Biological control of agricultural pests has gained importance in recent years due to increased pressure to reduce the use of agrochemicals and their residues in the environment and food. Viruses of a few families are known to infect insects but only those belonging to the highly specialized family Baculoviridae have been used as biopesticides. They are safe to people and wildlife, their specificity is very narrow. Their application as bioinsecticides was limited until recently because of their slow killing action and technical difficulties for in vitro commercial production. Two approaches for the wider application of baculoviruses as biopesticides will be implemented in future. In countries where use of genetically modified organisms is restricted, the improvements will be mainly at the level of diagnostics, in vitro production and changes in biopesticide formulations. In the second approach, the killing activity of baculoviruses may be augmented by genetic modifications of the baculovirus genome with genes of another natural pathogen. It is expected that the baculoviruses improved by genetic modifications will be gradually introduced in countries which have fewer concerns towards genetically modified organisms.

Current problems of forest protection concern the declining health of forest stands due to climate change and the resulting extreme weather events such as heat waves, droughts, hurricane winds, heavy rainfalls and floods. Repeated impacts of these factors increase susceptibility of forest stands to pest insects and fungal pathogens. Norway spruce (Picea abies [L.] H. Karst) is sensitive to high air temperatures and water shortage. Long lasting droughts during the last two decades, have been one of the reasons behind Norway spruce dieback due to severe outbreak of European spruce bark beetle Ips typographus (L.) in the mountainous regions of southern Poland. In the Scots pine (Pinus sylvestris L.) stands, water balance disorders have enhanced the colonization of weekend trees by steelblue jewel beetle Phaenops cyanea (F.) and engraver beetle Ips acuminatus (Gyll.), as well as contributed to the spread of fungal diseases caused by Gremmeniella abietina (Lagerb.) Cenangium ferruginosum Fr. and Sphaeropsis sapinea Fr. fungi. Water related stress leads to weakening of oak stands, which are attacked by Agrilus spp. beetles and pathogens from the genus Phytophthora. It is possible that long lasting droughts initiated the spread of infectious ash disease caused by Hymenoscyphus fraxineus (= Chalara fraxinea), which resulted in the epidemic of ash dieback throughout Europe. Until recently, the use of plant protection products was the most common method of forest protection against pest insects and pathogens. Poland’s accession to the European Union has affected the marketing and use of plant protection products in the country. The implementation of the EU legislation (Directive 91/414/EEC, Directive 2009/128/ EC and Regulation No 1107/2009) has resulted in decreased assortment of pesticides registered for the protection of forests. High costs and long registration process considerably limited the interest of producers in placing the plant protection products on the market. Systematic decrease in the number of plant protection products possible to register for use in forestry, as well as the principles of integrated plant protection established in the EU in 2014 call for seeking plant protection methods based on the natural enemies of pests, such as pathogenic microorganisms, parasites and predators. Therefore, contemporary forest protection requires the advancement of integrated methods for pest insect and disease control through developing methods of forecasting forest dangers, the use of natural enemies and agro-technical methods for regulation of pests, as well as the development of decision support systems as a tool facilitating introduction of integrated forest protection principles. Such support systems help to establish optimal terms for the implementation of protection measures, so as to increase their efficiency while limiting the use of chemical pesticides to an absolute minimum.

Bark beetles infest several pine tree species, often creating major economic losses. Biotic interactions between Scolytinae populations inhabiting Pinus sylvestris were analyzed using a new sampling method involving a two dimensional division of tree space resources into units and sections. The goal was to evaluate the effects of the type of available reproduction material on insect infestation. The P. sylvestris stands in this study included an analysis of bark beetles colonizing dead trees and uninfested living trees (trap trees). Analyzed trees were harvested and their stems divided into ten equal units; each unit was halved into two sections (half of the stem circumference). The colonization of dead trees by insects was evaluated immediately after cutting for infested trees and 2 months after cutting for trap trees. The type of breeding material significantly affected the species composition of bark beetles infesting P. sylvestris. The dead trees were colonized mostly by Trypodendron lineatum and Tomicus minor, and to a lesser extent by Tomicus piniperda, which dominated in trap trees. Tomicus piniperda and T. lineatum preferred thicker stems; however, T. minor, Hylurgops palliatus and Pityogenes bidentatus preferred thinner ones. The application of the new sampling method helped to increase the accuracy of niche segregation among insect species. The results of niche segregation indicate bark beetles exhibited spatial specialization in the use of resources, mainly related to the moisture content of breeding material and the availability of food resources which is the main factor determining the coexistence of bark beetles in the same environment.

