The disease outbreaks in aquaculture system of wetlands are the major cause of fish mortality. Among various bacterial septicaemic diseases, fish mortality caused by Acinetobacter spp. is recently reported in different fish species. Fish disease outbreak was investigated in a wetland of West Bengal, India to identify the aetiological factors involved. The moribund fish were examined and subjected to bacterial isolation. Two bacterial causative agents were identified as Acinetobacter junii and Acinetobacter pittii by biochemical characterization and 16S rRNA gene amplification. Both the isolates were oxidase-negative, nitrate-negative, catalase-positive and indole-negative. The molecular identification using 16S rRNA gene sequencing and phylogenetic tree analysis further confirmed the two Acinetobacter spp. with 97%-99% similarity. The antibiotic resistance patterns of these two bacteria revealed that both of them were resistant to β-lactam, cefalexin, cephalothin, amoxyclav, cefuroxime, cefadroxil, clindamycin, vancomycin and penicillin. In addition, A. pittii was also resistant to other antibiotics of cephams group such as ceftazidime and cefotaxime.In the challenge experiment, both A. junii and A. pittii were found to be pathogenic with LD 50 of 1.24 × 10 5 and 1.88 × 10 7 cfu/fish respectively. Histopathological examination of gill, liver and kidney revealed prominent changes supporting bacterial septicaemia. The investigation reports for the first time on concurrent infection by A. junii and multidrug-resistant (MDR)-A. pittii as emerging fish pathogens to cause severe mortality in Labeo catla and Hypophthalmichthys molitrix in a freshwater wetland.

Ichthyophthirius multifiliis, a ciliated parasite causing ichthyophthiriasis (white spot disease) in freshwater fishes, results in significant economic loss to the aquaculture sector. One of the important predisposing factors for ichthyophthiriasis is low water temperature (i.e., below 20°C), which affects the health and makes freshwater fishes more susceptible to parasitic infections. During ichthyophthiriasis, fishes are stressed and acute immune reactions are compromised, which enables the aquatic bacterial pathogens to simultaneously infect the host and increase the severity of disease. In the present work, we aimed to understand the parasite–bacteria co-infection mechanism in fish. Later, Curcuma longa (turmeric) essential oil was used as a promising management strategy to improve immunity and control co-infections in fish. A natural outbreak of I. multifiliis was reported (validated by 16S rRNA PCR and sequencing method) in Pangasianodon hypophthalmus from a culture facility of ICAR-CIFRI, India. The fish showed clinical signs including hemorrhage, ulcer, discoloration, and redness in the body surface. Further microbiological analysis revealed that Aeromonas hydrophila was associated (validated by 16S rRNA PCR and sequencing method) with the infection and mortality of P. hypophthalmus, confirmed by hemolysin and survival assay. This created a scenario of co-infections, where both infectious agents are active together, causing ichthyophthiriasis and motile Aeromonas septicemia (MAS) in P. hypophthalmus. Interestingly, turmeric oil supplementation induced protective immunity in P. hypophthalmus against the co-infection condition. The study showed that P. hypophthalmus fingerlings supplemented with turmeric oil, at an optimum concentration (10 ppm), exhibited significantly increased survival against co-infection. The optimum concentration induced anti-stress and antioxidative response in fingerlings, marked by a significant decrease in cortisol and elevated levels of superoxide dismutase (SOD) and catalase (CAT) in treated animals as compared with the controls. Furthermore, the study indicated that supplementation of turmeric oil increases both non-specific and specific immune response, and significantly higher values of immune genes (interleukin-1β, transferrin, and C3), HSP70, HSP90, and IgM were observed in P. hypophthalmus treatment groups. Our findings suggest that C. longa (turmeric) oil modulates stress, antioxidant, and immunological responses, probably contributing to enhanced protection in P. hypophthalmus. Hence, the application of turmeric oil treatment in aquaculture might become a management strategy to control co-infections in fishes. However, this hypothesis needs further validation.

Aphanomyces invadans is an aquatic oomycete pathogen and the causative agent of epizootic ulcerative syndrome (EUS) in fresh and brackish water fish, which is responsible for severe mortalities and economic losses in aquaculture. Therefore, there is an urgent need to develop anti-infective strategies to control EUS. An Oomycetes, a fungus-like eukaryotic microorganism, and a susceptible species, i.e., Heteropneustes fossilis, are used to establish whether an Eclipta alba leaf extract is effective against the EUS-causing A. invadans. We found that treatment with methanolic leaf extract, at concentrations between 50–100 ppm (T4–T6), protects the H. fossilis fingerlings against A. invadans infection. These optimum concentrations induced anti-stress and antioxidative response in fish, marked by a significant decrease in cortisol and elevated levels of superoxide dismutase (SOD) and catalase (CAT) levels in treated animals, as compared with the controls. We further demonstrated that the A. invadans-protective effect of methanolic leaf extract was caused by its immunomodulatory effect and is linked to the enhanced survival of fingerlings. The analysis of non-specific and specific immune factors confirms that methanolic leaf extract-induced HSP70, HSP90 and IgM levels mediate the survival of H. fossilis fingerlings against A. invadans infection. Taken together, our study provides evidence that the generation of anti-stress and antioxidative responses, as well as humoral immunity, may play a role in protecting H. fossilis fingerlings against A. invadans infection. It is probable that E. alba methanolic leaf extract treatment might become part of a holistic strategy to control EUS in fish species.

