Background and Objectives. Chronic obstructive pulmonary disease (COPD) is a complex disease characterized by airflow limitation. Although airway inflammation and oxidative stress are known to be important in the pathogenesis of COPD, the mechanism underlying airflow obstruction is not fully understood. Gene expression profiling of lung tissue was performed to define the molecular pathways that are dysregulated in COPD. Methods. RNA was isolated from lung tissues obtained from 98 subjects with COPD and 91 control subjects with normal spirometry. The RNA samples were processed with RNA-seq using the HiSeq 2000 system. Genes expressed differentially between the two groups were identified using Student's t-test. Results. After filtering for genes with zero counts and noncoding genes, 16,676 genes were evaluated. A total of 2312 genes were differentially expressed between the lung tissues of COPD and control subjects (false discovery rate corrected q < 0.01). The expression of genes related to oxidative phosphorylation and protein catabolism was reduced and genes related to chromatin modification were dysregulated in lung tissues of COPD subjects. Conclusions. Oxidative phosphorylation, protein degradation, and chromatin modification were the most dysregulated pathways in the lung tissues of COPD subjects. These findings may have clinical and mechanistic implications in COPD.
Double proton transfers in formic acid dimer and formamidine dimer were studied as prototypes of multiple proton transfer. The potential energy surface (PES) for the double proton transfer was studied using ab initio quantum mechanical methods. The solvent effect on the PES was also included using the Onsager self-consistent reaction field model. In the gas phase, the transition state for the double proton transfer in the formic acid dimer complex has D 2 h symmetry, but in water it is changed to a C 2 v structure, when the Hartree−Fock (HF) level of theory is used. When the density functional theory is used, the transition state has D 2 h symmetry with and without solvent. However the barrier height depends very much on the electron correlation. The double proton transfer occurs synchronously in all the cases. For the formamidine dimer complex, the transition state has C 2 v symmetry in the gas phase, and it changes to C s symmetry in water at the HF level of theory. The C 2 v structure becomes an intermediate in water, which means that double proton transfer occurs asynchronously. In the density functional theory for the gas phase, the transition state has D 2 h symmetry, and it changes to C 2 v structure in solution. However the double proton transfer occurs synchronously in both cases. These results suggest that the correlation is very important to the PES for double proton transfer, not only in the gas phase but also in solution.
Abstract. The Geostationary Ocean Color Imager (GOCI) onboard the Communication, Ocean, and Meteorological Satellite (COMS) is the first multi-channel ocean color imager in geostationary orbit. Hourly GOCI top-of-atmosphere radiance has been available for the retrieval of aerosol optical properties over East Asia since March 2011. This study presents improvements made to the GOCI Yonsei Aerosol Retrieval (YAER) algorithm together with validation results during the Distributed Regional Aerosol Gridded Observation Networks -Northeast Asia 2012 campaign (DRAGON-NE Asia 2012 campaign). The evaluation during the spring season over East Asia is important because of high aerosol concentrations and diverse types of Asian dust and haze. Optical properties of aerosol are retrieved from the GOCI YAER algorithm including aerosol optical depth (AOD) at 550 nm, fine-mode fraction (FMF) at 550 nm, single-scattering albedo (SSA) at 440 nm, Ångström exponent (AE) between 440 and 860 nm, and aerosol type. The aerosol models are created based on a global analysis of the Aerosol Robotic Networks (AERONET) inversion data, and covers a broad range of size distribution and absorptivity, including nonspherical dust properties. The Cox-Munk ocean bidirectional reflectance distribution function (BRDF) model is used over ocean, and an improved minimum reflectance technique is used over land. Because turbid water is persistent over the Yellow Sea, the land algorithm is used for such cases. The aerosol products are evaluated against AERONET observations and MODIS Collection 6 aerosol products retrieved from Dark Target (DT) and Deep Blue (DB) algorithms during the DRAGON-NE Asia 2012 campaign conducted from March to May 2012. Comparison of AOD from GOCI and AERONET resulted in a Pearson correlation coefficient of 0.881 and a linear regression equation with GOCI AOD = 1.083 × AERONET AOD − 0.042. The correlation between GOCI and MODIS AODs is higher over ocean than land. GOCI AOD shows better agreement with MODIS DB than MODIS DT. The other GOCI YAER products (AE, FMF, and SSA) show lower correlation with AERONET than AOD, but still show some skills for qualitative use.
