Purpose: ABTL0812 is a novel first-in-class, small molecule which showed antiproliferative effect on tumor cells in phenotypic assays. Here we describe the mechanism of action of this antitumor drug, which is currently in clinical development.Experimental Design: We investigated the effect of ABTL0812 on cancer cell death, proliferation, and modulation of intracellular signaling pathways, using human lung (A549) and pancreatic (MiaPaCa-2) cancer cells and tumor xenografts. To identify cellular targets, we performed in silico high-throughput screening comparing ABTL0812 chemical structure against ChEMBL15 database.Results: ABTL0812 inhibited Akt/mTORC1 axis, resulting in impaired cancer cell proliferation and autophagy-mediated cell death. In silico screening led us to identify PPARs, PPARa and PPARg as the cellular targets of ABTL0812. We showed that ABTL0812 activates both PPAR receptors, resulting in upregulation of Tribbles-3 pseudokinase (TRIB3) gene expression. Upregulated TRIB3 binds cellular Akt, preventing its activation by upstream kinases, resulting in Akt inhibition and suppression of the Akt/mTORC1 axis. Pharmacologic inhibition of PPARa/g or TRIB3 silencing prevented ABTL0812-induced cell death. ABTL0812 treatment induced Akt inhibition in cancer cells, tumor xenografts, and peripheral blood mononuclear cells from patients enrolled in phase I/Ib first-in-human clinical trial.Conclusions: ABTL0812 has a unique and novel mechanism of action, that defines a new and drugable cellular route that links PPARs to Akt/mTORC1 axis, where TRIB3 pseudokinase plays a central role. Activation of this route (PPARa/g-TRIB3-AktmTORC1) leads to autophagy-mediated cancer cell death. Given the low toxicity and high tolerability of ABTL0812, our results support further development of ABTL0812 as a promising anticancer therapy.

El colectivo adolescente no es una excepción: las investigaciones señalan una notable prevalencia de este problema, evidenciándose la necesidad de profundizar en su estudio, especialmente en Iberoamérica, dada la escasez de trabajos al respecto. Este estudio tiene como objetivos fortalecer el conocimiento sobre el comportamiento agresivo en parejas adolescentes de Costa Rica. Para ello, se toman en consideración diferentes formas de agresión, en lugar de analizar únicamente agresiones físicas o sexuales; además se estudian posibles diferencias en función del sexo. Basándose en un diseño ex post facto, se aplica un instrumento estandarizado a 468 adolescentes de Heredia (Costa Rica), que analiza cinco formas de agresión: verbal-emocional; física; sexual; relacional y amenazas. Los resultados muestran que son escasos los participantes en la investigación que no cometieron o sufrieron alguna agresión en pareja, especialmente verbales-emocionales o verbales-emocionales y sexuales, si bien la frecuencia de estas fue baja; además, se detectan algunas diferencias significativas intersexos. De los hallazgos obtenidos se concluye la existencia de dificultades en los adolescentes para afrontar, de forma constructiva, conflictos que se presentan en sus relaciones, algo que debe ser abordado de manera integral para incrementar la eficacia de los programas preventivos para jóvenes.
This paper proposes a new argument to explain why media firms silence information that may be relevant to consumers and why this behavior varies across firms. We build on the literature of career concerns and consider firms that seek to maximize their reputation for high quality. Crucial to our results is the idea that media firms can affect, with their reporting strategy, the probability that consumers learn the true state. Reputational concerns dictate that a monopoly firm suppresses scoops, even when evidence is strong. With competition, precise private information is published but weaker though informative signals are silenced. We obtain that silence is higher in media firms with high levels of initial reputation and/or great social influence. We draw predictions on a firm's optimal choice of an editorial standard, the persistence of news suppression when consumers believe one state to be more likely than another and the possibility that silence may be socially beneficial.
El liderazgo para la justicia social es un tema que ha cobrado relevancia en los espacios de diálogo vinculados a temas educativos, en muchos contextos alrededor del mundo y Costa Rica no es la excepción. El objetivo de este estudio es el análisis en relación con esta temática, así como el aporte de ideas para los educadores en formación y en ejercicio; igualmente, para las personas que dirigen el rumbo en materia de políticas educativas del país. Desde un enfoque cualitativo, se implementa el protocolo de entrevista de la Red Internacional de Desarrollo de Liderazgo (ISLDN), que surge como parte de las reflexiones en la Convención de la University Council for Educational Administration (UCEA) en el 2008, el cual está conformado por cinco preguntas. Los sujetos de análisis son dos directivos escolares, concretamente del nivel de preescolar público y de una unidad pedagógica privada. Los resultados sugieren un estilo de liderazgo democrático con algunas características de transformacional, centrados en aspectos de la justicia social como reconocimiento y capacidades de las personas. Además, se hallan coincidencias, en ambos casos, con los aportes teóricos revisados basados en las cuatro categorías de justicia social: económica, cultural, asociativa y para el desarrollo.
