ResumoO gênero Cryptosporidium é composto por protozoários com grande capacidade de reprodução e disseminação. Sua transmissão pode ocorrer indiretamente pela ingestão de água e alimentos contaminados com oocistos viáveis ou diretamente entre animais, entre humanos ou entre animais e humanos. Em diversas partes do mundo, animais de companhia tem sido citados como fontes potenciais de infecção por Cryptosporidium spp. O presente estudo teve como objetivo avaliar a ocorrência da infecção por Cryptosporidium spp. em cães domiciliados na cidade do Recife, estado de Pernambuco, Brasil, bem como a associação da infecção com dados epidemiológicos e sanitários. Foram analisadas 97 amostras de fezes de caninos domiciliados, utilizando-se as técnicas de centrífugo-sedimentação em formol-éter com posterior coloração pelo método de Kinyoun. A análise estatística foi realizada por meio dos testes Qui-quadrado de Pearson e Exato de Fisher, com nível de significância de 5,0%. Detectou-se positividade de 31,9% (31/97), tratando-se do primeiro registro de ocorrência da infecção por Cryptosporidium spp. na espécie canina no nordeste brasileiro, verificando-se associação significativa entre a taxa de infecção e a assistência médica veterinária, vacinação recebida e o tipo de ambiente de criação dos cães. Transmission can occur indirectly by ingestion of water contaminated with viable oocysts or directly between animals, between humans, or between animals and human. In various parts of the world, pets have been cited as potential sources of infection by Cryptosporidium spp. The present study aimed to evaluate the occurrence of natural Cryptosporidium spp. infection in domestic dogs in the city of Recife, Pernambuco State, Brazil, as well as the association of the infection with epidemiological and sanitary data. Stool samples from 97 dogs were analyzed using the techniques of centrifugal sedimentation in formalin-ether with subsequent staining by Kinyoun method. Statistical analysis was performed using the chi-square test of Pearson and Fisher Exact test, with significance level of 5.0%. Positivity was detected in 31.9% (31/97) of samples. It was the first record of infection by Cryptosporidium spp. in dogs from northeastern Brazil. We verified significant association between the rate of infection and veterinary medical care, vaccination, and type of environment in which the dogs were kept.
A esquistossomose, causada pelo trematódeo Schistosoma mansoni no Brasil, assume caráter de doença infecciosa com quadros agudos e crônicos, estando a instalação da doença intimamente ligada à persistência dos focos de transmissão, às precárias condições socioambientais e à presença do vetor Biomphalaria glabrata. Em Pernambuco, embora a doença predomine em áreas rurais, casos em áreas litorâneas vêm sendo registrados, principalmente onde existem problemas de saneamento básico. Objetivou-se com este estudo, avaliar a frequência da infecção por S. mansoni e analisar aspectos socioeconômicos e higiênico-sanitários ligados ao parasitismo em comunidades da Região Metropolitana de Recife -PE. O estudo foi desenvolvido em 14 comunidades nos municípios de Camaragibe, Goiana, Recife e Igarassu, com anuência das Secretárias de Saúde de cada município. Para o diagnóstico da doença, utilizaram-se as técnicas de Kato-katz, sistema Coprotest®, Hoffmann, Faust e Willis. Aplicou-se um questionário para a obtenção de dados epidemiológicos. Obteve-se positividade de 7,3%, sendo as variáveis de associação significativa a cidade e comunidade de residência dos indivíduos, a faixa etária, número de pessoas na residência, tipo de habitação, e, principalmente, aquelas ligadas à obtenção, uso e contato com a água.
O presente estudo teve como objetivo verificar a taxa de infecção por Cryptosporidium spp. e analisar a influência do manejo higiênico-sanitário em ovinos de propriedades localizadas na Região Metropolitana do Recife e Zona da Mata de Pernambuco. Foram analisadas 194 amostras de fezes de ovinos de 9 propriedades, utilizando-se as técnicas de sedimentação em formol-éter e coloração segundo Kinyoun. A taxa de infecção e os fatores de risco ligados à higiene e manejo foram analisados através do teste Quiquadrado de Pearson e do teste Exato de Fisher, com nível de significância de 5%. Foi observada positividade em 11,29% (21/186) das amostras. O sistema de criação extensivo teve associação significativa com a doença e os cuidados relativos ao manejo podem ser determinantes para a disseminação e presença de oocistos nas fezes de ovinos.
