The individual application of pure and active compounds such as methionine may help to address water scarcity issues without compromising the yield of wheat. As organic plant growth stimulants, amino acids are popularly used to promote the productivity of crops. However, the influence of the exogenous application of methionine in wheat remains elusive. The present investigation was planned in order to understand the impact of methionine in wheat under drought stress. Two wheat genotypes were allowed to grow with 100% field capacity (FC) up to the three-leaf stage. Twenty-five-day-old seedlings of two wheat genotypes, Galaxy-13 and Johar-16, were subjected to 40% FC, denoted as water deficit-stress (D), along with 100% FC, called control (C), with and without L-methionine (Met; 4 mM) foliar treatment. Water deficit significantly reduced shoot length, shoot fresh and dry weights, seed yield, photosynthetic, gas exchange attributes except for transpiration rate (E), and shoot mineral ions (potassium, calcium, and phosphorus) in both genotypes. A significant increase was recorded in superoxide dismutase (SOD), catalase (CAT), hydrogen peroxide (H2O2), malondialdehyde (MDA), and sodium ions (Na+) due to water deficiency. However, foliar application of Met substantially improved the studied growth, photosynthetic, and gas exchange attributes with water deficit conditions in both genotypes. The activities of SOD, POD, and CAT were further enhanced under stress with Met application. Met improved potassium (K), calcium (Ca2+), and phosphorus (P) content. In a nutshell, the foliar application of Met effectively amended water deficit stress tolerance by reducing MDA and H2O2 content under water deficit conditions in wheat plants. Thus, we are able to deduce a positive association between Met-induced improved growth attributes and drought tolerance.

In this study, the functional metabolites of Moringa oleifera (MO) were screened to evaluate their possible role in accelerated functional retrieval after peripheral nerve injury (PNI). MO leaves were used for extract preparation using solvents of different polarities. Each dry extract was uniformly mixed in rodents’ chow and supplemented daily at a dose rate of 2 g/kg body weight from the day of nerve crush until the completion of the trial. The sciatic functional index (SFI) and muscle grip strength were performed to assess the recovery of motor functions, whereas the hotplate test was performed to measure the regain of sensory functions. An optimal level of oxidative stress and a controlled glycemic level mediates a number of physio-biochemical pathways for the smooth progression of the regeneration process. Therefore, total oxidant status (TOS), total antioxidant capacity (TAC) and glycemic levels were analyzed in metabolite-enriched extract-treated groups compared to the control. The supplementation of polar extracts demonstrated a significantly high potential to induce the retrieval of sensory and motor functions. Further, they were highly effective in controlling oxidative stress, facilitating accelerated nerve generation. This study has highlighted MO as a sustainable source of nutritive metabolites and a valuable target for drug development.

Endometrial cancer is the fourth most common cancer in women. It arises from the endometrium and accompanied by the abnormal growth of the cells. Sign and symptoms include pelvic pain and abnormal vaginal bleeding. It has two categories. Type 1 tumors are estrogen-dependent and they have mutations in PTEN, PIK3CA while Type 2 tumors are more sensitive and have mutations in TP53. Overactivation of the signaling pathway (PI3K) results in anti-apoptosis. Here, this study aims to identify Tumor-Specific Antigen for germline mutations in endometrial cancer which can be used as a potential vaccine candidate. The germline mutations data are obtained from cancer gene census of the cosmic database. Genes mutating with crucial role in endometrial cancer are considered. Peptides libraries are generated using peptide design library. Human leukocyte antigen alleles are identified for the peptide library through NetMHC. Binding affinities of alleles with peptide are determined. Linear regression is performed to generate graphs. PTEN, TP53, PIK3CA, KRAS, and CTNNB1 proved to have critical role. About 575 overlapping peptide libraries are generated and each peptide has a length of 18-20 amino acids. Approximately 58 HLAs are identified, having strong interactions with HLAs. Regression analysis shows that the no. of mutations are directly associated with a binding affinity of peptides. From this, we suggest that the identified TSA can be used as personalized peptide vaccines that directly target the mutated genes in endometrial cancer. This research work can be used in the laboratories for further validation.

Hypothetical proteins (HPs) are those whose sequence is present but nothing is known about their structural and functional annotations. Elucidation of structural and functional insights would be effective enough in perceiving the protein interactions and their involvement in various pathways and networks. Ananas comosus is considered as third most essential tropical fruit and is given more consideration from the commercial point of view; therefore, in this research, it is taken for analysis using different approaches of bioinformatics. On the basis of subcellular localization and secondary structure analysis, it was suggested that HP is the nuclear protein consisting of α helices and more abundant coils. Homology modeling has been done through Swiss Model to determine their template structure but template identity score is not enough which uncover the fact that the protein OAY63476.1 is unpredicted in-vitro. In order to predict the 3D structure of protein, Phyre 2 server is utilized. Results are validated by different strategies revealing the stability of the developed model. The quality factor of this protein is 45.28. Functional analysis and domain identification have been performed by using NCBI-CDD and Pfam which suggested that protein contains ring variant domain. Comparative genome analysis has been performed with other proteins of plants which demonstrate that the selected protein has the highest similarity. This study paves the way in order to determine the structural and functional annotations of other uncharacterized proteins and effective in exploring other novel proteins along with their functions in the same way as that of this research study.
