Purpose: The present study focuses on threshold levels for cytotoxicity after long-term and repetitive exposure for HUVEC as a model for the specific microvascular endothelial system. Furthermore, possible genotoxic effects and functional impairment caused by ZnO NPs in HUVEC are elucidated. Methods: Thresholds for cytotoxic effects are determined by the 3-(4,5-dimethylthiazol-2yl)-2,5-diphenyltetrazolium bromide (MTT) and Annexin V assay. To demonstrate DNA damage, single-cell microgel electrophoresis (comet) assay is performed after exposure to sub-cytotoxic concentrations of ZnO NPs. The proliferation assay, dot blot assay and capillary tube formation assay are also carried out to analyze functional impairment. Results: NPs showed to be spherical in shape with an average size of 45-55 nm. Long-term exposure as well as repetitive exposure with ZnO NPs exceeding 25 µg/mL lead to decreased viability in HUVEC. In addition, DNA damage was indicated by the comet assay after longterm and repetitive exposure. Twenty-four hours after long-term exposure, the proliferation assay does not show any difference between negative control and exposed cells. Forty-eight hours after exposure, HUVEC show an inverse concentration-related ability to proliferate. The dot blot assay provides evidence that ZnO NPs lead to a decreased release of VEGF, while capillary tube formation assay shows restriction in the ability of HUVEC to build tubes and meshes as a first step in angiogenesis. Conclusion: Sub-cytotoxic concentrations of ZnO NPs lead to DNA damage and functional impairment in HUVEC. Based on these data, ZnO NPs may affect neo-angiogenesis. Further investigation based on tissue cultures is required to elucidate the impact of ZnO NPs on human cell systems.
ZusammenfassungShisha-Tabak benötigt im Vergleich zur Zigarette höhere Konzentrationen des Feuchthaltemittels Glycerin. Seit Mai 2016 ist die bis dahin gültige Limitierung von Feuchthaltemitteln in Tabak auf 5 % aufgehoben. Derzeit ist das toxikologische Profil des Glycerins jedoch noch nicht hinreichend erforscht.Ziel dieser Arbeit war es, Glycerin auf mögliche zyto- und genotoxische Effekte zu untersuchen, um so das Gefährdungspotenzial durch Glycerin im Shisha-Tabak zu beurteilen und die tabakkontrollpolitische Situation in Deutschland zu diskutieren. Dafür wurden Lymphozyten sowie Nasenschleimhautzellen von 10 Patienten für eine Stunde Glycerin (0,001 mol/l bis 6,0 mol/l) exponiert. Durch den Trypanblau-Ausschlusstest wurden die Zellen auf Zytotoxizität, mittels Einzelzellgelelektrophorese (Comet Assay) und Mikrokern-Test auf Genotoxizität untersucht.Im Trypanblau-Ausschlusstest traten bei Lymphozyten sowie nasalen Mukosazellen signifikante Vitalitätsabfälle ab Glycerin-Konzentrationen von 1,0 mol/l auf. Im Comet Assay konnten für beide Zellgruppen signifikante Unterschiede des Olive Tail Moments (OTM) ab 1,0 mol/l nachgewiesen werden. Beim Mikrokern-Test zeigten sich keine signifikanten Zunahmen der Mikrokern-Anzahl.Es konnten zyto- und genotoxische Effekte ab Konzentrationen von 1,0 mol/l nachgewiesen werden. Dies überschreitet die reale Glycerin-Belastung im Hauptstromrauch der Shisha jedoch deutlich. Dennoch handelt es sich bei Genotoxizität um ein stochastisches Risiko. Ebenso sind toxische Effekte, beispielsweise durch Erhitzung, bereits bei geringeren Konzentrationen denkbar. Für eine umfangreichere Beurteilung von Feuchthaltemitteln im Shisha-Tabak sind weitere Untersuchungen indiziert. Darüber hinaus besteht enormer Handlungsbedarf zur weiteren Einführung tabakkontrollpolitischer Maßnahmen in Deutschland.
Zusammenfassung Hintergrund Die Studienlage zu Kautabak und Zigarettenrauch ist eindeutig und zeigt karzinogenes Potenzial. Über Schnupftabak ist hingegen wenig bekannt, v. a. auf zellulärer Ebene gibt es keine ausreichenden wissenschaftlichen Publikationen. Somit lässt sich die eventuell mutagene Wirkung von Schnupftabak nur schwer einschätzen. In Konsequenz stützt sich die WHO in ihrer Einstufung des Schnupftabaks als nicht karzinogen auf eine sehr eingeschränkte Datenlage. Ziel Ziel der vorliegenden Arbeit war es, Schnupftabak auf mögliche zyto- und genotoxische Effekte auf humane Lymphozyten und Nasenschleimhautzellen zu untersuchen um ggf. tumorinitiierende Effekte darzustellen. Material und Methoden Eingesetzt wurden eine Schnupftabaksorte ohne Menthol und eine Sorte mit Mentholzusatz. Die benötigten Nasenschleimhautzellen und Lymphozyten wurden von 10 Probanden gewonnen und eine Stunde lang mit einem Schnupftabak-DMSO-Gemisch (2000 µg/ml bis 0,01 µg/ml) inkubiert. Zur Analyse wurde der Trypanblau-Test, der Comet-Assay und der Mikrokerntest verwendet. Ergebnis Der Trypanblau-Test zeigte keinen Abfall der Vitalität. Beim Comet-Assay ergab sich bei Lymphozyten ein signifikanter Anstieg der DNA-Fragmentierung ab 100 µg/ml, bei Nasenschleimhautzellen ab 1000 µg/ml. Der Mikrokerntest wies keine signifikante Zunahme der Mikrokerne auf. Es konnte kein Unterschied zwischen den beiden Tabaksorten aufgezeigt werden. Diskussion Es zeigte sich eine Schädigung der Erbsubstanz im Comet-Assay, die möglicherweise reparabel ist. Irreparable DNA-Schäden im Sinne von Mikrokernen wurden nicht gefunden. Nach diesen Ergebnissen muss die Einstufung der WHO in Zweifel gezogen werden. Untersuchungen mit weiteren Endpunkten der Genotoxizität sind somit gerechtfertigt, um zu einer fundierten Beurteilung des Risikopotenzials von Schnupftabak zu gelangen.
