ResumoSenna reticulata (Willd.) H. S. Irwin & Barneby é uma planta arbórea pioneira em áreas abertas de planícies aluviais da Amazônia e frequentemente usada como planta medicinal. Devido à escassez de informações sobre a diversidade da espécie, caracterizou-se a diversidade genética de populações naturais de S. reticulata por meio de marcadores AFLP. Foram coletadas amostras em quatro populações ao longo do trecho do gasoduto Coari-Manaus: Santa Luzia do Baixio, Nossa Senhora de Nazaré, Matrinchã e Esperança II. Foram utilizados quatro primers de AFLP, resultando num total de 330 locos polimórficos variando de 76,97% a 92,42% entre as populações. O dendrograma e a análise bayesiana de agrupamento permitiram a visualização de um grupo formado pelas populações de Nossa Senhora de Nazaré e Esperança II e outro agrupamento formado por Santa Luzia do Baixio. A população de Matrinchã foi caracterizada como a composição de genes dos dois grupos. O fluxo gênico médio estimado entre as populações foi de 4,04 e o teste de Mantel indicou baixa correlação (r = 0,268) entre as matrizes de distância geográfica e distância genética. De acordo com a análise molecular de variância, a maior diferenciação genética está dentro das populações (82,29%), enquanto a variação interpopulacional foi de 17,71%. Irwin & Barneby is a pioneer tree that grows in open areas of the Amazon floodplains and is often used as a medicinal plant species. Due to the lack of information on diversity for this species, this research aimed to characterize the genetic diversity of natural populations of S. reticulata by AFLP markers. The samples were collected in four populations along the natural gas pipeline pathway of Coari-Manaus: Santa Luzia do Baixio, Nossa Senhora de Nazaré, Matrinchã and Esperança II. Four AFLP primers were used, resulting in a total of 330 polymorphic loci ranged from 76.97% to 92.42% among populations. The clustering and Bayesian cluster analysis allowed to discriminate a group composed by the Nossa Senhora de Nazaré and Esperança II populations and another group composed by Santa Luzia do Baixio. Matrinchã population was characterized as a gene composition from both groups. Average gene flow among populations was 4.04 and Mantel test showed low correlation (r = 0.268) between the pairwise of geographic and genetic distance matrices. According to molecular variance analysis, the most genetic differentiation is within populations (82.29%), while the inter population variation was around 17.71%.
Sementes de leguminosas apresentam alta concentração de inibidores de tripsina; estas proteínas estão envolvidas no metabolismo celular e também em mecanismos de defesa vegetal. A fim de confirmar ou não, a possível ação fungicida destas proteínas a partir de extratos de sementes de leguminosas arbóreas, o objetivo deste estudo foi detectar inibidores de tripsina em sementes de Caesalpinia ferrea (CfTI) e Swartzia polyphylla (SpTI) e testar os extratos contra os fungos fitopatogênicos Colletotrichum guaranicola, Corynespora cassiicola, Fusarium oxysporum e Sclerotium rolfsii, avaliando o crescimento micelial e a esporulação. Para tanto, amostras do material biológico vegetal, sementes finamente pulverizadas, foram submetidas à extração em NaCl 150 mM. Os extratos protéicos foram parcialmente purificados em coluna Sephadex G-100, submetidos à detecção dos inibidores e SDS-PAGE (12,5%) e, utilizados nos bioensaios contra os fungos. O perfil eletroforético revelou uma única banda em CfTI e oito bandas em SpTI. Os extratos de C. ferrea e S. polyphylla exibiram efeito na diminuição da esporulação dos fungos testados, mas S. rolfsii foi inibido apenas por C. ferrea. Quanto ao crescimento micelial, os dois extratos tiveram efeito sobre F. oxysporum e S. rolfsii, ao passo que C. guaranicola foi inibido apenas por S. polyphylla, e C. cassiicola por C. ferrea. Concluiu-se que sementes de C. ferrea e S. polyphylla apresentam inibidores de tripsina. Além disso, os resultados sugerem que estas espécies de leguminosas arbóreas são promissoras no que concerne à prospecção de fungicidas naturais, uma vez que os extratos diminuíram o crescimento micelial e a esporulação de C. guaranicola, C. cassiicola, F. oxysporum e S. rolfsii.
