Low light is a major adversity affecting yield and quality of summer maize in the Huang‐Huai‐Hai region of China. We conducted a field experiment to explore the effects of shading on root development and yield formation in two summer maize hybrids (Zea mays L.), Denghai605 (DH605) and Zhengdan958 (ZD958). The experiment consisted of four treatments (CK: ambient sunlight, S1: shading from tasselling to physiological maturity stage, S2: shading from six‐leaf to tasselling stage, S3: shading from seeding to physiological maturity stage). Shading had a strong impact on the development of roots in the upper soil layer. Shading significantly decreased the root dry weight, root/shoot ratio, root length density, root absorption area and active absorption area. The results showed that treatments in a diminishing sequence of effects on root from S3, S1, S2 to CK. Overall, shading decreased the root morphologic and activity indices, and decreased yield in summer maize. During an average of 2 years, yields of ZD958 in S3, S2 and S1 decreased by 85%, 24% and 55%, yields of DH605 in S3, S2 and S1 decreased by 87%, 26% and 67%, in compare to CK. The results will be useful for hybrid selection and improving cultural practices for enhancing the maize shading resistance in Huang‐Huai‐Hai region.

Maize is one of the most important crops globally that provides food, feed, and bioenergy. However, shading stress threatens maize production. In this study, we investigated the effects of shading on photosynthate accumulation and distribution of summer maize in the field. Zhengdan958 (ZD958) and Denghai 605 (DH605) were used as experimental materials in a field experiment running from 2013 to 2015. Shading treatments were applied over different growth stages: from the tassel stage (VT) to physiological maturity (R6) (S1), from the six-leaf stage (V6) to VT (S2), and from emergence stage (VE) to R6 (S3). The effects of shading on plant photosynthesis, photosynthate accumulation and distribution, and yield were evaluated in comparison to ambient sunlight. Shading significantly decreased the leaf area, SPAD value, net photosynthetic rate, dry matter accumulation, and grain yield. During the 3-year experimental period, grain yields of ZD958 and DH605 were reduced by 83.4%, 34.2%, 53.1% and 79.3%, 24.2%, 57.6% as compared to the CK by treatments S3, S2, and S1, respectively. 13CO2 stable isotope tracing revealed that shading differentially affected the photosynthate transfer rate in different stages; photosynthates were transferred from top to bottom plant parts, in the order control > S2 > S1 > S3. We conclude that shading clearly disrupted photosynthate metabolism, and reduced the photosynthate accumulation in the grain, resulting in a yield reduction.

Maize (Zea mays L.) is the important crop over the world. Nitrogen (N) as necessary element affects photosynthetic characteristics and grain yield of summer maize. In this study, N0 (0 kg N ha-1), N1 (129 kg N ha-1), N2 (185 kg N ha-1), and N3 (300 kg N ha-1) was conducted using hybrid ‘ZhengDan958’ at Dawenkou research field (36°11′N, 117°06′E, 178 m altitude) in the North China Plain to explore the effects of N rate on photosynthetic characteristics and chloroplast ultrastructure. Gas exchange parameters, chlorophyll fluorescence parameters, leaf area index (LAI), chlorophyll SPAD value, chloroplast ultrastructure, dry matter weight and grain yield were measured. At physiological maturity stage, dry matter weight and grain yield of N2 increased by 33–52% (P ≤ 0.05) and 6–32% (P ≤ 0.05), respectively, compared with other treatments. During the growing from silking (R1) to milk (R3) stage, LAI of N0 and N1 were 35–38% (P ≤ 0.05) and 9–23% (P ≤ 0.05) less than that of N2, respectively. Chlorophyll SPAD value of N0 and N1 were 13–22% (P ≤ 0.05) and 5–11% (P ≤ 0.05) lower than that of N2. There was no significant difference in LAI and chlorophyll SPAD value between N2 and N3 during the period from R1 to R3 (P > 0.05). The net photosynthetic rate (Pn), maximal quantum efficiency of PSII (Fv/Fm) and quantum efficiency of PSII (ΦPSII) were higher with the increase of N rate up to N2 (P ≤ 0.05), and those of N3 were significantly less than N2 (P ≤ 0.05). In compared with N2, the chloroplast configuration of N0 and N1 became elliptical, almost circular or irregular. The membrane of chloroplast and thylakoid resolved with growing stage, and the number of chloroplast per cell and lamellae per grana decreased under N0 and N1 treatment (P ≤ 0.05). Under N0 and N1 treatments, summer maize had more negative photosynthetic characteristics. The more number of osmium granule and vesicle and the larger gap between lamellae were shown in N3. Therefore, N2 treatment, 185 kg N ha-1, is the appropriate application rate for grain yield, photosynthesis and chloroplast ultrastructure.

Biomass-derived carbon dots (CDs) are promising nanotools for agricultural applications and function as a reactive oxygen species (ROS) scavenger to alleviate plant oxidative stress under adverse environments. Nevertheless, plants need ROS burst to fully activate Ca 2+ -regulated defensive signaling pathway. The underlying mechanism of CDs to improve plant environmental adaptability without ROS is largely unknown. Here, Salvia miltiorrhiza-derived CDs triggered ROS-independent Ca 2+ mobilization in plant roots. Mechanistic investigation attributed this function mainly to the hydroxyl and carboxyl groups on CDs. CDs-triggered Ca 2+ mobilization was found to be dependent on the production of cyclic nucleotides and cyclic nucleotide-gated ion channels. Lectin receptor kinases were verified as essential for this Ca 2+ mobilization. CDs hydroponic application promoted Ca 2+ signaling and plant environmental adaptability under salinity and nutrient-deficient conditions. All these findings uncover that CDs have a Ca 2+ -mobilizing property and thus can be used as a simultaneous Ca 2+ signaling amplifier and ROS scavenger for crop improvement.

Background: Shade stress, a universal abiotic stress, suppresses plant growth and production seriously. However, little is known regarding the protein regulatory networks under shade stress. To better characterize the proteomic changes of maize leaves under shade stress, 60% shade (S) and supplementary lighting (L) on cloudy daylight from tasseling stage to physiological maturity stage were designed, the ambient sunlight treatment was used as control (CK). Isobaric tag for relative and absolute quantification (iTRAQ) technology was used to determine the proteome profiles in leaves. Results: Shading significantly decreased the SPAD value, net photosynthetic rate, and grain yield. During two experimental years, grain yields of S were reduced by 48 and 47%, and L increased by 6 and 11%, compared to CK. In total, 3958 proteins were identified by iTRAQ, and 2745 proteins were quantified including 349 proteins showed at least 1.2-fold changes in expression levels between treatments and CK. The differentially expressed proteins were classified into photosynthesis, stress defense, energy production, signal transduction, and protein and amino acid metabolism using the Web Gene Ontology Annotation Plot online tool. In addition, these proteins showed significant enrichment of the chloroplasts (58%) and cytosol (21%) for subcellular localization. Conclusions: 60% shade induced the expression of proteins involved in photosynthetic electron transport chain (especially light-harvesting complex) and stress/defense/detoxification. However, the proteins related to calvin cycle, starch and sucrose metabolisms, glycolysis, TCA cycle, and ribosome and protein synthesis were dramatically depressed. Together, our results might help to provide a valuable resource for protein function analysis and also clarify the proteomic and physiological mechanism of maize underlying shade stress.