Alkoxy-substituted benzaldoxime ethers, namely (i) 3-methoxy-4-ethoxybenzaldoxime N-O-alkyl ethers, (ii) 3,4-dimethoxybenzaldoxime N-O-alkyl ethers, and (iii) 3,4-methylenedioxybenzaldoxime N-O-alkyl ethers, have been synthesized and evaluated for their insect growth regulatory activity against fifth-instar nymphs of the desert locust Schistocerca gregaria F. When injected into insect hemolymph at the lowest dose level of 3 µg/nymph, 3,4-methylenedioxybenzaldoxime N-O-methyl ether and 3,4-methylenedioxybenzaldoxime N-O-isopropyl ether showed 40 and 50% growth deformities respectively. On topical application (at 20 µg/nymph) 3-methoxy-4-ethoxybenzaldoxime N-O-methyl ether inflicted 100% abnormalties in insect growth. Structure-activity relationship studies revealed that maximum activity was associated with compounds having three carbons in the oxime ether moiety.

Curcuminoids, the major colouring constituents of Curcuma longa (turmeric) rhizome powder, comprise mainly three closely related curcumins (I, II and III). A simple method has been devised for their ef®cient extraction and separation. Their structures have been con®rmed by 1 H NMR spectroscopy and unique mass fragmentation pattern. Curcumin-I, the major constituent has been converted to ®ve alkyl ether derivatives, which have been tested along with the parent compounds and other extractives for insect growth inhibitory activity against Schistocerca gregaria and Dysdercus koenigii nymphs. At 20 mg per nymph, benzene extract and dibutyl curcumin-I were the most active (60% inhibition) against S gregaria, whereas at 50 mg per nymph these substances exhibited moderate growth-inhibitory activity (45%) against D koenigii nymphs. At these concentrations, turmeric oil caused 50±60% nymphal mortality in both test insects. The insect control activity of most of the turmeric products was comparable to or better than that of a commercial neem formulation.

The one‐stripe spiny eel (Macrognathus aral, Bloch & Schneider, 1801) under the family Mastacembelidae, a native ornamental fish, were collected from the freshwater ponds and canals of West Bengal, with a purpose to raise the colony as ornamental fish broodstock. During acclimatization, fish were found to be lethargic and reluctant to consume feed followed by the appearance of deep wounds and red patches on the ventral part of the body surface. There was a mortality of 40.49% of affected fish within the next 48 hours. Post‐mortem examinations revealed the presence of metacercariae inside the body cavity and on internal organs, spreading up to the buccal cavity. The abundance of the parasite was 15 to 36 numbers per fish. The length of the metacercaria varied from 5.2 mm to 9.8 mm. The metacercariae were identified as Clinostomum complanatum by light microscopy. Further, molecular confirmation of identification was done using PCR amplification and sequencing of the 18S RNA gene. This is the first report of metacercarial infestation of Clinostomum complanatum with molecular confirmation in indigenous M. aral with high ornamental value.

In vitro study was conducted to test mycellial growth inhibition effect of plant extracts on Macrophomina phaseolina causing stem rot of jute as well as for feeding inhibition and mortality on 3rd instar larvae of Bihar hairy caterpillar, Spilosoma obliqua Walker. The result revealed, at 2000 ppm, acetone extracts of sunnhemp and Azeratum conyzoides exhibited maximum of 34.44% and 41.85% mycellial growth inhibition respectively. Whereas, 83.32% and 66.67% spore germination inhibition of the fungus was observed respectively. At 5000 ppm, methanolic extracts of Crotolaria quinquefolia, garlic, curry leaf and turmeric oil recorded 35.55%, 44.44%, 50.00% and 70.00% mycellial growth inhibition of the fungus. Methanolic extracts of C. juncea, C. quinquefolia, curry leaf and garlic recorded above 80.00% feeding inhibition on S. obliqua and larval mortality of 20.00%-44.44%. It is clear that neem, sunnhemp, garlic and turmeric extract possess antifungal, insecticidal, antifeedant properties and may be integrated for management of stem rot as well as S. obliqua in jute crop.
