Adaptation an und Resistenz gegen Antimetaboliten l>zw. Chemotherapeutika bei den pathogenen Mikroorganismen 1 gehören mit zu den seit mehr als zwei Jahrzehnten intensiv bearbeiteten, doch auch heute noch nicht gelösten Problemen der Biochemie. Die Erforschung der diesen Verhaltensweisen zugrunde liegenden Stoffwechselprozesse koinzidiert in vielen Punkten mit den für die Kausaltherapie bakterieller Erkrankungen und maligner Tumoren gleich bedeutungsvollen Fragen der Aufklärung von Stoffwechselhaupt-und -nebenwegen in Mikroorganismen und entarteten Geweben sowie der Wirkungsweise und -orte von Antimetaboliten im intermediären Stoffwechsel.Zur Bearbeitung solcher Fragen bediente man sich der pathogenen Mikroorganismen selbst; man verwendete aber auch einfacher zu handhabende "Modell-Lebewesen", wobei man in Kauf nehmen mußte, daß Ergebnisse mit diesen nicht ohne nähere Prüfung für jene gelten.Neurospora crassa ist ein in seiner Genetik und seinen Stoffwechselvorgängen bekanntes und experimentell gut zu verwendendes Lebewesen. Es reagiert empfindlich auf die Gegenwart von Antimetaboliten. Da es mit vielen Ascomyceten die Eigenschaft teilt, auf der Oberfläche eines geeigneten Agarnährmediums horizontal zu wachsen, ließ sich ein Horizontalwuchstest ausarbeiten, der, obgleich 1943 beschrieben 2 , bis heute zur Hemmungsanalyse wenig und dann nur in qualitativer Beziehung verwendet wurde 3 . Die Vorzüge von N. crassa als biologischem Testobjekt und des Horizontalwuchstestes als Arbeitsmethode veranlaßten uns, damit eine Reihe neuerer Antimetaboliten und Cytostatika zu prüfen. Wir beobachteten hierbei mehrere Verlaufsformen der Hemmung * Ausgeführt mit Mitteln der Deutschen Forschungsgemeinschaft und der Gesellschaft zur Förderung der Westfälischen Wilhelms-TJniversität Münster Bd. 315 (1959) Horizontalwuchstest mit Neurospora crassa 47 äes Horizontalwuchses von N. crassa und teilten sie in Gruppen typischer Hemmungsformen ein. In der vorliegenden Arbeit werden diese Verlaufsformen sowie das Verfahren zu ihrer quantitativen Auswertung beschrieben. Allgemeines zum Wuchstest mit N. crassa N. crassa wächst apikal, d. h. nur von den Mycelspitzen aus 4 . Einmal gebildete Zellen wachsen nicht mehr in der Längsachse, sondern nur noch geringfügig in der Querrichtung und verdicken ihre Chitinwände. Die Zellen sind nicht völlig gegeneinander abgeschlossen und die Trennwände besitzen Foramina, durch die nicht nur agranuläre Zellsubstanzen, sondern auch Zellkerne ausgetauscht werden, von denen jede Zelle viele enthält. Das in einem Horizontalrohr nach Ryan, Beadle und Tat um 2 eingeschlossene, darin vorwärts wachsende Mycel ist somit als ein einheitlicher Organismus anzusehen. Die in das Testrohr vordringenden Mycelspitzen treffen stets auf die gleichen Konzentrationen an Metaboliten und Antimetaboliten im Nährmedium. Die Verhaltensweise dieses Organismus während des Horizontalwuchses ist also die Antwort auf eine über Wachstumsstrecke und Versuchszeit konstante Dosis der Wirksubstanz.