RESUMOCryptosporidium spp. é um protozoário gastrointestinal que pode infectar as microvilosidades das células do intestino de mais de 150 espécies de vertebrados. Muitos fatores são determinantes para que os oocistos se disseminem e o risco da infecção depende da dose, virulência e infectividade dos mesmos, além do grau de susceptibilidade do hospedeiro. A transmissão ocorre pela ingestão de oocistos que são eliminados nas fezes em sua forma infectante. Água, alimentos contaminados e animais infectados são principal fonte de transmissão de Cryptosporidium spp. Esta enfermidade é considerada uma zoonose de grande importância em saúde pública devido à ocorrência de casos assintomáticos. Quando o sistema imunológico está comprometido os indivíduos podem vir a óbito. No presente estudo objetivou-se avaliar a freqüência de infecção por Cryptosporidium spp. e sua associação com as condições higiênico-sanitárias da população humana de comunidades dos municípios de Recife, Camaragibe, Goiana e Igarassu -PE, Brasil. As pessoas foram convidadas a assinar um termo de Consentimento Livre Esclarecido (TCLE), confirmando sua participação na pesquisa, selecionados por amostragem não probabilística. As amostras fecais foram processadas utilizando a técnica de centrífugo-flutuação em formol éter e cada lâmina foi corada segundo a Técnica de Ziehl-Neelsen modificado (Kinyoun). Foram realizadas três repetições para cada amostra. Da amostragem foi obtida uma positividade de 17,12% (62/362) para Cryptosporidium spp.nas onze comunidades das quatros cidades.
A survey was carried out to detect American cutaneous leishmaniasis (ACL) among dogs in an area where a human outbreak had occurred in the state of Pernambuco, in northeastern Brazil. Domiciled dogs living in the district of Três Ladeiras, Igarassu were used in the present study. The following procedures were performed: The Polymerase Chain Reaction (PCR) (n = 126); the Immunofluorescence Antibody Test (IFAT) (n = 80); and a parasitological examination to detect amastigote forms of Leishmania sp. in skin lesions (n = 43). Associations between the infection in animals and the clinical and epidemiological factors were analyzed using Fisher's exact test or the Pearson's chi-squared test. In total, 46.8% (59/126) of the samples tested were PCRpositive. Although a higher frequency of positivity was detected among males (46.3 %) and animals aged between 3 and 4 years (50.0 %), no significant associations were recorded for these variables (p> 0.05). Similarly, the clinical signs and aspects related to the environment in which the animal lives did not differ significantly, but differences were recorded for the variable locality. In the IFAT, only 6.2% (5/80) of the dogs were positive and no amastigote forms of Leishmania sp. were detected.
Sheep and goats suffer heavy losses due to the high prevalence of gastrointestinal nematodes, mainly from the nematode Haemonchus contortus. The parasite causes high morbidity and mortality, even though, the indiscriminate use of chemicals to control H. contortus has favored the selection of multidrug resistance parasites. Molecular diagnosis of resistance to the benzimidazole (BZs) group is important to indicate substitute strategies of control. The aim of this study was to determine the presence of the mutation at position F200Y of the beta-tubulin gene in seven populations of H. contortus from sheep and goats’ farms from the state of Pernambuco, Brazil. Twenty adult H. contortus male worms were recovered for DNAextraction from naturally infected animals, from farms from Recife, Sairé, Camocim deSão Félix, Gameleira, Moreno, Bonito, and Serra Talhada. The primers forward: 5` AAT GCT TCC ACC CTT GTC CAT C 3’, and reverse: 5’ CAA CAA CGG GCA TGA AGA AG 3’ were used to search for the main codon (F200Y) of the beta-tubulin gene to determine BZ resistance. The NC1/ITS-2 forward: 5’ ACG TCT GGT TCA GGG GTT TT 3’ and reverse: 5’ TTA TCT GTT TTT CCT GCC CT 3’ primers were used for confirming H. contortus species. A questionnaire was applied to obtain information on management practices, including the frequency of BZs use. The sequences from the NC1/ITS-2 confirmed the presence of H. contortus from all samples. The F200Y mutation responsible for BZ resistance (TTC/TAC) in H. contortus was found only in the samples from Sairé, as the TTC codon varied from 55.6 to 85.7%. In relation to the questionnaire, producers revealed the absence of quarantine for newly arrived animals; high drug treatment frequency (60-days interval); treatment of the whole flock; the visual estimation of the weight of the animals for dose determination; and high drug rotation. It was also mentioned the lack of veterinary assistance with no use of fecal exams. Even though, all these factors may be essential to increase parasite selection pressure, it was not possible to determine which of those showed a higher risk due to the high number of variables and the small number of participants. This is without any doubt one of the great difficulties to correlate phenotypic drug efficacy data (ex. FECRT) and genotypic-related (ex. PCR) resistance markers in helminths. We have included some risk factors reported by the farmers from Pernambuco and their influences in the process of drug selection, as well as a few strategies to mitigate this situation. The present report constitutes the first record of a molecular diagnosis of the mutation F200Y that confers resistance to the BZ group in ruminants in the state of Pernambuco, Brazil.
