In order to breed new strains of oak mushroom, Lentinula edodes, on the sawdust cultivation, we collected 10 from Korea, Taiwan and China respectively and examined somatic incompatibility, morphological features of fruiting body and productivity. Four strains(SANJO701HO, FMRI2534, FMRI2337 and FMRI2613) shown remarkable results, were confirmed with parental strains. 80 monokaryotic strains derived from the selected 4 parental strains, were selected and 117 hybrid strains were made by mono-mono mating. Aslo, Physiological characteristics were investigated. Average of the mycelial growth of hybrid strains of mating combination SANJO701HO-FMRI2337 was approximately 10% lower than other mating combinations. Overall, This study was founded to develop new varieties of L. edodes. The productivity of new 117 strains on sawdust cultivation needs continued research.
Lentinula edodes is a globally important edible mushroom species that is appreciated for its medicinal properties as well as its nutritional value. During commercial cultivation, a mycelial brown film forms on the surface of the sawdust growth medium at the late vegetative stage. Mycelial film formation is a critical developmental process that contributes to the quantity and quality of the mushroom yield. However, little is known regarding the genetic underpinnings of brown film formation on the surface of mycelial tissue. A novel causal gene associated with the formation of the mycelial brown film, named ABL (Abnormal browning related to light), was identified in this study. The comparative genetic analysis by dihybrid crosses between normal and abnormal browning film cultivars demonstrated that a single dominant allele was responsible for the abnormal mycelium browning phenotype. Whole-genome sequencing analysis of hybrid isolates revealed five missense single-nucleotide polymorphisms (SNPs) in the ABL locus of individuals forming abnormal partial brown films. Additional whole-genome resequencing of a further 16 cultivars showed that three of the five missense SNPs were strongly associated with the abnormal browning phenotype. Overexpression of the dominant abl-D allele in a wild-type background conferred the abnormal mycelial browning phenotype upon transformants, with slender hyphae observed as a general defective mycelial growth phenotype. Our methodology will aid the future discovery of candidate genes associated with favorable traits in edible mushrooms. The discovery of a novel gene, ABL, associated with mycelial film formation will facilitate marker-associated breeding in L. edodes.
In the structural investigation of cultivation facilities, the proportion of farmers to grown with double or triple structure were investigated 96%. It has been shown to grow with a stable cultivation facilities structure against environmental changes. The results of the analysis of the changes in the cultivation environment of Yeoju area in July-august, temperature and humidity of the external instrument shelter was 20.2~29.9 o C and 66.2~99.9% respectively. In the greenhouse 2 model capable of temperature environment regulation in the cultivation facility, temperature and humidity were investigated 19.3~25.7 o C and 81.6~99.9% respectively. Result of the survey of fruiting body characteristics and yields in the greenhous 2, pileus diameter and stipe length were investigated 66.2 and 54.1 mm, yield of mushroom and individual weight were 312 g and 26.6 g. High-temperature environment regulation shows the improvement of the quality and productivity of the mushroom. Power consumption of the air conditioning has been investigated using 56kwh/day, electricity costs were calculated 2195 won/day.
