In this study, a protocol to induce high amount of friable callus of Boerhaavia paniculata RICH and a lipidomics technique were applied to investigate the profile of lipids to relate to those present in the roots of this plant that presented anti-inflammatory activity in the crude hexane extract. The callus culture was induced from seeds in solidified Murashige and Skoog medium containing different amounts of glucose and different concentrations of 2,4-Dichlorophenoxyacetic acid . The explants were kept in a germination chamber at 30±2°C with a photoperiod of 16 h under light intensity of 27 µmol m -2 s -1 for 4 weeks. The best results for friable callus formation and development of the biomass were obtained in the treatment containing 2.26 μM 2.4-D and glucose (1.5 %; w/v). Lipidomics techniques were applied in hexane fraction showing higher concentrations of the steroids β-sitosterol (3.53 mg/100 g dc-dry cells), and fatty acids, especially 2-hydroxy-tetracosanoic acid (0.34 mg/100 g dc), eicosanoic acid (86.25 mg/100 g dc), stearic acid (420.83 mg/100 g dc), tetradecanoic acid (10.74 mg/100 g dc) and linoleic acid (100.61 mg/100 g dc). The lipid profile of callus versus that found in the roots of wild plant is described in this work.

Tancredo Neves nº 2501, Belém-PA, joanne.souza@ufra.edu.br 1 RESUMO Em função da carência na literatura de informações hidrológicas que possam contribuir para o desenvolvimento de uma agricultura sustentável no Pará e considerando a hipótese de que seja possível representar a distribuição de frequência das precipitações através da função Gama, o presente trabalho teve o objetivo de ajustar as distribuições de frequência das precipitações de localidades do estado do Pará e a partir destas determinar e espacializar as precipitações mensais e anuais prováveis, a um nível de 75% de probabilidade (P75%). As precipitações diárias foram obtidas no banco de dados da Agência Nacional de Águas (153 estações pluviométricas). Foram elaboradas 1.836 séries históricas de precipitações mensais e 153 séries de precipitações anuais. A determinação da precipitação provável foi realizada por meio da distribuição Gama, aprovada pelo teste de Kolmogorov-Smirnov em 98,1% das séries históricas. Somente séries mensais foram reprovadas no teste. As P75% foram devidamente espacializadas no estado do Pará e as maiores precipitações concentraram-se na região nordeste com destaque para o litoral do estado e a região próxima ao município de Portel. Na região sudeste do Pará pôde-se observar que as localidades próximas às divisas com Tocantins e Mato-Grosso permanecem durante cinco meses com predominância de P75% menores do que 50 mm. As precipitações prováveis do estado do Pará foram devidamente estimadas e espacializadas, e considerando que a maioria das séries analisadas foram aprovadas no teste de aderência, pode-se afirmar que a função Gama pode ser utilizada para representar a distribuição de frequência das precipitações do estado do Pará. PROBABLE MONTHLY AND ANNUAL RAINFALL FOR THE STATE OF PARÁ, BRAZIL 2 ABSTRACT Considering the scarcity of hydrological information in the literature that may contribute to the development of sustainable irrigated agriculture in Pará and considering the hypothesis that it is possible to represent rainfall frequency distribution through the Gamma function, the present study aimed at adjusting rainfall frequency distributions of localities in Pará state (Brazil) and, based on them, determine and spatialize problable monthly and annual rainfall, with 75% probability. Daily rainfall data were obtained in National Water Agency (ANA) database (153 pluviometric stations). 1,836 historical series of monthly rainfall and 153 annual rainfall series were made. The estimate of probable precipitation was made by Gamma function and this model was approved by the Kolmogorov-Smirnov test in 98.1% of the historical series. Only monthly series failed in the test. The P75% were adequately specialized in Pará state, and the highest rainfall are concentrated in the northeast region with emphasis on the coast of the state and the region near the city of Portel. In the southeast of Pará it was observed that the areas close to Tocantins and Mato Grosso states stayed during five months with predominance of P75% smaller than 50 mm. The probable rainfall of Pará state were adequately estimated and spatialized and the Gamma function can be used to represent therainfall frequency distribution in Pará State, considering that most of the historical series were approved in adherence test. This work aimed to investigate the effects of different combinations of auxin (0, 225 and 450 µM Picloram) and Murashige and Skoog salts (50% and 100% MS salt concentrations, respectively named as ½ MS and MS) in mediate the induction of callus and flowers from immature inflorescences (developmental stages I-9, I-12 and I-15) of the oil palm BRS – Manicoré hybrid. In I-9 inflorescence explants, the rate of callus induction was 17% higher in ½ MS than in MS; while in both I-12 and I-15 inflorescence explants, callus induction was over than 95% regardless of the MS salt concentration. The higher rate of callus induction in I-9 inflorescences was mediated by 450 µM Picloram, while the callus induction rates for either I-12 or I-15 inflorescences were higher than 92% regardless of Picloram concentration. Floral induction was lower in I-9 inflorescence explants (45% floral induction rate) than in both I-12 and I-15 inflorescence explants (floral induction rate higher than 90%). Our results suggest that Picloram (450 µM) and MS salts concentrations (½ MS) led to the highest rates of calluses and flowers induction in younger inflorescence explants (I-9); however, the medium composition is indifferent on callus and flower induction in I-12 and I-15 inflorescences.
Através da micropropagação na área agrícola as plantas medicinais têm sido bastante difundidas com perspectiva para obtenção de novas espécies que servirão como fonte de compostos biologicamente ativos e aprimoramento da produção de fitofármacos. A luz é um dos requisitos mais importantes para o desenvolvimento de plântulas cultivadas in vitro, pois pode influenciar nos aspectos morfológicos, anatômicos e fisiológicos através da irradiância ou irradiância de fótons que incidem numa superfície como a das folhas e com isso fenômenos biológicos importantes, como a fotossíntese, respondem de maneira diferentes na planta. O presente trabalho teve como objetivo avaliar a influência de diferentes irradiâncias de fótons no desenvolvimento de três espécies medicinais e aromáticas. O experimento consistiu em um esquema fatorial 3 X 3 com três diferentes irradiâncias de fótons: 35, 45, e 75 μmol.m-2.s-1 em três espécies: Aeollanthus suaveolens Mart. ex Spreng, Hypericum cavernicola L. B. SM e Hypericum teretiusculum A.St.-Hil. Cada tratamento foi composto por 4 repetições e 3 explantes em cada frasco, os segmentos utilizados foram os nodais e apicais. As variáveis analisadas foram número de brotos, altura dos brotos e a concentração da clorofila. Ocorreu diferença estatística significativa entre as diferentes irradiâncias para variável brotação, com a H. teretiusculum se destacando com 9,17 brotos para irradiância de 45 μmol.m-2.s-1 e para a variável altura dos brotos só houve diferença estatística para a espécie A. suaveollens, se destacando com 7,03 cm para irradiância de 75 μmol.m-2.s-1. Para os teores de clorofila, a espécies que apresentaram as maiores médias foram H. cavernicola e H. teretiusculum, com valores de 11,286 (35 μmol.m-2.s-1) e 10,801 (75 μmol.m-2.s-1) respectivamente, seguidas da A. suaveollens com 9,451 para irradiância de 35 μmol.m-2.s-1. Concluindo que as diferentes irradiâncias influenciam o crescimento e a produção dos pigmentos fotossintéticos diferentemente nas espécies medicinais e aromáticas avaliadas.