The control of Aedes aegypti has been considered one of the most important public health challenges worldwide. Chemical compounds have long been used for this purpose, but resistance to these molecules has also increased. Therefore, over the last few years several studies have focused on the development of alternative tools, particularly those based on plant metabolites. The purpose of this study was to assess the larvicidal activity of Caesalpinia ferrea and Lippia origanoides against Ae. aegypti. Larvae (L3) of Ae. aegypti Liverpool and Rockefeller strains, as well as of the Recife population were exposed to different concentrations of C. ferrea (ranging from 13.1 to 105 mg/mL) and L. origanoides (ranging from 16.3 to 130 mg/mL), and the mortality rate was evaluated up to 48 hours after the beginning of the experiment. All tested groups and ontrol group were quadruplicated. For C. ferrea, mortality ranged from 42.5% to 100% for Ae. aegypti Liverpool strain,from 67% to 100% for Ae. aegypti Rockfeller strain, and 57% to 100% for Ae. aegypti Recife population after 48 hours of larval exposure. For L. origanoides, the larvicidal activity ranged from 75% to 100% for Ae. aegypti Liverpool strain, from 61.5% to 100% for Ae. aegypti Rockfeller strain, and from 60.5% to 100% for Ae.aegypti Recife population. The hydro ethanol extract of C. ferrea and L. origanoides presented larvicidal activity against Ae. aegypti.KEY WORDS: Aedes aegypti; hydro ethanol extracts; Caesalpinia ferrea; Lippia sidoides; botanical insecticide; mosquitoes.
RESUMO -As zoonoses parasitárias são um problema de saúde pública e diante da proximidade entre animais e humanos, objetivou-se avaliar a percepção de tutores de cães e gatos residentes à cidade do Recife, estado de Pernambuco, Brasil, quanto à infecção por Cryptosporidium spp. e seus fatores de risco. Para tanto, foi aplicado um questionário, composto por perguntas fechadas, a 71 tutores. Os resultados foram tabulados em planilhas do Microsoft Excel® versão 2007 para obtenção das frequências absolutas e relativas. Entre os tutores participantes, 16,9% (n = 12) eram do sexo masculino e 83,1% (n = 59) do sexo feminino. Suas respectivas idades variaram de 18 a 75 anos. Em relação à criptosporidiose, apenas 11,3% (n = 8) dos tutores responderam ter conhecimento sobre o assunto e 88,7% (n = 63) citaram uma total insciência. Um percentual de 100% (n = 71) foi igualmente alcançado ao serem questionados sobre o uso de medicações anticoccidianas, respondendo não utilizá-las. Sobre a possibilidade de os animais transmitirem doenças para o ser humano, afirmaram ter esse conhecimento. Dentre os participantes, 73,2% (n = 52) afirmaram recolher as fezes de seus animais. Dos tutores 80,2% (n = 57) relataram que seus animais possuíam acompanhamento por médico veterinário. O conceito de zoonose foi corretamente respondido por apenas 21,1% (n = 15) deles. Os resultados obtidos evidenciaram um reduzido nível de conhecimento sobre fatores de risco relacionados à infecção por Cryptosporidium spp., o que expõe tanto os tutores e seus animais quanto a saúde coletiva da região estudada à criptosporidiose.Palavras-Chaves: criptosporidiose; caninos; felinos; zoonose.ABSTRACT -Parasitic zoonoses are a public health problem and given the proximity between animals and humans, aimed to evaluate the perception of tutors of dogs and cats living to the city of Recife, state of Pernambuco, Brazil, as to infection by Cryptosporidium spp. and their risk factors. On the possibility of the animals transmitting diseases to humans, they claimed to have that knowledge. Among the participants, 73.2% (n = 52) stated collect the feces of their pets. Tutors 80.2% (n = 57) reported that their animals had monitoring by a veterinarian. The concept of zoonosis was correctly answered by only 21.1% (n = 15) of them.
