Nucleus pulposus-like differentiation is always the challenge with application of stem cells for intervertebral disc repair. The combination of injectable biomaterials and stem cells may provide a resolution for this problem, as the transmembrane force can affect the intracellular environment through integrin αβ. In this study, we developed a strategy of photo-crosslinked gelatin-hyaluronic acid methacrylate (GelHA) hydrogel to commit the nucleus pulposus-like differentiation of adipose stromal cells (ASCs) for intervertebral disc repair. ASCs were isolated and cultured in GelHA hydrogel to evaluate nucleus pulposus-like differentiation. The function of integrin αvβ6 was investigated with neutralising antibody. The efficacy of ASCs with GelHA hydrogel for intervertebral disc repair was studied in a rat model of intervertebral disc degeneration. The results showed that GelHA hydrogel promoted ASCs nucleus pulposus-like differentiation and that integrin αvβ6 neutralising antibody prevented ASCs from expression of nucleus pulposus matrix in vitro. The combination of GelHA hydrogel and ASCs promoted quality intervertebral disc repair in rats with much more nucleus pulposus matrix and significantly higher disc height index. The findings have demonstrated that the combination of photo-crosslinked GelHA hydrogel and ASCs can commit ASCs to nucleus pulposus-like differentiation and improve the efficacy of ASCs for intervertebral disc repair. These findings suggest a promising stem cellbased strategy for intervertebral disc repair.
Intervertebral disc degeneration is associated with back pain and disc herniation. This study established a modified protocol for intervertebral disc (IVD) decellularization and prepared its extracellular matrix (ECM). By culturing mesenchymal stem cells (MSCs)(3, 7, 14 and 21 days) and human degenerative IVD cells (7 days) in the ECM, implanting it subcutaneously in rabbit and injecting ECM microparticles into degenerative disc, the biological safety and efficacy of decellularized IVD was evaluated both in vitro and in vivo. Here, we demonstrated that cellular components can be removed completely after decellularization and maximally retain the structure and biomechanics of native IVD. We revealed that allogeneic ECM did not evoke any apparent inflammatory reaction in vivo and no cytotoxicity was found in vitro. Moreover, IVD ECM can induce differentiation of MSCs into IVD-like cells in vitro. Furthermore, allogeneic ECM microparticles are effective on the treatment of rabbit disc degeneration in vivo. In conclusion, our study developed an optimized method for IVD decellularization and we proved decellularized IVD is safe and effective for the treatment of degenerated disc diseases.
Previous studies proposed that serum uric acid (UA), an endogenous antioxidant, could be a protective factor against bone loss. However, recently, a study with a population of US adults did not note the protective effects of serum UA. Therefore, the exact association between serum UA and bone health remains unclear. We performed a retrospective consecutive cohort study in a Chinese population to examine the association between serum UA and bone health. This cross-sectional study included 17,735 individuals who underwent lumbar spine bone mineral density (BMD) measurements as part of a health examination. In covariance analyses (multivariable-adjusted), a high serum UA level was associated with a high BMD, T-score, and Z-score. In binary logistic regression analyses (multivariable-adjusted), a high serum UA level was associated with low odds ratios (ORs) for at least osteopenia and osteoporosis in male (age ≥50 years) (OR = 0.72–0.60 and OR = 0.49–0.39, respectively) and postmenopausal female participants (OR = 0.61–0.51 and OR = 0.66–0.49, respectively). In conclusion, serum UA is associated with BMD, the T-score, and the Z-score, and has a strong protective effect against at least osteopenia and osteoporosis.
Intracellular ion channel inositol 1,4,5-triphosphate receptor (IP3R1) releases Ca2+ from endoplasmic reticulum. The disturbance of IP3R1 is related to several neurodegenerative diseases. This study investigated the mechanism of IP3R1 in myocardial ischemia/reperfusion (MI/R). After MI/R modeling, IP3R1 expression was silenced in myocardium of MI/R rats to explore its role in the concentration of myocardial enzymes, infarct area, Ca2+ level, NLRP3/Caspase-1, and pyroptosis markers and inflammatory factors. The adult rat cardiomyocytes were isolated and cultured to establish hypoxia/reperfusion (H/R) cell model. The expression of IP3R1 was downregulated or ERP44 was overexpressed in H/R-induced cells. Nifedipine D6 was added to H/R-induced cells to block Ca2+ channel or Nigericin was added to activate NLRP3. IP3R1 was highly expressed in myocardium of MI/R rats, and silencing IP3R1 alleviated MI/R injury, reduced Ca2+ overload, inflammation and pyroptosis in MI/R rats, and H/R-induced cells. The binding of ERP44 to IP3R1 inhibited Ca2+ overload, alleviated cardiomyocyte inflammation, and pyroptosis. The increase of intracellular Ca2+ level caused H/R-induced cardiomyocyte pyroptosis through the NLRP3/Caspase-1 pathway. Activation of NLRP3 pathway reversed the protection of IP3R1 inhibition/ERP44 overexpression/Nifedipine D6 on H/R-induced cells. Overall, ERP44 binding to IP3R1 inhibits Ca2+ overload, thus alleviating pyroptosis and MI/R injury.
Termites are eusocial arthropod decomposers, and improve soil fertility, crop yield, and also are used by humans for their benefits across the world. However, some species of termites are becoming a threat to the farming community as they are directly and indirectly causing major losses to the agricultural system. It is estimated that termites cost the global economy more than 40 billion USD annually, and considerable research has been done on their management. In this review, we present the available information related to sustainable and integrated termite management practices (ITM). Furthermore, we insist that the better management of this menace can be possible through: (i) improving traditional methods to keep termites away from crops; (ii) improving agricultural practices to maintain plants with more vigor and less susceptible to termite attack; and (iii) integration of available techniques to reduce termite infestation in crops and surroundings. The application of an effective combination of traditional practices with recently developed approaches is the best option for agricultural growers. Moreover, keeping in mind the beneficial nature of this pest, more innovative efforts for its management, particularly using rapidly emerging technology (e.g., RNA interference), are needed.
Bone metastasis is a severe complication of advanced breast cancer, resulting in osteolysis and increased mortality in patients. Raddeanin A (RA), isolated from traditional Chinese herbs, is an oleanane-type triterpenoid saponin with anticancer potential. In this study, we investigated the effects of RA in breast cancer-induced osteolysis and elucidated the possible mechanisms involved in this process. We first verified that RA could suppress osteoclast formation and bone resorption in vitro. Next, we confirmed that RA suppressed Ti-particle-induced osteolysis in a mouse calvarial model, possibly through inhibition of the SRC/AKT signaling pathway. A breast cancer-induced osteolysis mouse model further revealed the positive protective effects of RA by micro-computed tomography and histology. Finally, we demonstrated that RA inhibited invasion and AKT/mammalian target of rapamycin signaling and induced apoptosis in MDA-MB-231 cells. These results indicate that RA is an effective inhibitor of breast cancer-induced osteolysis.
