(Primolius couloni) se considera un loro del Nuevo Mundo en peligro de extinción. Para establecer rangos hematológicos de referencia, se tomaron muestras de sangre de 13 especímenes (7 machos, 6 hembras) criados en cautividad en el Zoológico del Patronato del Parque de Las Leyendas de Lima, Perú. El promedio de eritrocitos y leucocitos fue 2608 x 10 3 /ml y de 10.11 x 10 3 /ml, respectivamente; 50.15% de hematocrito y 200 fl de volumen corpuscular medio; asimismo, en el recuento diferencial leucocitario se observó 64.7 y 25.3% de heterófilos y leucocitos, respectivamente. El diámetro mayor y menor de los eritrocitos fue de 11.3 y 7.0 µm, respectivamente. Estos resultados podrían emplearse como valores locales de referencia para esta especie.
RESUMENObjetivos. Evaluar el efecto de las nanopartículas de ZnO, TiO 2 y SiO 2 sobre la viabilidad celular y la expresión génica de las interleuquinas 7 y 3 y del factor estimulante de colonias de granulocito -macrófago (GM-CSF) en Mus musculus. Materiales y métodos. Se extrajo médula ósea roja de cinco roedores (Balb/c) para el estudio de viabilidad celular mediante la prueba de MTT. Por otro lado, grupos cinco roedores fueron inoculados vía intraperitoneal con dosis de 0,5; 1; 2,5; 5 y 10 mg/kg de nanopartículas de ZnO y SiO 2 y de 5; 10; 15; 20 y 25 mg/kg de nanopartículas deTiO 2 , 30 h después, se obtuvo el ARN a partir de la médula ósea roja para los análisis de expresión génica empleando las técnicas de PCR y RT-PCR cuantitativa. Resultados. Las nanopartículas de ZnO y SiO 2 redujeron la viabilidad celular de una manera dosis-dependiente en un 37 y 26%, respectivamente, a partir de una dosis de 1 mg/kg. En cuanto al efecto sobre la expresión génica, a las dosis 5 y 10 mg/kg, las nanopartículas de TiO 2 redujeron en mayor porcentaje la expresión de las interleuquinas 7 y 3 (55,3 y 70,2% respectivamente), con respecto a la expresión del GM-CSF, el mayor porcentaje de reducción lo produjo las nanopartículas de SiO 2 (91%). Las nanopartículas de ZnO redujeron a partir de las dosis de 20 y 25 mg/kg. Conclusiones. Las nanopartículas de ZnO, SiO 2 y TiO 2 alteran la viabilidad celular y la expresión génica en la médula ósea de ratón. EFFECT OF THE INDUSTRIAL NANOPARTICLES TiO 2 , SiO 2 and ZnO ON CELL VIABILITY AND GENE EXPRESSION IN RED BONE MARROW OF Mus musculus ABSTRACT Objectives.To evaluate the effect of ZnO, TiO 2 and SiO 2 nanoparticles on cell viability and expression of the interleukin 7, interleukin 3, and granulocyte-macrophage colony stimulating factor (GM-CSF) genes in Mus musculus. Materials and methods. Red bone marrow was extracted from five Balb/c mice for the analysis of cell viability using the MTT test. The mice were divided into two groups of five each: one group was inoculated intraperitoneally with 0.5, 1.0, 2.5, 5.0, and 10 mg/kg of ZnO and SiO 2 nanoparticles, respectively, and the other group was inoculated with 5.0, 10.0, 15.0, 20.0, and 25 mg/kg of TiO 2 nanoparticles, respectively. Thirty hours later, RNA was extracted from the red bone marrow of the mice in both groups for gene expression analysis using quantitative PCR and RT-PCR. Results. ZnO and SiO 2 nanoparticles reduced cell viability in a dose-dependent manner by 37% and 26%, respectively, starting at a dose of 1 mg/kg. TiO 2 nanoparticles at 5 mg/kg and 10 mg/kg reduced the gene expression of interleukins 7 and 3 by 55.3% and 70.2%, respectively, and SiO 2 nanoparticles caused the greatest decrease (91%) in the expression of GM-CSF.
