AbstractC optoborus ochromactonus, is a recently described scolytid pest associated with the balsa plantations, causing damage and death in young trees (from 12 to 36 months). The objectives were: Characterize the distribution of damage into the tree, define the incidence of C. ochromactonus in two years old plantations in Buena Fe and El Empalme, and evaluate dasometrics parameters of its host. C. ochromactonus has two types of damages that correspond to the initial attack, when the shaft is unsuccessful and no mass attack, whereas when it occurs in upper branches and exceed 300 drillings produces symptoms of dieback and then spike the shaft. The incidence of injury was higher in El Empalme 22.90%, attacking trees 20.10 cm dbh and 18.40 m high, relative to Buena Fe, with 11.20%, 18.20 attacking trees dbh cm and 15.90 m high
Knowledge about pollen biology is fundamental for the planning of controlled crosses in programs of conservation and genetic improvement of plant species. The objectives of this research were to determine the ideal medium for in vitro germination of pollen and to evaluate the effect of temperature and drying on the viability of stored pollen from Ochroma pyramidale. Pollen was obtained from flowers collected before anthesis. The appropriate culture medium for in vitro germination of pollen was determined by testing modifications of the medium proposed by Brewbaker and Kwack. Pollen drying was performed during different times (60, 120, 180 and 240 minutes) at 29 °C and 45 % relative humidity. These samples were stored under temperatures of 5,-20 and-196 °C, and evaluated for 45 days. The ideal medium for pollen germination was composed of 10 % sucrose, 30 mg L-1 boric acid, 430 mg L-1 calcium nitrate. The initial viability calculated by in vitro germination was high, which indicates that the individuals evaluated can be used as pollen donors in the controlled crossing plans. Humidity below 5 % allowed keeping the pollen stored, independently of the storage temperature during the 45 days of the test, without significant decrease in their viability and germination rate. However, pollen viability decreased rapidly when it was stored with high moisture content, regardless of storage temperatures.
Palabras clave: cultivo in vitro, organogénesis, Nothofagus alpina, medios de cultivo, enraizamiento. Abstract Nothofagus alpina is one of the most important forest species in the south of Chile and Argentina, where genetic improvement programs have been started. Micropropagation is a very useful technique when cloning trees for improvement purposes, so the objectives stated in this investigation were: to determine the best of 6-bencilaminopurine phytohormones (BAP) and the ideal culture medium to induce bud rooting and N. alpina nodal section sprouting. Murashige Skoog (MS) and Wood Plant (WPM) media were evaluated with half macronutrients (rooting) and full micronutrients (multiplication), supplemented with 0.10% peptone, 2.00% saccharose, 0.70% agar and different auxin concentrations. MS medium supplemented with 2 mg L -1 AIB was found to be the best for inducing root formation in both number and length of the main root. The greatest percentage of rooting was shown by WPM supplemented with 2 mg L -1 AIB. Treatments with cytokinin for inducing multiplication originated calluses and failed to root, with a prevalence of calluses in WPM medium. The application of cytokinin is not indispensable for sprouting since in witness treatments, a greater number of buds was observed. This leads to the idea that the plant material used contains enough endogenous levels of this phytohormone
