-Three experiments were conducted to evaluate factors affecting number of surface ovarian follicles and oocytes yield and quality in buffalo. In Experiment 1, ovaries (n = 126) were collected in pairs from slaughtered anoestrus, early pregnant and cyclic buffaloes. Ovarian follicles (1-3, 4-9 and ≥ 10 mm diameter) were counted, aspirated and oocytes were recovered and evaluated. In Experiment 2, ovaries were divided into 2 groups. Group 1, ovaries bearing a CL (n = 74) and Group 2 non-bearing CL (n = 74), ovarian follicles (2-8 mm) were counted, aspirated and oocytes evaluated. In Experiment 3, oocytes were recovered using aspiration or slicing methods. In all experiments, oocytes were classified into good, fair, poor and denuded. Results showed that the development of small and total ovarian follicles are continuous and independent in early pregnant or cyclic buffalo cows, however, it significantly decreased (P < 0.01) in the ovaries of anoestrus buffaloes. Number of medium and large size follicles was significantly increased (P < 0.01) in cyclic buffaloes on Days 10-16 and 17-22 of oestrous cycle, while large follicles was significantly decreased (P < 0.01) in the ovaries of pregnant buffaloes. A significantly higher (P < 0.01) percentage of poor and denuded oocytes were recovered from ovaries of anoestrus and pregnant buffalo. While, the highest (P < 0.01) percentage of good quality oocytes were recovered from ovaries of cyclic buffaloes on Days 1-3 and 10-16 of oestrous cycle, eliciting that the stage of oestrous cycle is affecting the quality of buffalo oocytes. In addition, the presence of a CL stimulates the development of a significantly higher (P < 0.01) number ovarian follicles which produced a significantly higher (P < 0.05) number of good quality oocytes. Slicing of buffalo ovaries produced a significantly higher number of fair, poor and denuded oocytes. 232The application of biotechnology in cattle breeding has allowed us to reduce the generation interval and to increase selection pressure in animal populations on the basis of more objective evaluation of the genotype. It facilitates more effective utilization of the reproductive potential of animals, makes a broader range of parental combinations available and thus allows for modification of the animal population in a desired way.The prerequisite for achieving these goals involves efficient cryopreservation methods for long-term storage of embryos, exchange of genetic material and creation of genetic resources. Today the embryos obtained by superovulation can be fro- ABSTRACT: The present study was designed to compare the efficiency of bovine embryo production for cryopreservation between oocytes collected from donors in the growth phase of follicular development (GPFD) and those recovered from donors in the undefined phase (UPFD). Cyclic cows, Czech Siemental or Holstein dairy breeds, 4-6 years of age, slaughtered at the local abbatoir were used. They were divided into two groups based on ovarian morphology: I. GPFD donors with ovaries corresponding to the growth phase of the first follicular wave (estrus cycle days 3-4; n = 52), and II. UPFD donors with ovaries in any other phase of follicular development (undefined estrus cycle days; n = 89). A total of 3 771 oocytes were collected and 1 134 embryos were prepared as two separate populations by standard protocol. In total 352 excellent or good quality embryos at the early, advanced or expanded blastocyst stage from both donor groups were pooled and used for cryotolerance assessment. They were frozen on day 7 (D7) or day 8 (D8) after fertilization by the standard procedure. After thawing, the embryos were cultured for 72 h to the hatching stage. The percentages of both D7 embryos and advanced blastocysts were significantly higher (P ≤ 0.01) for oocytes collected from GPFD donors than for oocytes collected from UPFD donors (33.7 vs 28.6% and 43.0 vs 29.5%, respectively). The percentages of excellent or good quality embryos obtained from both D7 embryos and fertilized oocytes were significantly higher (P ≤ 0.01) for oocytes collected from GPFD donors than for oocytes collected from UPFD donors (63.6 vs 49.4% and 21.4 vs 14.1%, respectively). The post-thaw survival rates were significantly higher (P ≤ 0.01) for D7 than D8 embryos (80.4 vs 66.3%). In relation to the developmental stage, the development and hatching rates were significantly higher (P ≤ 0.01) for D7 than D8 early blastocysts (75.0 vs 41.2% and 50.0 vs 5.9%, respectively) and for D7 than D8 advanced blastocysts, (73.7 vs 57.1 and 52.6 vs 28.6%, respectively). No differences were found between D7 and D8 expanded blastocysts after freezing-thawing. Resumo: As biotécnicas da reprodução são importantes ferramentas para conservação de espécies domésticas e silvestres, pois permitem a recuperação e uso futuro de material reprodutivo. O objetivo deste estudo foi avaliar parâmetros morfológicos de CCOs de cutias, obtidos pela técnica de fatiamento do ovário para utilização em protocolos de maturação e fecundação na produção in vitro de embriões. Foram utilizadas dezessete cutias fêmeas do NEPAS, CCA- UFPI, com idade e peso médios de 3,9 anos e 2,16Kg, respectivamente, que foram submetidas à ovariossalpingohisterectomia. Os ovários após dissecados e pesados em balança de precisão, foram fatiados individualmente. Procedeu-se a busca e seleção dos CCOs em estereomicroscópio, os quais foram identificados e quantificados por cada ovário, além de classificados quanto a sua morfologia segundo a quantidade de camadas de células do cumulus e ao citoplasma em quatro graus. Verificou-se que a técnica de fatiamento do ovário possibilitou a obtenção de CCOs de cutias, com recuperação de grande quantidade e de variados graus de qualidade. Não houve correlações entre a idade dos animais e o peso dos ovários; a idade e o número de CCOs obtidos; entre o peso das cutias e o peso dos ovários e o número de CCOs obtidos.
