Cis-dichlorodiammineplatinum II (CDDP) is a chemotherapeutic agent that induces nephrotoxicity by different mechanisms, including oxidative stress, mitochondrial dysfunction, autophagy, and endoplasmic reticulum stress. This study aimed to evaluate if the protective effects of the antioxidant alpha-mangostin (αM) in CDDP-induced damage in proximal tubule Lilly laboratory culture porcine kidney (LLC-PK1) cells, are related to mitochondrial function preservation. It was found that αM co-incubation prevented CDDP-induced cell death. Furthermore, αM prevented the CDDP-induced decrease in cell respiratory states, in the maximum capacity of the electron transfer system (E) and in the respiration associated to oxidative phosphorylation (OXPHOS). CDDP also decreased the protein levels of voltage dependence anion channel (VDAC) and mitochondrial complex subunits, which together with the reduction in E, the mitofusin 2 decrease and the mitochondrial network fragmentation observed by MitoTracker Green, suggest the mitochondrial morphology alteration and the decrease in mitochondrial mass induced by CDDP. CDDP also induced the reduction in mitochondrial biogenesis observed by transcription factor A, mitochondria (TFAM) decreased protein-level and the increase in mitophagy. All these changes were prevented by αM. Taken together, our results imply that αM’s protective effects in CDDP-induced toxicity in LLC-PK1 cells are associated to mitochondrial function preservation.

Z-Ligustilide (1) and Z-6,6',7,3'-alpha-diligustilide (2), two of the major active phthalides of the medicinal plant Ligusticum porteri (osha), were chosen for the development and validation of an HPLC-diode array detection method suitable for QC of the crude drug. The method used gradient elution to achieve separation on a Hibar RT LiChrospher 100 C18 column. The LOD values were 29 and 45 microg/mL, and the LOQs were 89 and 125 microg/mL, respectively. The method showed good intraday precision (%RSD: 0.7 for 1 and 3.1 for 2) and interday precision (%RSD: 1.2 for 1 and 1.8 for 2). The method was used for the analysis of 1 and 2 in crude drug samples and several herbal preparations from Mexico and the United States. Quantitative analysis showed that the content of the two phthalides varied significantly among the samples. All the samples contained higher concentrations of 1 (0.15-2.5%) than 2 (0.002-1.0%). The profiles of volatile compounds in the essential oil obtained by hydrodistillation and solid-phase microextraction of L. porteri roots were analyzed by GC-MS. Thirty one chemical constituents (> 99.7% of the total content) were identified in the essential oil, which was characterized by the presence of a high percentage of phthalides (44.61%) and sesquiterpenes (10.69%). The major light volatile components extracted by solid-phase microextraction were monoterpenes.

Neuropeptide Y (NPY), a peptide released by nervous cells, appears to contribute to adiposity regulation by increasing food intake and inhibiting lipolysis. New NPY receptor related antagonists such as S.A.0204 are being developed as potential anti-obesity drugs affecting adipocyte lipid metabolism and thermogenesis. In this sense, those animals fed on a high-energy yielding (cafeteria) diet decreased body fat weight as compared to overweight controls, when they were administered with S.A.0204, and increased body temperature, which statistically correlated with high UCP2 mRNA expression levels in white adipose tissue. In addition, the in vivo NPY-antagonist administration was able to prevent white adipose tissue growth in animals fed the cafeteria (high-fat) diet by impairing PPARy and CIEBPalpha mRNA expression in white fat cells. In summary, this novel NPY related-antagonist S.A.0204 may regulate body fat deposition by affecting both energy dissipation and white adipose tissue deposition, representing a potential new pharmacological strategy for obesity management.

Potent and selective rNPY antagonists will be able to probe the merits of this approach for the treatment of obesity. We report the synthesis and preliminary evaluation of some hydrazide derivatives as antagonists of rNPY.
