This study was aimed at finding the optimal conditions for hydrogen production based on statistical experiments and using a simulation approach. A Plackett–Burman design and steepest ascent were used to screen the key factors to obtain the best hydrogen concentration. According to the regression analysis, cysteine, acetate, and aeration had the best effect. The optimal conditions, using the method of steepest ascent, were aeration (0.125 L/min), acetate (200 mg/L), cysteine (498 mg/L). Once this was determined, an experiment with more than two factors was considered. The combinations: acetate + cysteine without aeration and cysteine without aeration increased hydrogen concentration. These last two criteria were used to validate the dynamic model based on unstructured kinetics. Biomass, nitrogen, acetate, and hydrogen concentrations were monitored. The proposed model was used to perform the multi-objective optimization for various desired combinations. The simultaneous optimization for a minimum ratio of cysteine-acetate improved the concentration of hydrogen to 20 mg/L. Biomass optimized the concentration of hydrogen to 11.5 mg/L. The simultaneous optimization of reaction time (RT) and cysteine improved hydrogen concentration to 28.19 mg/L. The experimental hydrogen production was 11.4 mg/L at 24 h under discontinuous operation. Finally, the proposed model and the optimization methodology calculated a higher hydrogen concentration than the experimental data.
Las reacciones de hipersensibilidad tipo I o alergias se distinguen por presentar una respuesta inmune a antígenos ambientales. Están implicadas células secretoras de citocinas tipo 2 (linfocitos Th2 e ILC2), eosinófilos, linfocitos B productores de IgE que se unen a receptores en mastocitos y basófilos, liberando mediadores que aumentan la permeabilidad vascular, vasodilatación, contracción del musculo liso bronquial y visceral e inflamación, siendo la manifestación sistémica más grave el choque anafiláctico. Una alternativa de tratamiento regulador de la respuesta a alérgenos es el Factor de Transferencia (FT), el cual se consigue a partir de un Extracto Dializable Leucocitario (EDL) obtenido del lisado de células linfoides, que contiene al menos 200 diferentes moléculas entre 1 a 12 kDa. De ellas a la fracción de péptidos con pesos moleculares (PM) entre 3.5 y 5 kDa se les denomina FT, con actividad inmunomoduladora interespecífica. El FT está compuesto por péptidos hidrofílicos polares, con partes ácidas y dos regiones: una variable y una constante. Debido a su baja probabilidad de transmisión de zoonosis se seleccionó al caballo para la obtención del EDL y purificación del FT. Su obtención se llevó a cabo a partir de la lisis de células sanguíneas, ultra filtración e identificación de péptidos. El FT así obtenido tuvo un 51.3% de incremento en concentración comparado con el FT humano comercial con un patrón electroforético similar. A partir del FT de caballo purificado se evalúo su efecto en el control de reacciones alérgicas en cobayo previamente sensibilizado con ovoalbúmina. Cuatro grupos de cobayos fueron estudiados: un grupo control sensibilizado con ovovalbúmina, uno al que se le administró el FT humano, otro que se le dosificó FT de caballo y un cuarto grupo que no se le administró FT como control negativo. El FT de caballo registró un efecto inmunomodulador sobre las reacciones alérgicas equivalente al presentado con el FT humano, por tanto es una alternativa para el tratamiento de alergias en humanos.
Factores ambientales, genéticos así como malos hábitos de la vida contemporánea han deteriorado su calidad en la población, incrementándose los padecimientos crónicos degenerativos como enfermedades cardiacas, Diabetes mellitus o el cáncer. En el caso del cáncer, la cirugía o la quimioterapia afectan drásticamente la calidad de vida del paciente. Investigaciones recientes buscan sustancias inocuas para su tratamiento como la fototerapia, que se basa en la administración de oxo clorofila como derivado del pigmento, la cual es absorbida por células con alta división celular como las cancerígenas y activada al exponer a las células que la acumularon a luz roja causando la formación de radicales libres que alteran el metabolismo celular. Por ello, se obtuvo la clorofila A a partir de Chlorella spp. en fotobiorreactores con medio basal de Bold modificado, utilizando como fuente de carbono almidón de arroz en fotoperiodos de 12 h. La clorofila A se purifico por cromatografía en columna de sílica gel eluyendo con etanol absoluto y se concentró por rotavapor. La oxoclorofila A se obtuvo por pirolisis en condiciones anóxicas y por acidificación. Se midió su actividad antioxidante por el método del guayacol. Posteriormente, se determinó su efecto citotóxico en Artemia salinaa diluciones 1:13, 1:20 y 1:40 de oxoclorofila A, registrando una mayor citotoxicidad en la obtenida por pirolisis, donde la DL50 calculada (Graphpad Prism) fue de 1.198%. En trabajos futuros se evaluará su efecto citotóxico en líneas celulares humanas sanas y malignas y en ratón.
