The development and validation of different types of molecular markers is crucial to conducting marker-assisted sesame breeding. Insertion-deletion (InDel) markers are highly polymorphic and suitable for low-cost gel-based genotyping. From this perspective, this study aimed to discover and develop InDel markers through bioinformatic analysis of double digest restriction site-associated DNA sequencing (ddRADSeq) data from 95 accessions belonging to the Mediterranean sesame core collection. Bioinformatic analysis indicated the presence of 7477 InDel positions genome wide. Deletions accounted for 61% of the InDels and short deletions (1–2 bp) were the most abundant type (94.9%). On average, InDels of at least 2 bp in length had a frequency of 2.99 InDels/Mb. The 86 InDel sites having length ≥8 bp were detected in genome-wide analysis. These regions can be used for the development of InDel markers considering low-cost genotyping with agarose gels. In order to validate these InDels, a total of 38 InDel regions were selected and primers were successfully amplified. About 13% of these InDels were in the coding sequences (CDSs) and in the 3′- and 5′- untranslated regions (UTRs). Furthermore, the efficiencies of these 16 InDel markers were assessed on 32 sesame accessions. The polymorphic information content (PIC) of these 16 markers ranged from 0.06 to 0.62 (average: 0.33). These results demonstrated the success of InDel identification and marker development for sesame with the use of ddRADSeq data. These agarose-resolvable InDel markers are expected to be useful for sesame breeders.

The concept of ‘crop ideotype’ is coined as a desirable plant model expected to better perform for seed yield, oils and other useful characteristics when developed as a cultivar, and it consists of two major approaches, namely, (i) ‘defect elimination’, that is, integration of disease resistance to a susceptible genotype from a resistant genotype and (ii) ‘selection for yield’ by improving yield after crosses between desirable parents. For consideration of these approaches, here we introduced an ideotype in kabuli chickpea (Cicer arietinum L.) which is high-yielding, extra-large-seeded, and double- or multi-podded, has high plant height and imparipinnate-leafed traits, and is heat tolerant and resistant to ascochyta blight [Ascochyta rabiei (Pass.) Labr.], which causes considerable yield losses, via marker-assisted selection. F3 and F4 lines were evaluated for agro-morphological traits divided into six classes, namely, (i) imparipinnate-leafed and single-podded progeny, (ii) imparipinnate-leafed and double-podded progeny, (iii) imparipinnate-leafed and multi-podded progeny, (iv) unifoliolate-leafed and single-podded progeny, (v) unifoliolate-leafed and double-podded progeny, (vi) unifoliolate-leafed and multi-podded progeny. F3:4 lines having 100-seed weight ≥ 45 g and double- or multi-podded traits were additionally assessed for resistance to ascochyta blight using molecular markers including SCY17590 and CaETR-1. Superior lines having higher values than their best parents were determined for all studied traits indicating that economic and important traits including yield and seed size in chickpea could be improved by crossing suitable parents. Imparipinnate-leafed and multi-podded plants had not only the highest number of pods and seeds per plant but also the highest yield. On the other hand, imparipinnate-leafed and single podded progeny had the largest seed size, followed by imparipinnate-leafed and double-podded progeny. Multi-podded plants produced 23% more seed yield than that of single-podded plants, while multi-podded plants attained 7.6% more seed yield than that of double-podded plants. SCY17590 and CaETR-1 markers located on LG4 related to QTLAR2 and QTLAR1 were found in 14 lines among 152 F3:4 lines. Six superior lines were selected for being double- or multi-podded, imparipinnate-leafed, suitable for combine harvest, heat-tolerant, and resistant to ascochyta blight, and having both of two resistance markers and extra-large seeds as high as 50–60 g per 100-seed weight. Resistance alleles from two different backgrounds for resistance to ascochyta blight were integrated with double- or multi-podded kabuli chickpea lines having high yield, extra-large seeds, high plant height, imparipinnate-leaves and high heat tolerance, playing a crucial role for future demands of population and food security. These approaches seem to be applicable in ideotype breeding for other important crop plants.

