The binding of two 5-substituted-l,3,4-thiadiazole-2-thione inhibitors to the matrix metalloproteinase stromelysin (MMP-3) have been characterized by protein crystallography. Both inhibitors coordinate to the catalytic zinc cation via an exocyclic sulfur and lay in an unusual position across the unprimed (Pl-P3) side of the proteinase active site. Nitrogen atoms in the thiadiazole moiety make specific hydrogen bond interactions with enzyme structural elements that are conserved across all enzymes in the matrix metalloproteinase class. Strong hydrophobic interactions between the inhibitors and the side chain of tyrosine-I55 appear to be responsible for the very high selectivity of these inhibitors for stromelysin. In these enzymehnhibitor complexes, the S 1 ' enzyme subsite is unoccupied. A conformational rearrangement of the catalytic domain occurs that reveals an inherent flexibility of the substrate binding region leading to speculation about a possible mechanism for modulation of stromelysin activity and selectivity.
Overexpression of the D2 dopamine receptor has been proposed to be part of the pathology of schizophrenia. The isolation of a D2 dopamine receptor clone has assisted the molecular characterization of D2 receptor. We have now isolated an identical rat clone along with two other clones--a second related rat clone (RD-2in) and a homologous bovine clone (BD-2in), both of which contain an insert encoding an additional 29 amino acids relative to the original rat clone (RD-2o). All three clones encode D2 receptor binding sites when expressed in COS-7 cells. The amino-acid insert encoded by D-2in lies in the domain of the receptor believed to interact with the GTP-binding proteins (G proteins) of various signal transduction pathways. By using oligonucleotide probes specific for either D-2o or D-2in RNA transcripts, we have found that the level of expression of the D-2in-encoded form of the receptor is seven times that of the D-2o form in the caudate nucleus, the richest brain source of D2 receptors.
The synthesis and biological activity of a series of seco-oxysterol analogs designed to be inhibitors of transcription of the gene for 3-hydroxy-3-methylglutaryl-Coenzyme A reductase (HMGR) are described. The compound possessing the most significant activity, [1 alpha (E),4 beta]-3-[2-(4- hydroxy-1-methylcyclohexyl)ethenyl]-alpha,alpha-dimethylbenzenepentan ol (4, U-88156), inhibited (IC50 = 10 microM) the expression of beta-galactosidase (beta-gal) in a transfected human HepG2 cell line wherein the beta-gal gene was driven by a 5 kB segment of the promoter for hamster HMGR. Furthermore, using wild-type HepG2 cells, it was shown that 10 microM 4 reduced HMGR mRNA levels by 73% while stimulating LDL-receptor activity by 47%. In the same system, the related oxysterol, 25-hydroxycholesterol (1), at 10 microM lowered both HMGR mRNA levels and LDL-receptor activity by 58% and 64%, respectively. Overall HMGR activity in wild-type HepG2 cells was inhibited 30% by 4 at 10 microM. These findings collectively demonstrate that a seco-oxysterol analog is capable of regulating HMGR gene expression and that this regulation can occur without a concomitant attenuation of the level of LDL-receptor activity.

Adaptation an und Resistenz gegen Antimetaboliten l>zw. Chemotherapeutika bei den pathogenen Mikroorganismen 1 gehören mit zu den seit mehr als zwei Jahrzehnten intensiv bearbeiteten, doch auch heute noch nicht gelösten Problemen der Biochemie. Die Erforschung der diesen Verhaltensweisen zugrunde liegenden Stoffwechselprozesse koinzidiert in vielen Punkten mit den für die Kausaltherapie bakterieller Erkrankungen und maligner Tumoren gleich bedeutungsvollen Fragen der Aufklärung von Stoffwechselhaupt-und -nebenwegen in Mikroorganismen und entarteten Geweben sowie der Wirkungsweise und -orte von Antimetaboliten im intermediären Stoffwechsel.Zur Bearbeitung solcher Fragen bediente man sich der pathogenen Mikroorganismen selbst; man verwendete aber auch einfacher zu handhabende "Modell-Lebewesen", wobei man in Kauf nehmen mußte, daß Ergebnisse mit diesen nicht ohne nähere Prüfung für jene gelten.Neurospora crassa ist ein in seiner Genetik und seinen Stoffwechselvorgängen bekanntes und experimentell gut zu verwendendes Lebewesen. Es reagiert empfindlich auf die Gegenwart von Antimetaboliten. Da es mit vielen Ascomyceten die Eigenschaft teilt, auf der Oberfläche eines geeigneten Agarnährmediums horizontal zu wachsen, ließ sich ein Horizontalwuchstest ausarbeiten, der, obgleich 1943 beschrieben 2 , bis heute zur Hemmungsanalyse wenig und dann nur in qualitativer Beziehung verwendet wurde 3 . Die Vorzüge von N. crassa als biologischem Testobjekt und des Horizontalwuchstestes als Arbeitsmethode veranlaßten uns, damit eine Reihe neuerer Antimetaboliten und Cytostatika zu prüfen. Wir beobachteten hierbei mehrere Verlaufsformen der Hemmung * Ausgeführt mit Mitteln der Deutschen Forschungsgemeinschaft und der Gesellschaft zur Förderung der Westfälischen Wilhelms-TJniversität Münster Bd. 315 (1959) Horizontalwuchstest mit Neurospora crassa 47 äes Horizontalwuchses von N. crassa und teilten sie in Gruppen typischer Hemmungsformen ein. In der vorliegenden Arbeit werden diese Verlaufsformen sowie das Verfahren zu ihrer quantitativen Auswertung beschrieben. Allgemeines zum Wuchstest mit N. crassa N. crassa wächst apikal, d. h. nur von den Mycelspitzen aus 4 . Einmal gebildete Zellen wachsen nicht mehr in der Längsachse, sondern nur noch geringfügig in der Querrichtung und verdicken ihre Chitinwände. Die Zellen sind nicht völlig gegeneinander abgeschlossen und die Trennwände besitzen Foramina, durch die nicht nur agranuläre Zellsubstanzen, sondern auch Zellkerne ausgetauscht werden, von denen jede Zelle viele enthält. Das in einem Horizontalrohr nach Ryan, Beadle und Tat um 2 eingeschlossene, darin vorwärts wachsende Mycel ist somit als ein einheitlicher Organismus anzusehen. Die in das Testrohr vordringenden Mycelspitzen treffen stets auf die gleichen Konzentrationen an Metaboliten und Antimetaboliten im Nährmedium. Die Verhaltensweise dieses Organismus während des Horizontalwuchses ist also die Antwort auf eine über Wachstumsstrecke und Versuchszeit konstante Dosis der Wirksubstanz.