The study estimated the efficacy of a net coated with alpha-cypermethrin used to protect Norway spruce [Picea abies (L.) H. Karst] wood against bark beetle infestations. The inside of the net was coated with 100 mg/m2 of alpha-cypermethrin. The studies carried out in 2010 and 2011 took place in southern Poland in the Beskid Żywiecki and Beskid Sądecki mountains in P. abies stands threatened by Ips typographus (Linnaeus, 1758). The research material consisted of wood logs taken off of 70–80-year-old P. abies trees. The logs were wrapped in the net in the early spring before spring swarming of bark beetles. The unwrapped logs were considered as the study controls. An evaluation of the treatments was performed after 2 months. The evaluation was based on the counting of bark beetles galleries found after removing the bark from the wrapped and unwrapped logs. There were no insect galleries on the wrapped logs. The net was a barrier, on which bark beetles died. About 10 dead I. typographus beetles were found on 0.01 m2 of the net surface. On the bark from the unwrapped-control logs there were 3,156 galleries/entrance holes of bark beetles, in total, of which 73% belonged to I. typographus, 13% to Pityogenes chalcographus (Linnaeus, 1761), and almost 10% to Xyloterus lineatus (Oliv.). These results indicated the high efficacy of the net coated with alpha-cypermethrin used for the protection of Norway spruce wood against the bark beetles. At the same time, the net was found to have a negative effect on non-target entomofauna, mainly Hylobius spp., Thanasimus formicarius (L.), and Tetropium castaneum (F.). However, the use of a net did not affect other predatory (Carabidae) and parasitic (Ichneumonidae and Tachinidae) entomofauna.

The method of single-strand conformational polymorphism (SSCP) was modified in our laboratories for the characterization of baculoviruses, insect viruses with great potential for use as bioinsecticides in biological protection programs. A series of primers were synthesized after the comparison of the polyhedrin gene sequences of over 20 baculoviruses. Polyhedrin is a highly conserved protein which is responsible for the persistence of the virus in the environment. Universal primers were designed which could be used in polymerase chain reactions (PCR) containing genomic DNA from an array of nucleopolyhedrosis viruses (NPVs) including these which are used as biopesticides against important pests of forests and crops, such as Anticarsia gemmatalis, Spodoptera frugiperda, Lymantria dispar, Lymantria monacha and many others. PCR products were denatured and subjected to single-strand DNA electrophoresis at variable temperatures (MSSCP) where, after silver staining, they gave ssDNA band patterns characteristic for each baculovirus species. This technique can be potentially applied to detect baculoviruses in insects collected in the field, as well as to plant tissues and the excrements or bodies of predators without need for sequencing the PCR products. Sometimes MSSCP can be used not only for species determination but also as an indication of genomic variability which can be related to infectivity.

A new isolate of baculovirus, Lymantria dispar multiple nucleopolyhedrovirus-BNP (LdMNPV-BNP), was found in dead gypsy moth (L. dispar) caterpillars collected in the Biebrzanski National Park in Poland. Here, we examined its biological activity, structure, genetic content and phylogeny. Multiple nucleocapsids of LdMNPV-BNP are enveloped together in 2-26 virions embedded in occluded bodies (OBs) very similar to the OBs previously described in viruses infecting Lymantriinae. This isolate kills pest larvae in a relatively short time (LT50 of approximately 9days for a dose of 2×10(7)OBs/ml), highlighting the possibility for its use as a biopesticide. Next-generation sequencing of LdMNPV-BNP revealed gene content (e.g. DNA photolyase) that is not present in any LdMNPV isolate sequenced to date. The genome is 157,270 base pairs long and has a notably lower G+C content in comparison to other LdMNPVs (50.3% G+C content compared to an average of 57.4% among other LdMNPVs). According to our phylogenetic analysis based on 37 core genes, LdMNPV-BNP is a member of group II alphabaculoviruses, which are closely related to LdMNPV and LyxyMNPV (Lymantria xylina multiple nucleopolyhedrovirus). Molecular evolution inference based on the partial sequence of lef-8, lef-9 and polh genes shows that LdMNPV-BNP and isolates of Lymantria monacha nucleopolyhedrovirus (LymoNPV) may share a very recent common ancestor or be isolates of the same virus species. LdMNPV-BNP, like other baculoviruses, could be beneficial as an active component of biopesticides that can be used during forest integrated pest management.