During storage, chickpea (Cicer arietinum L.) is severely attacked by some bruchid species, especially Callosobruchus chinensis L. (Coleoptera: Bruchidae), resulting in losses in quantity and nutritional quality. In the present study, three species of Cicer including five accessions of Cicer arietinum L. (three kabuli and two desi chickpeas), four accessions of C. echinospermum P.H. Davis and five accessions of C. reticulatum Ladiz. were screened for resistance to C. chinensis in both free-choice and no-choice tests in the laboratory. Resistance was evaluated by measuring oviposition (number of eggs per seed), adult emergence (number of holes per seed), damaged seed rate and seed weight loss (%). The results revealed that no eggs were laid by the bruchid females to the C. echinospermum accessions in free-choice test, but in nochoice test only two C. echinospermum accessions (AWC 304 and AWC 305) had few eggs (4.3 and 3.3 eggs/seed, respectively). The highest rate of oviposition occurred in kabuli chickpeas, especially in YAR (25.1 eggs/seed in free-choice test). The accessions exhibited a similar pattern for adult emergence like in the oviposition rates. As for damaged seed rate, no damage was observed in both tests in the C. echinospermum accessions, except AWC 304 (6.7%) in no-choice test. The highest seed damage was seen in kabuli-type accessions, being 100% in YAR and ILC 8617. Considering seed weight loss, no weight loss occurred in the C. echinospermum accessions in both tests (except, AWC 304 in no-choice test) whereas the highest weight loss was seen in the kabuli-type accession, CA 2969 (28.6%) in free-choice test and in the desi type accession, ICC 4957 (35.0%) in no-choice test. Since lower numbers of eggs were laid on hairy, wrinkled/reticulated and dark seed accessions, these features seemed to be important in the preference of the bruchid for host selection and oviposition. These results suggest that resistant varieties can be used as gene sources in breeding new cultivars resistant to C. chinensis.

The rose sawfly, Arge rosae L. (Hymenoptera: Argidae), is one of the most destructive pests of roses grown both indoors and outdoors in Antalya province (south-western part of Turkey). Especially in greenhouse rose cultivation, growers depend heavily on synthetic pesticides to control this pest along with other arthropod pests in their crops. The aim of this study was to evaluate biocontrol potential of some indigenous soil-borne entomopathogenic fungi (EPF) against the pest. In pathogenicity assays, a total of 17 EPF isolates, belonging to three species [Beauveria bassiana (Bals.) Vuill. -14, Clonostachys rosea (Link) Schroers -2 and Isaria farinosa (Holmsk.) Fr. -1] were tested against the 4 th instar larvae of A. rosae under laboratory conditions. All the isolates were applied at a conidial suspension of 1 × 10 7 conidia/ml, using the spray method. The results from the assays showed that bioefficacy of tested isolates increased significantly with elapsed time up to 9 days after inoculation. Of the 17 isolates tested, 5 isolates of B. bassiana (BbDm-1, BbKp-1, BbMp-1, BbSr-1 and BbMg-2) and 1 isolate of I. farinosa (IfGp-1) appeared to be most promising, causing mortalities between 76.7 and 86.7% 3 days post treatment. Probit analysis of mortality data showed that there were significant differences in the susceptibility of 4 th instar larvae of A. rosae to tested EPF isolates. The above-mentioned six isolates had the lowest LT50 and LT95 values (ranged from 1.66 to 7.50 and 4.81 to 8.49 days, respectively), implying their high virulence and their biocontrol potential against the pest. Phylogenetic analysis based on the ITS region sequence revealed that all tested isolates showed high similarities (ranged from 99 to 100%) with other isolates of their respective fungal species in GenBank.Overall results suggested that the most virulent above-mentioned six isolates had significant potential as biological control agents against A. rosae.

Background The Colorado potato beetle (CPB) Leptinotarsa decemlineata (Say) (Coleoptera: Chrysomelidae) is one of the most damaging pests of potatoes all over the world as well as in Turkey. Both adults and larvae of this pest feed on the foliage of potatoes, and often cause complete defoliation of potato plants, with considerable yield losses of up to 60%. Chemical control of this pest has induced the selection of resistant CPB populations and negative environmental impact. The aim of the present study was to evaluate the pathogenicity of 14 Turkish isolates of Beauveria bassiana (Balsamo) Vuillemin (Deuteromycota: Hyphomycetes) against different stages of CPB under laboratory conditions. Results All isolates were tested as conidial suspensions of (1 × 107 conidia/ml), using spray method. The results from pathogenicity assays showed that virulence of tested isolates was variable, and mortality increased with the time of exposure. The highest mortality was seen in larvae of CPB; however, young larvae (1st and 2nd instars) were more susceptible to fungal isolates than older ones (3rd and 4th instars). Four isolates (BbDm-1, BbDs-2, BbMg-2, and BbMp-1) were more pathogenic than others against L. decemlineata, causing mortalities between 96.7 and 100% in the 2 youngest larval instars, respectively, between 91.7 and 96.7% in the 2 oldest larval instars, respectively, and between 93.3 and 96.7% in 0–48-h-old adults, respectively, 9 days post treatment. Furthermore, these 4 isolates had the most potent egg-hatching inhibitory effects (66.7–90%). The results of molecular phylogenetic analyses based on the ITS region sequence indicated that all tested B. bassiana isolates showed a high similarity (99%) with other B. bassiana isolates in GenBank. Conclusions Results suggested that the 4 B. bassiana isolates were highly virulence and had the potential for biological control of CPB. Further tests under field conditions are recommended to validate the laboratory results.