Corynespora cassiicola is a cosmopolitan ascomycete widely known as phytopathogen in several crops, and more recently as an emerging pathogen in humans. In this study the genetic variability of 60 isolates of Corynespora cassiicola from different hosts and cities of Amazonas was evaluated, using AFLP molecular markers. Seven genetic groups were identified according to a dendrogram obtained by the Unweighted Pair Group Method using Arithmetical Averages, indicating significant variability among the isolates. Three isolates of different hosts (28, obtained from papaya; 55, obtained from cucumber; and 58, from tomato) remained as single individuals in distinct groups, suggesting marked genetic variation in comparison to the other isolates and possible specificity by the host.
A mancha-alvo, causada pelo fungo Corynespora cassiicola, é a principal doença da parte área do tomateiro na região Norte do Brasil. No Estado do Amazonas o patógeno em plantas de pepino, causa severos danos à produção tanto em cultivo protegido quanto em campo aberto. O objetivo deste trabalho foi avaliar a reação de dez genótipos de pepino a C. cassiicola. O experimento foi conduzido em casa de vegetação em delineamento inteiramente ao acaso com dez tratamentos e dez repetições. Foram avaliados a cultivar Aodai e os híbridos tipo japonês General Lee F1, Hokushim, Japonês Híbrido F1 (Soudai), Jóia, Marketmore 76 (verde comprido), Natsubayashi, Natsu Suzumi, Sprint 440 II e Tsuyataro. Os isolados do patógeno foram obtidos a partir de folhas de plantas de pepino naturalmente infectadas, e multiplicado em meio de cultura BDA. As plantas foram inoculadas nas duas primeiras folhas definitivas com suspensão de inóculo na concentração 104 conídios mL-1 e mantidas em câmara úmida por 48 horas. A severidade foi avaliada e calculada a área abaixo da curva de progresso da doença (AACPD) para cada tratamento. Houve variação na reação dos genótipos ao patógeno e a cultivar Aodai apresentou menor AACPD considerada como resistente. Os híbridos Hokuchim, Soudai, Verde Comprido e Natsubayashi apresentaram menores valores de AACPD entre os híbridos avaliados em contraposição ao Tsuyataro o mais suscetível.
Foi realizada uma reavaliação morfológica de estruturas reprodutivas e de infecção do agente causal da antracnose do guaranazeiro (Paullinia cupana var. sorbilis), visando confirmar a identidade do patógeno. Os resultados reforçam o reconhecimento da espécie Colletotrichum guaranicola Albuq., como um novo taxon dentro do gênero Colletotrichum.
This study investigated the effect of ozone on papaya postharvest conservation, aiming at controlling anthracnose and increasing fruit shelf life. The experiment was conducted at the Laboratory of Product Technology, Federal University of Amazonas, Manaus-AM. Fruits were treated with 0; 0.6; 1.5 and 3.3 ppm ozone and commercial fungicide. They were then stored at room temperature (27 ± 2 °C and 65 ± 5% RH). Incidence, severity, number and size of anthracnose lesions were determined, as well as postharvest quality by assessing shelf life; accumulated and daily fresh mass loss (%), peel color, electrolyte leakage (%), soluble solids (%), titratable acidity (g 100 g-1), soluble solids / titratable acidity ratio, pH and vitamin C (mg 100 g-1). Ozone reduced anthracnose severity in the short (3.3 ppm) and long term (1.5 ppm), making it as effective as a commercial fungicide. Ozone increased shelf life by seven days and maintained the postharvest quality of papaya fruits. Thus, ozone becomes a potential technology for postharvest use as a safe alternative to the use of pesticides in reducing anthracnose and increasing shelf life of papaya fruits.
Sacha inchi (Plukenetia volubilis L.) is a species native to the Amazon region for which studies of genetic diversity are required to ensure the success in programs to breed cultivars for agriculture. The purpose of this study was to assess the genetic diversity among sacha inchi accessions using amplified fragment length polymorphism (AFLP) markers. Thirty-seven accessions of the sacha inchi germplasm bank of Embrapa Amazônia Ocidental were analyzed. The markers were identified with four primer combinations. The percentage of polymorphic loci was estimated and the similarity of accessions analyzed by calculating the arithmetic complement of the Jaccard coefficient and cluster analysis. The primers revealed 191 polymorphic loci. The parameters of the similarity values in class intervals were on average 0.739 and variance 0.01. The estimates of the number and percentage of the genetic distance values per class interval were highest in the range of 0.7 to 0.79, (248 and 37.24%, respectively); the highest percentage, 69.22%, corresponded to values of genetic distance above 0.7, showing mostly high genetic diversity among the accessions. The results indicated a geographic structure among accessions in relation to their origin, generating consistent and valuable data for breeding programs and species conservation.
