Summary: A method of acidic extraction at pH‐1.3 was used for quantitative estimation of the proacrosin and acrosin content in boar spermatozoa after various times of storage in four different sperm dilutors. The proacrosin and acrosin contents were correlated with the results of biological tests for sperm penetration in zona‐free hamster eggs and sperm survival. The results have shown that proacrosin quantitation should be a convenient biochemical test of sperm fertilizing capacity.

Die Methode der Säureextraktion bei pH‐1.3 wurde für die quantitative Bestimmung der Proakrosin‐ und Akrosin‐Mengen in Eberspermien nach verschiedenen Konservierungszeiten in vier Sperma‐Dilutoren verwendet. Die Proakrosin‐ und Akrosin‐Mengen wurden mit den Ergebnissen der Spermatozoen‐Penetrationstests in Zona‐freien Hamstereiern und des Spermatozoen‐Überlebens verglichen. Die Resultate zeigen, daß die beschriebene Methode der Proakrosin‐Bestimmung ein geeigneter biochemischer Ergänzungstest für die Bestimmung der Spermien‐Befruchtungskapazität sein könnte.
Two partially purified proacrosin forms have been obtained from acid extracts of r a m ejaculated spermatozoa by Sephadex G-100 column chromatography in 0.001 M HCl. The form obtained from the extracts of whole ejaculates with apparently higher molecular mass was free of any acrosin inhibitor and was used for autoactivation studies. The autoactivation followed a classical S-shaped activation curve and was a twostep process. The presence of calcium ions slowed down the autoactivation process and stabilized the active acrosin formed. The activity of latter rapidly decreased when the pH of the fully activated mixture was adjusted to pH 3. The activity was not restored by readjusting the pH to 8. A proacrosin with apparently lower molecular mass was isolated from the extract of washed spermatozoa. Both proacrosins, however, showed the same molecular mass Mr -58 000 daltons, when examined by gel filtration in 0.1 M NaCl (pH 3).

Eine einfache Methode zur lsolierung von Inhibitorenfreiem Proakrosin und Proakrosin AutoaktivationStudienZusammenfassung: Von Saure-Extrakten der aus Schafbock ejakulierten Spermatozoa wurde durch Gelfiitrierung an Sephadex G-100 Saulen in 0.001 M HCl Proakrosin isoliert. Aus dem Extrakt des vollen Ejakulates wurde ein Priiparat isoliert mit einer scheinbar hoheren Molekularmasse, welches keine Akrosin-Inhibitoren enthielt und zu Autoaktivationsstudien verwendet wurde. Die Autoaktivation zeigte eine klassische Aktivationskurve in S-Form und zeigte einen zweistufigen ProzeB an. Die Anwesenheit von Calcium-Ionen verlanpmt den Autoaktivations-ProzeB und stabilisiert das gebildete Akrosin. Die Aktivitat des Akrosins vermindert sich, wenn das pH des voll aktivierten Reaktionsgemischs auf pH 3 adjustiert wird. Die Aktivitat erneuert sich nicht nach der Readjustierung des Gemisches auf pH 8. Aus dem Extrakt der gewaschenen Spermatozoa w d e ein Roakrosinpriiparat mit scheinbar niedrigerer Molekularmasse isoliert . Beide unter verschiedenen Bedingungen isolierte Akrosinpdparate zeigten jedoch bei der Gelfdtrierung in 0.1 M NaCl (pH 3) eine identische Molekularmasse M, -58 000.
