BackgroundUveal melanoma(UVM) is the most common intraocular malignancy and has a poor prognosis. The clinical significance of necroptosis(NCPS) in UVM is unclear.MethodsWe first identified necroptosis genes in UVM by single-cell analysis of the GSE139829 dataset from the GEO database and weighted co-expression network analysis of TCGA data. COX regression and Lasso regression were used to construct the prognostic model. Then survival analysis, immune microenvironment analysis, and mutation analysis were carried out. Finally, cell experiments were performed to verify the role of ITPA in UVM.ResultBy necroptosis-related prognostic model, UVM patients in both TCGA and GEO cohorts could be classified as high-NCPS and low-NCPS groups, with significant differences in survival time between the two groups (P<0.001). Besides, the high-NCPS group had higher levels of immune checkpoint-related gene expression, suggesting that they might be more likely to benefit from immunotherapy. The cell experiments confirmed the role of ITPA, the most significant gene in the model, in UVM. After ITPA was knocked down, the activity, proliferation, and invasion ability of the MuM-2B cell line were significantly reduced.ConclusionOur study can provide a reference for the diagnosis and treatment of patients with UVM.
The near infrared transmission of corrugated metal films deposited on hetero-colloidal crystals is investigated. The transmission response of the quasi-three-dimensional (quasi-3D) metal film is modified by controlling the nominal thickness of a dielectric layer pre-deposited on the top surface of the colloidal crystal to form a new hetero-colloidal crystal. An extraordinary optical transmission (EOT) phenomenon could be presented in such metallodielectric (MD) architectures. We have found that the main transmission peak is suppressed as the thickness of the intercalated dielectric layer is increased. We propose that the observed EOT is a result of constructive interference between a localized sphere-like plasmon mode and an index-guided eigen mode mainly confined in the colloidal crystal, which is confirmed by our numerical simulations. Based on the MD microstructures, a distinct plasmon sensitivity response difference is achieved, which indicates potential applications for biochemical sensing.

Here we report an experimental study of the temperature dependence of the current and applied field thresholds for droplet soliton formation, as well as the nanocontact's electrical characteristics associated with droplet dynamics. Nucleation requires lower current densities at lower temperatures, in contrast to typical spin-transfer-torque-induced switching between static magnetic states. Magnetoresistance and electrical noise measurements (10 MHz-1 GHz) show that droplet solitons become more stable at lower temperature. These results are of fundamental interest in understanding the influence of thermal noise on droplet solitons and have implications for the design of devices using the spin-transfertorque effects to create and control collective spin excitations.
Magnetic droplet solitons were first predicted to occur in materials with uniaxial magnetic anisotropy due to a long-range attractive interaction between elementary magnetic excitations, magnons. A non-equilibrium magnon population provided by a spin-polarized current in nanocontacts enables their creation and there is now clear experimental evidence for their formation, including direct images obtained with scanning x-ray transmission microscopy. Interest in magnetic droplets is associated with their unique magnetic dynamics that can lead to new types of high frequency nanometer scale oscillators of interest for information processing, including in neuromorphic computing. However, there are no direct measurements of the time required to nucleate droplet solitons or their lifetime–experiments to date only probe their steady-state characteristics, their response to dc spin-currents. Here we determine the timescales for droplet annihilation and generation using current pulses. Annihilation occurs in a few nanoseconds while generation can take several nanoseconds to a microsecond depending on the pulse amplitude. Micromagnetic simulations show that there is an incubation time for droplet generation that depends sensitively on the initial magnetic state of the nanocontact. An understanding of these processes is essential to utilizing the unique characteristics of magnetic droplet solitons oscillators, including their high frequency, tunable and hysteretic response.
Objectives The present study is aimed at evaluating the diagnostic value of combining shear wave elastography (SWE) parameters and the thyroid imaging reporting and data system (TIRADS) for differentiating between benign and malignant thyroid nodules. Methods Patients who underwent conventional ultrasonography (US) and SWE before surgery were enrolled in the current study. Each nodule was given a TIRADS risk score. The effectiveness of the SWE parameters was assessed by odds ratios (ORs). The SWE scoring risk stratification was proposed beyond 95% probability, and the desired values were obtained according to the log-normal distribution. The area under the receiver-operating characteristic (AUC) was used to compare the diagnostic performance between TIRADS-alone and TIRADS + SWE. Results A total of 262 patients with 298 thyroid nodules were enrolled in our study. The pathological analyses were conducted on 121 benign and 177 malignant nodules. The AUC values for TIRADS-alone and TIRADS + SWE were 0.896 (accuracy 83.2%) and 0.917 (accuracy 84.2%), respectively. However, the TIRADS + SWE scores showed a higher specificity (88.4%) and positive predictive value (91.2%) as compared with the TIRADS-alone of 73.6% and 83.2%, respectively. Conclusions Combining SWE and TIRADS improves the specificity of TIRADS-alone in differentiating between benign and malignant thyroid nodules.
Objectives: To explore the predictive value of radiomics nomogram using pretreatment ultrasound for disease-free survival (DFS) after resection of triple negative breast cancer (TNBC). Methods and materials: A total of 486 TNBC patients from 3 different institutions were consecutively recruited for this study. They were categorized into the primary cohort (n = 216), as well as the internal validation cohort (n = 108) and external validation cohort (n = 162). In primary cohort, least absolute shrinkage and selection operator logistic regression algorithm was used to select recurrence-related radiomics features extracted from the breast tumor and peritumor regions, and a radiomics signature was constructed derived from the grayscale ultrasound images. A radiomic nomogram integrating independent clinicopathological variables and radiomic signature was established with uni- and multivariate cox regressions. The predictive nomogram was validated using an internal cohort and an independent external cohort regarding abilities of discrimination, calibration and clinical usefulness. Results: The patients with higher Rad-score had a worse prognostic outcome than those with lower Rad-score in primary cohort and two validation cohorts (All p < 0.05).The radiomics nomogram indicated more effective prognostic performance compared with the clinicopathological model and tumor node metastasis staging system (p < 0.01), with a training C-index of 0.75 (95% confidence interval (CI), 0.71–0.80), an internal validation C-index of 0.73 (95% CI, 0.69–0.78) and an external validation 0.71 (95% CI,0.66–0.76). Moreover, the calibration curves revealed a good consistency for survival prediction of the radiomics model. Conclusions: The ultrasound-based radiomics signature was a promising biomarker for risk stratification for TNBC patients. Furthermore, the proposed radiomics modal integrating the optimal radiomics features and clinical data provided individual relapse risk accurately. Advances in knowledge: The radiomics model integrating radiomic signature and independent clinicopathological variables could improve individual prognostic evaluation and facilitate therapeutic decision-making, which demonstrated the incremental value of the radiomics signature for prognostic prediction in TNBC.
