Ocular retardation (or) is a murine eye mutation causing microphthalmia, a thin hypocellular retina and optic nerve aplasia. Here we show that mice carrying the OrJ allele have a premature stop codon in the homeobox of the Chx10 gene, a gene expressed at high levels in uncommitted retinal progenitor cells and mature bipolar cells. No CHX10 protein was detectable in the retinal neuroepithelium of orJ homozygotes. The loss of CHX10 leads both to reduced proliferation of retinal progenitors and to a specific absence of differentiated bipolar cells. Other major retinal cell types were present and correctly positioned in the mutant retina, although rod outer segments were short and retinal lamination was incomplete. These results indicate that Chx10 is an essential component in the network of genes required for the development of the mammalian eye, with profound effects on retinal progenitor proliferation and bipolar cell specification or differentiation. off
Bordetellae, pathogenic to mammals, produce an immunomodulatory adenylate cyclase toxin–hemolysin (CyaA, ACT or AC-Hly) that enables them to overcome the innate immune defense of the host. CyaA subverts host phagocytic cells by an orchestrated action of its functional domains, where an extremely catalytically active adenylyl cyclase enzyme is delivered into phagocyte cytosol by a pore-forming repeat-in-toxin (RTX) cytolysin moiety. By targeting sentinel cells expressing the complement receptor 3, known as the CD11b/CD18 (αMβ2) integrin, CyaA compromises the bactericidal functions of host phagocytes and supports infection of host airways by Bordetellae. Here, we review the state of knowledge on structural and functional aspects of CyaA toxin action, placing particular emphasis on signaling mechanisms by which the toxin-produced 3′,5′-cyclic adenosine monophosphate (cAMP) subverts the physiology of phagocytic cells.
2007: A city in motion: Time-space activity and mobility patterns of suburban inhabitants and the structuration of the spatial organization of the Prague metropolitan area. Geogr. Ann., 89 B (2): 147-167.ABSTRACT. This contribution attempts to reveal the relations between new suburban areas and other parts of the Prague metropolitan area by investigating the time-space activity and mobility patterns of the inhabitants of newly built suburban districts. The focus on some aspects of the everyday life of people in new suburbs helps us to identify the impact of suburbanization on the changing geography of the metropolitan region and to better understand how the spatial organization of the Prague metropolitan area is produced, reproduced and transformed. We use several interrelated concepts, which serve the theoretical foundation of our work, namely time geography, structuration theory and the postcommunist city. The empirical data utilized are primarily based on 262 diaries completed by eighty-eight individuals from thirtyeight households, accompanied by household questionnaires and interviews with the heads of households. The research confirmed the implicit, generally unspoken view that new suburbs in the Prague metropolitan region are heavily dependent on the core of the metropolitan area for the provision of jobs and services. However, newly built suburban shopping facilities to some extent disrupt this pattern, keeping some daily activities of inhabitants within the suburban zone. In addition to empirical observations, the key purpose of this contribution has been to discuss and apply time geography concepts and methods to the research of urban restructuring, and to understand the structuration of metropolitan spatial organization.
To avoid organ dysfunction as a consequence of tissue diminution or tumorous growth, a tight balance between cell proliferation and differentiation is maintained in metazoans. However, cell-intrinsic gene expression mechanisms controlling adult tissue homeostasis remain poorly understood. By focusing on the adult Caenorhabditis elegans reproductive tissue, we show that translational activation of mRNAs is a fundamental mechanism to maintain tissue homeostasis. Our genetic experiments identified the Trf4/5-type cytoplasmic poly(A) polymerase (cytoPAP) GLD-4 and its enzymatic activator GLS-1 to perform a dual role in regulating the size of the proliferative zone. Consistent with a ubiquitous expression of GLD-4 cytoPAP in proliferative germ cells, its genetic activity is required to maintain a robust proliferative adult germ cell pool, presumably by regulating many mRNA targets encoding proliferation-promoting factors. Based on translational reporters and endogenous protein expression analyses, we found that gld-4 activity promotes GLP-1/Notch receptor expression, an essential factor of continued germ cell proliferation. RNA-protein interaction assays documented also a physical association of the GLD-4/GLS-1 cytoPAP complex with glp-1 mRNA, and ribosomal fractionation studies established that GLD-4 cytoPAP activity facilitates translational efficiency of glp-1 mRNA. Moreover, we found that in proliferative cells the differentiation-promoting factor, GLD-2 cytoPAP, is translationally repressed by the stem cell factor and PUF-type RNA-binding protein, FBF. This suggests that cytoPAP-mediated translational activation of proliferation-promoting factors, paired with PUF-mediated translational repression of differentiation factors, forms a translational control circuit that expands the proliferative germ cell pool. Our additional genetic experiments uncovered that the GLD-4/GLS-1 cytoPAP complex promotes also differentiation, forming a redundant translational circuit with GLD-2 cytoPAP and the translational repressor GLD-1 to restrict proliferation. Together with previous findings, our combined data reveals two interconnected translational activation/repression circuitries of broadly conserved RNA regulators that maintain the balance between adult germ cell proliferation and differentiation.
The main purpose of this paper is to identify ongoing changes in post-socialist large housing estates and to clarify their main factors, using a case study approach. The paper examines the transformations of nine housing estates in the third largest Polish city -Łódź. The transformations were analysed in two dimensions: social (socio-demographic changes) and physical (changes in the spatial and functional structure and changes in the physiognomy). The key question was whether the changes occurring in large housing estates in Łódź protect them before the development of the social and physical degradation, identified in many estates in Western Europe, referred as the "large housing estate syndrome". Research has shown that in examined housing estates there are no symptoms of physical or social degradation. Moreover, processes found there contribute to the prevention of the negative phenomena leading to the development of the "large housing estate syndrome".
The complexity of head and neck cancers (HNC) mandates a multidisciplinary approach and radiation therapy (RT) plays a critical role in the optimal management of patients with HNC, either as frontline or adjuvant treatment postoperatively. The advent of both definitive and post-operative RT has significantly improved the outcomes of patients with HNC. Herein, we discuss the role of postoperative RT in different subtypes of HNC, its side effects, and the importance of surveillance. The treatment regions discussed in this paper are the oral cavity, nasopharynx, paranasal sinus cavity, oropharynx, larynx and hypopharynx. Multiple studies that demonstrate the importance of definitive and/or postoperative RT, which led to an improved outlook of survival for HNC patients will be discussed.