Percutaneous endoscopic transforaminal lumbar discectomy (PETLD) has now become a standard of care for the management of lumbar disc disease. There are two techniques for the introduction of a working cannula with respect to disc-outside-in and inside-out. The aim of this prospective study is to describe the technical aspects of a novel mobile outside-in method in dealing with different types of disc prolapse. A total of 184 consecutive patients with unilateral lower limb radiculopathy due to lumbar disc prolapse were operated on with the mobile outside-in technique of PETLD. Their clinical outcomes were evaluated based on the type of disc prolapse they had, a visual analog scale (VAS) leg pain score, the Oswestry Disability Index (ODI), and the Macnab criteria. The completeness of the decompression was documented with a postoperative magnetic resonance imaging. The mean age of the patients was 50 ± 16 years and the male/female ratio was 2:1. The mean follow-up was 19 ± 6 months. A total of 190 lumbar levels were operated on (L1-L2: n = 4, L2-L3: n = 17, L3-L4: n = 27, L4-5: n = 123, and L5-S1: n = 19). Divided into types, the patient distribution was central: n = 14, paracentral: n = 74, foraminal: n = 28, far lateral: n = 13, superior-migrated: n = 8, inferior migrated: n = 38, and high canal compromise: n = 9. The mean operative time was 35 ± 12 (25 - 56) min and the mean hospital stay was 1.2 ± 0.5 (1-3) days. The VAS score for leg pain improved from 7.5 ± 1 to 1.7 ± 0.9. The ODI improved from 70 ± 8.3 to 23 ± 5. According to the Macnab criteria, 75 patients (40.8%) had excellent results, 104 patients (56.5%) had good results, and 5 patients (2.7%) had fair results. Recurrence (including early and late) was seen in 15 out of the 190 levels that were operated on (7.89%). This article presents a novel outside-in approach that relies on a precise landing within the foramen in a mobile manner and does not solely depend upon the enlargement of the foramen. It is more versatile in application and useful in the management of all types of disc prolapse, even in severe canal compromise and high migration.
In recent years, indoor localization systems have been the object of significant research activity and of growing interest for their great expected social impact and their impressive business potential. Application areas include tracking and navigation, activity monitoring, personalized advertising, Active and Assisted Living (AAL), traceability, Internet of Things (IoT) networks, and Home-land Security. In spite of the numerous research advances and the great industrial interest, no canned solutions have yet been defined. The diversity and heterogeneity of applications, scenarios, sensor and user requirements, make it difficult to create uniform solutions. From that diverse reality, a main problem is derived that consists in the lack of a consensus both in terms of the metrics and the procedures used to measure the performance of the different indoor localization and navigation proposals. This paper introduces the general lines of the EvAAL benchmarking framework, which is aimed at a fair comparison of indoor positioning systems through a challenging competition under complex, realistic conditions. To evaluate the framework capabilities, we show how it was used in the 2016 Indoor Positioning and Indoor Navigation (IPIN) Competition. The 2016 IPIN competition considered three different scenario dimensions, with a variety of use cases: (1) pedestrian versus robotic navigation, (2) smartphones versus custom hardware usage and (3) real-time positioning versus off-line post-processing. A total of four competition tracks were evaluated under the same EvAAL benchmark framework in order to validate its potential to become a standard for evaluating indoor localization solutions. The experience gained during the competition and feedback from track organizers and competitors showed that the EvAAL framework is flexible enough to successfully fit the very different tracks and appears adequate to compare indoor positioning systems.
Poly- N -isopropyl acrylamide (PNIPAM) nanogels have been modified with different acrylic acid (AAc) contents for the efficient control of lower critical solution temperature (LCST). In this study, PNIPAM-co-AAc nanogels nanogels showed two volume phase transitions in comparison with PNIPAM. The transition temperature of PNIPAM nanogels was increased with AAc contents. The controlled drug release performance of PNIPAM-co-AAc nanogels loaded with β-lapachone was attributed to the AAc content ratio and was efficiently triggered in response to temperature and pH. Moreover, a colorimetric cell proliferation assay and direct fluorescence-based live/dead staining were used to confirm the concurrence on drug release profiles. Finally, PNIPAM-co-AAc20 showed a relatively low level of drug release in the range of acidic to neutral pH at body temperature, while maximizing drug release at basic pH. Therefore, we demonstrated that the PNIPAM-based nanogel with the temperature- and pH-responsive features could be a promising nanocarrier for potential intestine-specific drug delivery.
