Potato leafroll virus (PLRV), genus Polerovirus, family Luteoviridae, is a major pathogen of potato worldwide. PLRV is transmitted among host plants by aphids in a circulative–nonpropagative manner. Previous studies have demonstrated that PLRV infection increases aphid fecundity on, and attraction to, infected plants as compared to controls. However, the molecular mechanisms mediating this relationship are still poorly understood. In this study, we measured the impact of PLRV infection on plant–aphid interactions and plant chemistry in two hosts: Solanum tuberosum and Nicotiana benthamiana. Our study demonstrates that PLRV infection attenuates the induction of aphid‐induced jasmonic acid and ethylene in S. tuberosum and N. benthamiana. Using transient expression experiments, insect bioassays and chemical analysis, we show that expression of three PLRV proteins (P0, P1, and P7) mediate changes in plant–aphid interactions and inhibition of aphid‐induced jasmonic acid and ethylene in N. benthamiana. This study enhances our understanding of the plant‐vector‐pathogen interface by elucidating new mechanisms by which plant viruses transmitted in a circulative manner can manipulate plant hosts.
Most restoration projects involving invasive plant eradication tend to focus on plant removal with little consideration given to how these invasives change soil microbial communities. However, soil microorganisms can determine invasibility of habitats and, in turn, be altered by invasives once established, potentially inhibiting native plant establishment. We studied soil microbial communities in coastal dunes with varying invasion intensity and different restoration approaches (herbicide, mechanical excavation) at Point Reyes National Seashore. Overall, we found evidence of a strong link between bacterial and fungal soil communities and the presence of invasives and restoration approach. Heavily invaded sites were characterized by a lower abundance of putatively identified nitrifiers, fermentative bacteria, fungal parasites, and fungal dung saprotrophs and a higher abundance of cellulolytic bacteria and a class of arbuscular mycorrhizal fungi (Archaeosporomycetes). Changes in soil microbiota did not fully dissipate following removal of invasives using herbicide, with exception of reductions in cellulolytic bacteria and Archaeosporomycetes abundance. Mechanical restoration effectively removed both invasives and soil legacy effects by inverting or "flipping" rhizome-contaminated surface soils with soils from below and may have inadvertently induced other adverse effects on soils that impeded reestablishment of native dune plants. Land managers should consider additional measures to counteract lingering legacy effects and/or focus restoration efforts in areas where legacy effects are less pronounced.
Composting is an effective strategy to process agricultural and urban waste into forms that may be beneficial to crops. The objectives of this orchard field study were to characterize how a dairy manure compost and a food waste compost influenced: (1) soil nitrogen and carbon pools, (2) bacterial and nematode soil food webs and (3) tree growth and leaf N. The effects of composts were compared with fertilized and unfertilized control plots over two years in a newly planted almond orchard. Both dairy manure compost and food waste compost increased soil organic matter pools, as well as soil nitrate and ammonium at certain time points. Both composts also distinctly altered bacterial communities after application, specifically those groups with carbon degrading potential, and increased populations of bacterial feeding nematodes, although in different timeframes. Unique correlations were observed between nematode and bacterial groups within compost treatments that were not present in controls. Food waste compost increased trunk diameters compared to controls and had greater relative abundance of herbivorous root tip feeding nematodes. Results suggest that recycled waste composts contribute to biologically based nitrogen cycling and can increase tree growth, mainly within the first year after application.
