A Gram-stain-negative, strictly aerobic, moderately halophilic bacterium, designated A-1, was isolated from a tidal flat of the Taean coast in South Korea. Cells were motile rods with a single flagellum showing oxidase-negative and catalase-positive activities and contained poly-β-hydroxyalkanoic acid granules. Growth of strain A-1 was observed at 20-40 °C (optimum, 30 °C), pH 6.0-10.5 (optimum, pH 7.0) and in the presence of 1.0-6.0 % (w/v) NaCl (optimum, 2.0 %). Strain A-1 contained C16 : 0, summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c) and summed feature 8 (comprising C18 : 1ω7c and/or C18 : 1ω6c) as the major fatty acids. The major polar lipids of strain A-1 were phosphatidylethanolamine, phosphatidylglycerol and diphosphatidylglycerol. The isoprenoid quinones detected were ubiquinone-7 and ubiquinone-8. The G+C content of the genomic DNA was 51.5 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain A-1 formed a distinct phylogenetic lineage from other genera within the family Oceanospirillaceae. Strain A-1 shared low 16S rRNA gene sequence similarities with other taxa (≤94.9 %). On the basis of phenotypic, chemotaxonomic and molecular properties, it is clear that strain A-1 represents a novel genus and species of the family Oceanospirillaceae, for which the name Pontibacterium granulatum gen. nov., sp. nov. is proposed. The type strain is A-1 (=KACC 18119=JCM 30136).
A Gram-stain-negative, strictly aerobic bacterium, designated R1-4T, was isolated from soil from a military shooting range in the Republic of Korea. Cells were non-motile short rods, oxidase-positive and catalase-negative. Growth of R1-4T was observed at 15-45 °C (optimum, 30 °C) and pH 6.0-9.0 (optimum, pH 7.0). R1-4T contained summed feature 8 (comprising C18 : 1ω7c/C18 : 1ω6c), summed feature 3 (comprising C16 : 1ω7c/C16 : 1ω6c), cyclo-C19 : 0ω8c and C16 : 0 as the major fatty acids and ubiquinone-10 as the sole isoprenoid quinone. Phosphatidylglycerol, phosphatidylethanolamine, diphosphatidylglycerol, sphingoglycolipid, phosphatidylcholine, an unknown glycolipid and four unknown lipids were detected as polar lipids. The major polyamine was spermidine. The G+C content of the genomic DNA was 64.4 mol%. The results of phylogenetic analysis based on 16S rRNA gene sequences indicated that R1-4T formed a tight phylogenetic lineage with Novosphingobium sediminicola HU1-AH51T within the genus Novosphingobium. R1-4T was most closely related to N. sediminicola HU1-AH51T with a 98.8 % 16S rRNA gene sequence similarity. The DNA-DNA relatedness between R1-4T and the type strain of N. sediminicola was 37.8±4.2 %. On the basis of phenotypic, chemotaxonomic and molecular properties, it is clear that R1-4T represents a novel species of the genus Novosphingobium, for which the name Novosphingobium humi sp. nov. is proposed. The type strain is R1-4T (=KACC 19094T=JCM 31879T).
A Gram-stain-negative, strictly aerobic bacterium, designated HC1, was isolated from an air conditioner in South Korea. Cells were orange, non-motile cocci with oxidase- and catalase-positive activities and did not contain bacteriochlorophyll a. Growth of strain HC1 was observed at 10-45 °C (optimum, 30 °C), pH 4.5-9.5 (optimum, pH 7.0) and 0-3 % (w/v) NaCl (optimum, 0 %). Strain HC1 contained summed feature 8 (comprising C18 : 1ω7c/C18 : 1ω6c), C16 : 0 and cyclo-C19 : 0ω8c as the major fatty acids and ubiquinone-10 as the sole isoprenoid quinone. Phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine and an unknown aminolipid were detected as the major polar lipids. The major carotenoid was hydroxyspirilloxanthin. The G+C content of the genomic DNA was 70.1 mol%. Phylogenetic analysis, based on 16S rRNA gene sequences, showed that strain HC1 formed a phylogenetic lineage within the genus Roseomonas. Strain HC1 was most closely related to the type strains of Roseomonas oryzae, Roseomonas rubra, Roseomonas aestuarii and Roseomonas rhizosphaerae with 98.1, 97.9, 97.6 and 96.8 % 16S rRNA gene sequence similarities, respectively, but the DNA-DNA relatedness values between strain HC1 and closely related type strains were less than 70 %. Based on phenotypic, chemotaxonomic and molecular properties, strain HC1 represents a novel species of the genus Roseomonas, for which the name Roseomonas aerofrigidensis sp. nov. is proposed. The type strain is HC1 (=KACC 19097=JCM 31878).
A Gram-stain-negative and strictly aerobic bacterial strain, designated 6-4T, was isolated from forest soil in Jeju island, South Korea. Cells showing oxidase-positive and catalase-negative reactions were thin and long non-motile rods. Growth of strain 6-4T was observed at 20-35 °C (optimum, 30 °C) and pH 6.0-9.0 (optimum, pH 7.0). Strain 6-4T contained iso-C15 : 0, anteiso-C15 : 0, iso-C15 : 1 G and iso-C17 : 0 3-OH as the major fatty acids and menaquinone-7 (MK-7) as the sole isoprenoid quinone. Phosphatidylethanolamine was the major polar lipid and five unidentified aminolipids, one unidentified aminophospholipid and one unidentified lipid were also detected as minor polar lipids. The G+C content of the genomic DNA was 45.8 mol%. Strain 6-4T was most closely related to Flavihumibacter solisilvae 3-3T with a low 16S rRNA gene sequence similarity (94.2 %) and phylogenetic analysis indicated that the strain formed a distinct phylogenetic lineage from members of the genus Flavihumibacter and other closely related genera. On the basis of phylogenetic inference and phenotypic, chemotaxonomic and molecular properties, strain 6-4T represents a novel species of a new genus of the family Chitinophagaceae, for which the name Niveitalea solisilvae gen. nov., sp. nov. is proposed. The type strain of Niveitalea solisilvae is 6-4T (=KACC 18808T=JCM 31525T). An emended description of the genus Flavihumibacter is also proposed.
